    Notizen: A self-similar model for the ablation driven by thermal radiation is developed for the case in which the ablated flow cannot freely expand and it is optically thick to the impinging radiation. The results show that the motion is governed by a dimensionless parameter, which determines the importance of the radiation heat conduction relative to the convective energy flux. The model can be applied to the situation arising in ion beam fusion targets, where a cold pusher is ablated by blackbody radiation emitted by the beam absorber layer that surrounds it and tamps the ablated flow. The scaling laws for the magnitudes of the flow are found and the main differences with free flow ablation are discussed.

    Notizen: The structures of the choline-dependent pneumococcal murein hydrolases, LYTA amtdase and CPL1 Iysozyme, and the choline-independent CPL7 Iysozyme were analysed by controlled proteolytic digestions. The trypsin cleavage of the CPL1 and CPL7 lysozymes produced two resistant polypeptides, F1 and F7 respectively, corresponding to the N-terminal domain of the enzymes, whereas the amidase LYTA was completely hydrolysed by the protease. Interestingly, the F1 and F7 fragments showed a low, but significant, choline-independent lysozyme activity. Choline reduced the rate of proteolytic hydrolysis of choline-dependent enzymes, suggesting that the C-terminal choline-binding domain adopts a more resistant conformation in the presence of the ligand. On the other hand, the regions encoding the N-terminal domains of the three enzymes have been cloned and expressed in Escherichia coli, showing that these domains adopt an active conformation even in the absence of their C-terminal domains. The lower activity shown by the catalytic domains when compared with that of the complete enzymes suggests that the acquisition of a substrate-binding domain represents a noticeable evolutionary advantage for enzymes that interact with polymeric substrates, allowing them to achieve a higher catalytic efficiency. These results strongly reinforce the hypothesis that the pneumococcal murein hydrolases have been originated by fusion of two structural and functional independent domains, and provide new experimental support to the theory of modular evolution of proteins.

    Notizen: Abstract The 29Si and 27Al nuclear magnetic resonance (NMR) analysis of synthetic trioctahedral phyllosilicates 2:1, with tetrahedral ratios Al T/(Si + Al T) ranging from 0 to 0.5, has shown that the ditrigonal distortion of tetrahedral rings (angle α) is the main factor controlling chemical shift values of tetrahedral components in both signals. The increase of ditrigonal rotation angle shifts these components towards more positive values. For each sample, the composition of tetrahedral and octahedral sheets determine the value of α, and from this parameter, the mean tetrahedral Tot angle and the chemical shift values of components are deduced. For a given environment, variations on ditrigonal angle are responsibles for the observed evolution of chemical shift values with bulk composition. The comparative analysis of micas and saponite samples has demonstrated that the location of compensating charge (interlayer and octahedral sheet) does not affect chemical shift values.

    Notizen: Abstract The thermal behaviour of bovine-brain myelin membrane has been studied by high-sensitivity differential scanning calorimetry, Fourier-transform infrared spectroscopy and thermal gel analysis. Spectroscopic results indicate that protein transitions take place between 60°C and 90°C, while thermal gel analysis has provided the thermal denaturation profiles of myelin proteolipid, DM-20 protein and the Wolfgram Fraction. An irreversible calorimetric transition centred at 80.3 ± 0.2°C with a specific enthalpy of 4.7 ± 0.6 J/g of total protein has been assigned to the thermal denaturation of myelin proteolipid and DM-20 protein. The effects of the myelin storage conditions, scan rate, ionic strength and pH on this calorimetric transition have also been investigated. The thermal transition of the proteolipid practically disappears after treatment of the myelin with different amounts of chloroform-methanol 2:1 (v/v), a treatment which is generally used in proteolipid purification. On the other hand, the addition of several detergents to myelin only causes minor modifications to this transition, which then occurs at about 70°C, with a specific enthalpy of between 2.5 and 3.6 J/g of total protein. These results appear to show that detergents preserve the native conformation of the proteolipid far more than do organic solvents. Hence the use of detergents would seem to be the appropriate method for proteolipid purification.

