    Notes: A technique was developed to surface immobilize plant cells and was scaled up in laboratory size bioreactors. This technique was shown not to hinder the biosynthetic potential of Catharanthus roseus immobilized cells and to induce a partial release (300 μg/L) of serpentine into the culture medium contrary to suspension cultured cells. The release pattern seemed to follow the biosynthesis trends of the product. This release mechanism could be stimulated by a factor of 10 within 2 h by increasing the pH of the culture from 5.0 to 5.5.

    Notes: The scaleup of the technique of plant cell surface immobilization was performed successfully in specifically designed laboratory size bioreactors. The immobilizing matrix was formed into a vertically wound spiral providing for a high immobilizing area-to-volume ratio (0.8-1.2 cm-1). A modified airlift and a mechanically stirred vessel delivered a best bioreactor performance characterized by low biomass frothing and highly efficient plant cell attachment and retention (≥96%). The growth of Catharanthus roseus cells investigated in these bioreactors was found not to be mass transfer limited. It required mild mixing and aeration levels (kLa ∼ 10-15 h-1). The biomass formation pattern of surface immobilized plant cells generally exhibited a linear growth phase followed by a stationary phase characterized by the presence of residual carbohydrates in the medium, contrary to suspension cultures. This behavior was found to depend on the plant cell type and/or line cultured, as well as on the inoculum age. The space restriction and unidirectional growth of the SIPC biofilm combined with the limited availability of essential intracellular nutrients rapidly accumulated from the medium by the stationary phase inoculated plant cells all likely contributed to the culture behavior.

    Notes: In the series of analytical techniques for identification of chemical substances, infrared spectrometry presents by far the highest information content. However, the information is most complicated too. It concerns a multitude of band positions, band intensities and band shapes, which, moreover, can be disturbed by matrix and other effects. The high redundancy, however, allows conclusions to be made by a qualitative, subjective procedure.IR is often used to prove the equality between a sample and a reference material, e.g. in quality control of a production process. In forensic control, the question to be answered is mostly not to prove equality, but whether or not the presence of a compound in a sample, e.g. a drug, can be proved. Moreover, testing has to be performed according to objective rules.To fulfil these requirements, a new retrieval algorithm, the ‘Adequate Peaks Search’, is presented. It concerns representing the reference spectra by sets of adequate peak positions and the sample spectrum by a set of all peak positions, whereafter the cross-sections of the sample set and the reference sets are determined. The concept ‘adequate peak’ is defined and criteria have been formulated to evaluate the results into a positive (presence of the analyte is proved) or negative (presence is not proved) conclusion.The detection limit when the Adequate Peaks Search (APS) method was applied was four to seven times lower than that attained by a number of experts.

    Notes: In view of reports on the involvement of high-molecular-weight (HMW) kininogen and high-density lipoprotein (HDL) in the Vroman effect, we studied the adsorption of fibrinogen, HMW kininogen, HDL and several other proteins from pooled human plasma and congenitally HMW kininogen-deficient plasma onto glass and low-density polyethylene, both as a function of the plasma concentration and the contact time. Mixtures of purified (lipo)proteins were also included in the study. Protein adsorption was determined by means of a two-step enzyme-immunoassay. Our results support the hypothesis that HMW kininogen is involved in the displacement of fibrinogen, which is almost instantly adsorbed from normal plasma onto glass. On hydrophobic polymers like polyethylene, the low amounts of adsorbed fibrinogen and HMW kininogen from plasma and concentrated plasma solutions may be due to a preferential adsorption of HDL.

    Notes: An apparatus was designed and assembled to study the solvent removal from solution-cast thin polymer films. The computer interfacing of a thermogravimetric analyzer, spectrophotometer, electronic flowmeters, and control valves for the apparatus enabled the preprogramming of the carrier gas velocity, carrier gas solvent content, and temperature profiles to simulate the environment experienced in large parallel flow industrial driers. The apparatus has also been designed and operated to enable the visual observation of the drying film with an optical microscope. Initial experimental studies conducted with the apparatus involved the effect of temperature on solvent removal. The results indicate that high dryer gas temperatures can apparently cause skinning of the film surface resulting in slower solvent removal rates. The skin formation can be suppressed by higher solvent concentration in the carrier gas. The visual observations revealed the formation of standing waves in the film surface during drying at high gas velocities (〉2OO cm/min). The wave formation at least partially overcomes the effect of skinning by increasing the surface area of the film, and may be the manifestation of flow instabilities involving circulation within the film.

    Notes: Polyetheretherketone-polydimethylsiloxane (PEEK-PDMS) block copolymers were synthesized from the condensation of dimethylamino terminated PDMS and hydroxy terminated PEEK oligomers in 1-chloronapthalene. Yields for block copolymers synthesised from low molecular weight PDMS oligomers were good but yields were significantly reduced when higher molecular weight PDMS oligomers were used. This was related to the limited solubility of higher molecular weight PDMS in the reaction solvent. Differential scanning calorimetry (DSC) studies indicated that phase separation of the block copolymers occurred at very short segment length (M̄n 〈 4000). A depression in the crystallinity of both the PEEK and PDMS phases in the block copolymer was observed. Thermogravimetric analysis (TGA) studies indicated that the PEEK-PDMS block copolymers displayed insufficient thermo-oxidative stability to be melt-processed successfully in PEEK based blends.

