    Notes: The role of hydrodynamic flexibility stemming from a lack of configurational rigidity in the sedimentary and dispersive transport of macromolecules in dilute polymer solutions is analyzed within the framework of generalized Taylor dispersion theory. A macromolecular chain is modeled as a thermally fluctuating porous Brownian spongelike sphere which—in contrast with the classical investigations of Debye–Bueche and Brinkman for the rigid porous sphere case—is allowed to undergo thermal fluctuations in size (assumed governed by a Hookean elastic potential). Our results show an increase of up to about 20% in the average translational mobility of the flexible (size-fluctuating) sphere above that of the comparable rigid Debye–Bueche/Brinkman sphere (of equilibrium preaveraged radius). Coupling between mobility variations (arising from instantaneous fluctuations in sphere radius) and diffusive sampling of such sphere radii in size space, respectively, gives rise to a "Taylor'' dispersion mechanism, which enhances the diffusivity of the macromolecule above and beyond its purely molecular value. Indeed, the dispersivity (dyadic) of the sedimenting sphere is shown to be anisotropic, possessing a value different from its (mean) molecular diffusivity in the direction of net sedimentation. Both sedimentation and dispersion effects associated with size fluctuations are shown to be maximized at finite, intermediate values of the nondimensional sponge stiffness parameter S, rather than being monotonically decreasing functions of S. The relative importance of both effects increase with decreasing permeability of the sphere. With an increasing number (N(very-much-greater-than)1) of monomer units composing the chain, the isotropic (molecular) portion of the dispersivity dyadic decreases like N−1/2, whereas the anisotropic Taylor-dispersion portion, which is exclusively associated with the size fluctuations, increases like N3. The latter contribution, which is negligible for ordinary gravitational settling (owing to the smallness of the relevant Langevin parameter), may, however, become significant for ultracentrifugation of polymer solutions composed of long (N〉105) macromolecular chains.

    Notes: This past-presidential address considers the relationship between small-scale assembly and large-scale patterning in ciliates. Certain model examples of assembly-based patterning are reviewed, followed by the introduction of Williams and Honts' concept of “meta-assembly” as applied to the development of the oral apparatus. The major part of this review then explores two topics: the nature and origin of large-scale circumferential order, and the manner by which the large-scale order of cell directions influences the organization of membranelles of the oral apparatus. In this review, I summarize an existing formal description of large-scale positional order, allude to a more precise abstract theoretical model, and end with a brief discussion of the problem of searching for molecular mechanisms.

    Notes: Epitaxial GaAs grown by molecular beam epitaxy (MBE) at low substrate temperatures is observed to have a significantly shorter carrier lifetime than GaAs grown at normal substrate temperatures. Using femtosecond time-resolved-reflectance techniques, a sub-picosecond (〈0.4 ps) carrier lifetime has been measured for GaAs grown by MBE at ∼200°C and annealed at 600 °C. With the same material as a photoconductive switch we have measured electrical pulses with a full-width at half-maximum of 0.6 ps using the technique of electro-optic sampling. Good responsivity for a photoconductive switch is observed, corresponding to a mobility of the photoexcited carriers of ∼120–150 cm2/V s. GaAs grown by MBE at 200 °C and annealed at 600 °C is also semi-insulating, which results in a low dark current in the switch application. The combination of fast recombination lifetime, high carrier mobility, and high resistivity makes this material ideal for a number of subpicosecond photoconductive applications.

Anhand der Ergebnisse wurden Phasendiagramme für diese Lösungen erstellt. Die gemessenen Schmelzpunkte dieser Flüssigkeiten wurden mit den theoretischen Berech nungen der Schmelzpunkterniedrigung verglichen. Die Schmelztemperaturen der HAN-Lösungen in einem bestimmten Konzentrationsbereich können viel besser unter Zuhilfenahme der Zwei-Elektrolyten-Annahme abgeschätzt werden. Das Phasendiagramm der TEAN-Lösungen belegt eine Instabilität in Übereinstimmung mit einer Phasenseparierung in dieser Flüssigkeit.
    Notes: Abstract We present Differential Scanning Calorimetry (DSC) results on Hydroxyl Ammonium Nitrate (HAN) solutions and Triethanol Ammonium Nitrate (TEAN) solutions with varying concentrations. These results are used to generate phase diagrams of these solutions. The results of the melting points of these liquids are compared with the theoretical calculations of the depression of melting points. The melting temperatures of the HAN solutions at some specified concentration range are predicted rather well using the two electrolyte assumption. The phase diagram of the TEAN solutions explains an instability with respect to phase separation of this liquid.

    Notes: Summary Yeast transformants containing integrated copies of a galactose-regulated, ricin toxin A chain (RTA) expression plasmid were constructed and used in an attempt to isolate RTA-resistant yeast mutants. Analysis of RNA from mutant strains demonstrated that approximately half contained ribosomes that had been partially modified by RTA, although all the strains analysed transcribed full-length RTA RNA. The mutant strains could have mutations in yeast genes giving rise to RTA-resistant ribosomes or they could contain alterations within the RTA-encoding DNA causing production of mutant toxin. Ribosomes isolated from mutant strains were shown to be susceptible to RTA modification in vitro suggesting that the strains contain alterations in RTA. This paper describes the detailed analysis of one mutant strain which has a point mutation that changes serine 203 to asparagine in RTA protein. Although serine 203 lies outside the proposed active site of RTA its alteration leads to the production of RTA protein with a greatly reduced level of ribosome modifying activity. This decrease in activity apparently allows yeast cells to survive expression of RTA as only a proportion of the ribosomes become modified. We demonstrate that the mutant RTA preferentially modifies 26S rRNA in free 60S subunits and has lower catalytic activity compared with native RTA when produced in Escherichia coli. Such mutations provide a valuable means of identifying residues important in RTA catalysis and of further understanding the precise mechanism of action of RTA.

    Notes: Abstract The average blocking temperatures of ferritin molecules containing differing amounts of iron were determined by Mossbauer spectroscopy. The results imply that the magnetic anisotropy of the ferritin core particles is a function of particle volume. By addition of57Fe to ferritin core particles it was determined that, at a given temperature within the superpara-magnetic temperature region, the “last-in” ferric ions have average relaxation times that are shorter than those of the bulk ferric ions.
