    Notes: : The environment surrounding urban streams imposes constraints upon stream enhancement projects. Constraints include bridges, culverts, highways, sewer and water lines, lack of easements, and other floodplain structures. The consequences of failure of these infrastructure constraints can be significant and should be considered in the design process. Fault tree analysis provides a systematic technique for analyzing the interactions of events that could lead to infrastructure failure. A case study of a stream in Pittsburgh, Pennsylvania, shows that fault tree analysis can effectively model the interactions between the stream system and the infrastructure constraints and predict the most likely modes of failure. In addition, the relative success of alternative designs and failure mitigation techniques can be assessed using this analysis tool, lending insight into the urban stream enhancement design process. The method could also provide justification in the design permitting process and input for risk assessment.

    Notes: Striga hermonthica (Del.) Benth., a parasitic weed of grasses, causes major yield reductions in the principal cereal crops of semi-arid Africa. Cultivar resistance is the most economic control measure, since adapted, resistant cultivars can be grown without additional input from the subsistence farmer. Information on the genetics of resistance to S. hermonthica is scant. This is partially attributable to the rarity of germplasms which exhibit stable resistance across geographical regions. The objective of this study was to determine if the stable resistance observed in sorghum [Sorghum bicolor (L.) Moench] cultivar SRN39 is heritable. Crosses were made between SRN39 and a susceptible parent, P954063. Parental, F1, F2 and backcross generations were grown in infested pots and development of both host and parasite was monitored. Significant variation among genotypes was observed for both host traits and effects on parasite populations. The F1 did not differ significantly in Striga resistance from the susceptible parent, suggesting recessive inheritance. However, hybrid vigor was exhibited by the F1 which yielded and developed as well as the resistant parent. Broad sense heritability ranged from 0.23 to 0.55 for host traits and from 0.10 to 0.43 for effect of genotypes on the Striga population. Joint scaling tests showed that observed variation in each host or parasite trait consisted of additive and dominance components, suggesting possible progress could be made with appropriate selection schemes.

    Notes: To estimate quantitative-genetic parameters of sorghum for resistance to the hemi-parasitic weed striga [Striga hermonthica (Del.) Benth.] and for agronomic traits, 36 diallel F2 populations and their nine parental lines were evaluated under severe striga infestation at two locations each in Mali and Kenya. Location means for grain yield ranged from 132 to 254 g/m2. F2 populations outyielded lines on average by 18%. For striga emergence traits, F2 heterosis values ranged from -36% to 232% among populations. Genetic and genotype x environment interaction variances of lines and F2s were highly significant for all traits. Broad-sense heritabilities for areas under striga severity progress curves and grain yield were 0.83 and 0.90 in lines, and 0.81 and 0.89 in F2s, respectively. General and specific combining ability, and their interaction effects with locations were significant for most traits. F2 superiority for grain yield under striga infestation demonstrates the potential merit of heterozygous cultivars in the target areas. Significant genotype x environment interaction entails multilocational testing to identify stable resistance. A combination of resistance with striga tolerance is recommended to breeders.

    Notes: Multi-year Instrumental records for input, throughflow and output waters of the Lilburn Cave system provide control on denudation rates as they respond to seasonal and spatial variability. Data suggest that maximum denudation is in the late fall and early winter. This is when non-snowmelt discharge is at Its maximum. At lower discharge rates the volume of water moving through the cave system Is the limiting control on the volume of denudation. During periods of snowmelt the limiting control is the rate at which the calcite dissolves. This is probably the result of water flowing through wider channels during these times. Based on instrumental measurements, there is considerable variation in terms of where denudation occurs inside the cave. The loci of dissolution change from year to year. This is to be expected in the dynamic environment of the cave where materials shift routinely. This variability should be studied over longer periods of time in order to more fully understand its extent. The relatively small area of carbonate exposure relative to the area of the drainage basin gives rise to relatively high denudation rates. The carbonate Is being removed at a rate of about 5000 metric tons per year, or at about 830 mm/y. This is about five times the rate reported in the humid karst regions of Malaysia. This information indicates that the relative proportion of carbonate in the drainage basin needs to be considered when trying to estimate denudation in other areas.

    Notes: Blocking glycolytic breakdown of glucose by inactivation of phosphoglucose isomerase (Pgi) in Escherichia coli led to a greatly reduced maximum specific growth rate. Examination of the operational catabolic pathways and their flux ratios using [U-13C6]glucose-labeling experiments and metabolic flux ratio analysis provide evidence for the pentose phosphate (PP) pathway as the primary route of glucose catabolism in the knock-out mutant. The resulting extensive flux through the PP pathway disturbs apparently the reducing power balance, since overexpression of the recently identified soluble transhydrogenase UdhA improves significantly the growth rate of the Pgi mutant. The presented results provide first evidence that UdhA restores the cellular redox balance by catalyzing electron transfer from NADPH to NADH.

    Notes: Polar lipid-derived fatty acids (PLFA) commonly found in sulfate-reducing bacteria were detected in high abundance in the sediment harvested from a monitoring well of a petroleum-hydrocarbon (PHC)-contaminated aquifer. Aquifer microcosms were incubated under sulfate-reducing conditions with [methyl-14C]toluene to determine the 14C-mass balances and with [methyl-13C]toluene to follow the flow of carbon from toluene into biomarker fatty acids. An aliquot was used to establish an aquifer-derived toluene-degrading sulfate-reducing consortium, which grew well in liquid medium. Whole-cell hybridization using 16S rRNA-targeted oligonucleotide probes specific for different phylogenetic levels within the sulfate-reducing bacteria was applied in order to characterize the sulfate-reducing populations in the original sediment, the aquifer microcosms, and the aquifer-derived consortium. In the aquifer microcosms, the 14C quantification revealed that 61.6% of the [methyl-14C]toluene was mineralized and 2.7% was assimilated. Following [methyl-13C]toluene depletion (〈1 μM), the highest 13C-enrichment was found in PLFA 16:1ω5c. In addition, biomarker fatty acids characteristic for the genera Desulfobacter and Desulfobacula (cy17:0 and 10Me16:0) were also 13C-enriched, contrary to those of other sulfate-reducing genera, e.g. Desulfovibrio and Synthrophobacter (i17:1ω7c), Desulfobulbus and Desulforhabdus (15:1ω6c and 17:1ω6c). Although hybridization detection rates remained low, indicating low bacterial activities, 43% (aquifer sediment) and 30% (aquifer microcosm) of the total active bacteria belonged to the Desulfobacteriaceae thus supporting the PLFA-based results. Desulfobacter-species (42%), which belong to the Desulfobacteriaceae, dominated the community of the consortium. Our study showed that carbon stable isotope analysis in combination with whole-cell hybridization could link toluene degradation in aquifer microcosms to the metabolic activity of the Desulfobacter-like populations. These populations could play an important role in the clean up of aromatic PHC-contaminated aquifers.