    Notizen: Abstract Myelin proteolipid has been isolated from bovine brain and purified using organic solvents according to conventional procedures. The protein content of the purified sample, or crude proteolipid, contains a minimum of 75% w/w of proteolipid, with DM-20, a proteolipid molecule with an internal deletion of 35 out of 276 amino acid residues, as the only other component. Biochemical analysis has shown the differences in lipid composition between brain white matter, myelin and crude proteolipid preparations. The latter contained practically no cholesterol, while the other two samples had about 22−23% w/w. High-sensitivity differential scanning calorimetry experiments with both crude proteolipid and its extracted pool of lipids have shown similar reversible thermal transitions at 52°C and 48°C. The effect of increasing amounts of cholesterol on the two calorimetric transitions led in both cases to a continuous decrease in the melting temperature and in the transition enthalpy. Parallel Fourier-transform infrared spectroscopy studies of crude proteolipid have detected a reversible, co-operative lipid transition centred at 49 °C, with no detectable change in the amide region between 20°C and 60°C. Once more an increase in cholesterol content led to a decrease in the sharpness of this transition. It is concluded that the thermal transition detected in crude proteolipid, which has in the past been attributed to proteolipid thermal denaturation (Mateo et al. 1986), actually corresponds to a thermotropic phase transition of the lipids included in the crude proteolipid sample.

    Notizen: Abstract Grafting reactions of phenyl groups on silica substrates using as reagents various phenylsilanes with variable distances between the silicon atoms and the aromatic ring by 0, 1, and 2 methylene groups [≡Si−(CH2) n −C6H5] were studied. Two different silicates have been selected as source of silica: sepiolite and tetraethyl-orthosilicate (TEOS). Sulfonic- and nitro-derivatives prepared from these phenyl compounds by electrophilic substitution reactions, have been obtained. The mechanism of these processes has been studied in relation to the number of methylene separating groups belonging to the starting reagents. The characterization of such systems has been achieved by XRD,29Si and13C NMR/MAS, IR, and laser microprobe mass spectrometry.

    Notizen: A method is proposed which evaluates the averaged composition of a sample in terms of the centroid of the core level XP spectrum. The method is based on the basic principle of electron spectroscopy for chemical analysis, i.e. the additive property of the chemical shifts and their proportionality to the ligand co-ordination. It allows one to calculate the averaged composition of compounds if the line shapes of spectra corresponding to stoichiometric samples (including the pure metal) are known and a few simple conditions are assumed. The main advantage of this method is that the composition can be obtained even if the individual contributions of the different oxidation states cannot be resolved or deconvoluted, so that no previous knowledge of the oxidation states or co-ordination structure is necessary. This method of calculating the composition is illustrated by its application to ZrOx, HfOx, NbNx, TaNxOy and SiNxOy obtained by different methods. The results are then compared with those obtained from the usual XPS quantitative analysis, so that further insight to the sample near-surface structure and composition can be gained by this combined study.

    Notizen: The reduction effects and compositional changes induced by 3.5 keV Ar+ bombardment of several titanates (i.e. SrTiO3, Al2TiO5 and NiTiO3) have been quantitatively investigated by XPS.In all the samples studied here the original Ti4+ species were reduced to lower oxidation states (i.e. Ti3+ and Ti2+), although to a lesser extent than in pure TiO2. On the contrary, whereas Sr2+ and Al3+ seem to remain unaffected by Ar+ bombardment, in agreement with the behaviour of the respective oxides (i.e. SrO and Al2O3), Ni2+ appears more easily reducible to Ni0 in NiTiO3 than in NiO. In addition, other specific differences were observed between the titanates, which reveal the existence of interesting chemical effects related to the presence of the different counter-ions in the titanates.In the case of Al2TiO5, its Ar+-induced decomposition to form TiO2 + Al2O3 could be followed by XPS.