    Description / Table of Contents: Corrosion of glassfibre-reinforced plastics (GRP) in alkalic mediaWithin the corrosion of mechanical loaded GRP in acid media the dangerous stress corrosion cracking can take place. This is well known. In the absence of applications there was no need for basic investigations in alkalic media up to now. In this context the behaviour of single glass-filaments with various glass-compositions is as interesting as therewith produced unidirectional reinforced rods with thermosetting resins as matrix materials. As a standard media thinned soda lye (1 N = 4%) was used for the experiments. The composites are immersed under definite flexural load.It was found out, that corrosion in alkalic media in both cases (single filaments and GRP) is a diffusion-controlled decomposition. This plain, equal surface attack, which - in case of the composites - also the matrix underlies, can be classified as less critical as the stress crack corrosion in acids. The properties remain computable even after definite damage progresses. Suitable protective measures are discussed.
    Notes: Bei der Korrosion von mechanisch belastetem GFK in sauren Medien kann es schnell zur gefährlichen Spannungsrißkorrosion kommen. Dies ist bekannt. Aufgrund mangelnder Anwendungsfälle fehlen jedoch grundlegende Untersuchungen in basischen Medien. In diesem Zusammenhang interessiert sowohl das Verhalten von einzelnen Glasfasern verschiedener Zusammensetzung als auch von damit hergestellten unidirektional verstärkten Rundprofilen mit duroplastischer Matrix. Als Standardmedium wird bei den Versuchen verdünnte Natronlauge (1 N = 4%) eingesetzt. Die Verbunde werden einer Biegebelastung im Medium ausgesetzt.Es stellt sich heraus, daß es sich bei der Alkalikorrosion von Einzelfasern wie auch von GFK um einen diffusionskontrollierten Abbau handelt. Dieser flächige, gleichmäßige Abbau, dem im Fall der Verbunde auch die Matrix unterliegt, kann als weniger kritisch eingestuft werden als die Spannungsrißkorrosion in Säuren. Die Eigenschaften bleiben auch nach bestimmten Schädigungsfortschritten kalkulierbar. Geeignete Gegenmaßnahmen werden diskutiert.

    Notes: The crystallization behavior of two ethylene/octene copolymers, which differ in hexyl branch concentration, and their fractions were assessed. Fractionation of the crystalline linear low density polyethylenes (LLDPEs) was achieved by temperature rising elution fractionation. As the column temperature was raised, the eluted fractions exhibited a reduction in branch concentration and an increase in molecular weight. This was attributed to the difference in reactivity between ethylene and octene and the subsequent depletion of the ethylene monomer in the solution process. Spherulites formed during the crystallization of the whole polymers were well developed, banded, and displayed a wide distribution of sizes. However, spherulites of the LLDPE fractions were less well developed, more uniform in size, and tended to progressively deteriorate and become smaller as the concentration of branches increased. The ethylene and octene blocks of the copolymer crystallized independently, and it was proposed that the octene portion formed short, curved lamellae in the interfacial region of the lamellae formed from the linear ethylene portion of the molecule. Decreases in d spacing for fractions with increased short chain branching corresponded with similar drops in molecular weight.

    Notes: Distortion of ceramometal restorations during porcelain application is often attributed to mismatch of thermal expansion coefficients. Little attention has been paid to treatment the metal receives after casting. Oxide formation, lattice parameter changes, and macroscopic distortion were studied for flat specimens of two ceramometal alloys, a high Pd alloy and a high Au alloy. SnO2 and In2O3 formed on the surface of the high Au alloy after the oxidation firing, while SnO2 and CuGa2O4 formed on the high Pd alloy. Lattice parameter changes reflected the loss of base metal elements in the formation of surface oxides. The edges of specimens distorted toward the ground side. Such distortion can be explained by the induction of residual stress in the metal surface during grinding and subsequent surface stress annealing accompanied with subsurface elastic recovery during firings. Oxide formation was not related to specimen distortion. This study suggests a relationship between surface preparation and specimen distortion during porcelain application procedures.

Synthesis and Transamination of Enamino Ketones of the Tetrazole SeriesTetrazole-5-acetaldehyde enamines 5 react with aliphatic and aromatic carboxylic acid chlorides in the presence of an auxiliary base (e.g. pyridine or triethylamine) to give tetrazolylsubstituted enamino ketones 6. The latter can be modified by acid-catalyzed transamination with secondary aliphatic amines (e.g. diethylamine, pyrrolidine, piperidine, morpholine, hexamethylene imine) yielding 5-(1-acyl-2-dialkylamino-vinyl)-1-aryl-1 H-tetrazoles 7. The analogue transamination of 6 with primary alkyl, aralkyl, and aryl amines results in corresponding secondary enamino ketones 8, which exist in solution as an equilibrium mixture of the Z and E form. Acylation of secondary tetrazole-5-acetaldehyde enamines 9 does not lead to 8, but to the isomeric N-acyl derivatives 10. - Characteristic 1H-n.m.r., u.v., and i.r. spectroscopic data of the novel products are reported.
