    Notes: Abstract The presence of double minute chromosomes (dmin) in cancer cells is known to be correlated with gene amplifications. In human high grade astrocytomas or glioblastomas, about 50% of cytogenetically characterized cases display dmin. G5 is a cell line which has been established from a human glioblastoma containing multiple dmin. In order to identify the DNA content of these dmin, three techniques were successively used: conventional cytogenetic analysis, comparative genomic hybridization (CGH), and fluorescent in situ hybridization (FISH). The karyotype of G5 cells showed numerical chromosome changes (hypertriploidy), several marker chromosomes, and multiple dmin. CGH experiments detected two strong DNA amplification areas located in 9p21-22 and 9p24, as well as an underrepresentation of chromosomes 6, 10, 11, 13, 14, and 18q. By using FISH with a chromosome 9-specific painting probe to metaphase chromosomes of the G5 cell line, dmin were shown to contain DNA sequences originating from chromosome 9. This study demonstrates the usefulness of a combination of classical karyotyping, CGH, and FISH to identify the chromosomal origin of amplified DNA sequences in dmin.

    Notes: Abstract Genomic and cDNA clones have been isolated for a new gene (PO22) expressed in mature pollen of alfalfa. PO22 represents a single-or low-copy gene containing 4 introns, 2 transcriptional start sites, and coding for a predicted protein of 340 amino acids with no signal peptide and a coiled structure of39 amino acids near the carboxyl end. The amino acid sequence of PO22 is 38% identical to that of an early nodule-specific gene (ENOD8), also from alfalfa.

    Notes: Abstract PO149 is a low-copy-number gene expressed in the late stages of pollen development. The promoter region contains no similarities in DNA sequence to those of other pollen-specific genes, except for a tobacco sequence (AAATGA), which occurs four times in this alfalfa gene and much further upstream than in tobacco. Four distinct TATA boxes were detected in the promoter with the distal and proximal TATA boxes being separated by a spacer of 269 nucleotides. Hairpin loop structures were found in the 5′-and 3′-untranslated regions of PO149 mRNA. The coding region of PO149 is interrupted by two introns and encodes a putative prepeptide of 450 amino acids with homology to pollen pectate lyase-like proteins and pollen allergens. The coding region also contains sequences characteristic of both a signal peptide and a nuclear localization signal.

    Notes: Abstract We examined the long-term dynamics of plant biomass of Filifolium sibiricum steppe, Stipa baicalensis steppe and Leymus chinense (syn. Aneurolepidium chinense) steppe relative to interannual variation of precipitation and temperature during 1981–1990 in the Tumugi, Xingan League, eastern Inner Mongolia of China. On the average, annual precipitation was 41.1 cm. Peak live aboveground biomass (PLAB) was 152 g m-2 for F. sibiricum steppe and S. baicalensis steppe and 162 g m-2 for L. chinense steppe. Peak live belowground biomass (PLBB) varied between 968 g m-2 for F. sibiricum steppe and 1022 g m-2 for L. chinense steppe. The coefficient of variation (CV) in annual precipitation (25%) was lower than the CV in PLAB (35% to 37%) but larger than the CV in PLBB (10%) of the three meadow steppe sites. Rain use efficiency was 3.6 gDM m-2 cm-1 yr-1 for F. sibiricum steppe and S. baicalensis steppe, and 3.9 gDM m-2 cm-1 yr-1 for L. chinense steppe, respectively. Using the CENTURY ecosystem model, simulation results agreed reasonably well with the observed soil organic matter, seasonal dynamics and interannual variation of plant biomass of these three steppe sites during 1981–1990. The CENTURY model is slightly more successful than the empirical regression models that use annual precipitation to estimate PLAB of these meadow steppe over time. Both seasonal distribution and interannal variation in precipitation and temperature are the important controls of temporal dynamics of plant biomass, rain use efficiency, carbon flux and storage of these meadow steppe ecosystems over time.

    Notes: Abstract This paper reports on preliminary experiments on the controlled culturing of the freshwater ciliated protozoan Spirostomum ambiguum, as a first step in the development of a new low cost microbiotest, to complement the limited battery of standardized cost-effective toxicity tests presently available, with a representative of unicellular biota. Three strains of S. ambiguum originating from France and Poland respectively, have been grown in natural as well as in reconstituted waters, on several types of inert diets and in different temperature and light conditions. The best results in terms of survival and reproduction of the ciliates, leading to a standard recipe for stock culturing, were obtained at 20 °C, in reconstituted Volvic water and on a diet of oat flakes supplemented with dried leave powder as vegetal adjuvant, with weekly renewal of the medium and food. An average generation time of 4 days was obtained in the latter set of stock culturing conditions.

    Notes: Abstract The responses of femaleAphidius ervi to odors from a host food plant (Vicia faba), host aphids (Acyrthosiphon pisum), nonhost aphids (Aphis fabae), and aphid-plant complexes were investigated in a specially designed wind tunnel and a Y-tube olfactometer. In single-target (no-choice) and two-target (dual-choice) experiments, plant volatiles played a crucial role in the host foraging behavior ofA. ervi. The odor from theA. pisum-plant complex elicited the strongest responses byA. ervi females, followed by the odor from plants previously damaged by the feeding ofA. pisum. There was a significantly weaker response to odor fromA. pisum in the absence of the plant and to undamaged plants. Similarly, mechanically damaged plants and plants infested with the nonhost aphidA. fabae did not elicit strong responses. A plant that had been damaged byA. pisum and subsequently washed with distilled water was as attractive as an unwashed, previously infested plant.Aphidius ervi probably overcomes the reliability-detectability problem by selectively responding to herbivore-induced, volatile, semiochemical cues emitted by the first trophic level and by distinguishing between the volatiles induced by host and nonhost aphids.

    Notes: Abstract The traditional brainstem auditory evoked response (BAER) measurement technique (ensemble averaging) is time-consuming and is not acceptable for some time-critical clinical applications. In the paper the application of a pseudorandom binary sequence, the maximum length sequence, to human BAER measurements is examined. This technique permits a faster click rate to stimulate the test subject, and obtaines a higher signal-to-noise ratio (SNR) response through deconvolution. When compared with conventional averaging, the method can result in an improved SNR or in faster measurement of BAER. The theory of the technique and the experimental setup are presented, and theoretical analysis on the SNR improvement by this technique against averaging is also given. Actual measurements of BAER on both humans and cats indicate that this technique is effective, especially when the measurement time is not too long, or the number of trials is not too large.
