    Notes: Abstract. The diurnal cycling of leaf water potential (Ψleaf) in field-grown sunflower (Helianthus annuus) was used to investigate the cause of water deficitinduced limitation of net photosynthesis. Daily midafternoon decreases in Ψleaf of up to 1.5 MPa and in net photosynthesis of up to 50% were typical for irrigated sunflower during seed filling. These midafternoon values were lowered an additional 0.6 to 0.8 MPa by prolonged drought treatment. There was a nearly linear relationship between the decline in net photosynthesis and reductions in leaf conductance over the course of the day. Thus, it was unexpected to find that the low, midafternoon rates of photosynthesis were associated with the highest intercellular CO2 concentrations. These and other observations suggest that the daily decline in photosynthesis represents a ‘down regulation’ of the biochemical demand for CO2 that is coordinated with the diurnally developing need to conserve water, thus establishing a balanced limitation of photosynthesis involving both stomatal and non-stomatal factors. There were no indications that either short term (i.e. diurnal declines in Ψleaf) or long term (i.e. drought treatment) water deficits caused any damage or malfunctioning of photosynthesis. Rather, both the daily declines in photosynthesis and the nearly 25% decrease in leaf area induced by prolonged drought appeared to be well-controlled adaptive responses by field-grown sunflower plants to limited water availability.

    Notes: Permanently marked tillers of five perennial grasses, native to the mixed-grass prairie of North America, were monitored to determine patterns of defoliation, architectural attributes influencing probabilities of defoliation. and post-defoliation responses. Frequency of tiller defoliation was greatest for one of the dominant midgrasses, intermediate for the remaining midgrass and two dominant shortgrasses. and least for the subdominant midgrass. Midgrass species, including the infrequently grazed subordinate species, were consistently grazed more intensively than the shortgrass species. However, the relative intensity of defoliation did not vary among species or grazing periods indicating that intensity of defoliation was primarily a function of pre-defoliation tiller height. Tiller architecture, including height, lamina number and the presence of reproductive culms, did not significantly influence frequency or intensity of defoliation within a species. The lack of evidence supporting tiller architecture as a selection criterion within a species suggests that animals were selecting on vegetation parameters at higher levels of vegetation organization than individual tillers. Grazed tillers of the three midgrasses exhibited greater relative rates of tiller elongation between grazing periods than non-grazed tillers, particularly early in the season when environmental conditions for growth were most favourable. The patterns of interspecific tiller defoliation observed in this study parallel the long-term patterns of grazing-induced species replacement observed in this grassland.

    Notes: Resistance to intercalating dyes (ethidium, acriflavine) and other organic cations, such as quaternary ammonium-type antiseptic compounds, mediated by the Staphylococcus aureus ptasmid pSK1 is specified by an energy-dependent export mechanism encoded by the qacA gene. From nucleotide sequence analysis, qacA is predicted to encode a protein of Mr 55017 containing 514 amino acids. The gene is likely to initiate with a CUG codon, and a 36bp palindrome immediately preceding qacA, along with an upstream reading frame with homology to the TetR repressors, may be components of a regulatory circuit. The putative polypeptide specified by qacA has properties typical of a cytoplasmic membrane protein, and is indicated to be a member of a transport protein family that includes proteins reponsible for export-mediated resistance to tetracycline and methylenomycin, and uptake of sugars and quinate. The analysis suggests that N- and C-terminal regions of these proteins are involved in energy coupling (proton translocation) and substrate transport, respectively. The last common ancestor of the qacA and related tet (tetracycline resistance) lineages is inferred to have been repressor controlled, as occurs for modern tet determinants from Gram-negative, but not those from Gram-positive, bacteria.

    Notes: A repetitive element (IS986), previously isolated from Mycobacterium tuberculosis and shown to detect multiple restriction fragment-length polymorphisms (RFLPs), has been sequenced. It consists of a potential insertion sequence of 1358bp, with 30-bp inverted repeat ends. IS986 has four potentially significant open reading frames (ORFs): ORFa1, ORFa2 and ORFb on one strand and ORFc on the complementary strand. The sequences of the potential translated products identify IS986 as a member of the IS3 family, with an apparent frameshift between ORFa1 and ORFa2. IS986 has potential as a highly specific probe for detection and typing of M. tuberculosis, as well as for transposon mutagenesis of mycobacteria. The sequence of IS986 is virtually identical to that of another recently described element, IS6110 (Thierry et al., 1990).

    Notes: Polygalacturonase (Peh) and other pectolytic enzymes play a crucial role in the maceration of vegetables by soft rot Erwinia spp. We have sequenced the peh gene of Erwinia carotovora subsp. carotovora, and identified its product as a precursor of molecular weight 42639, and a mature protein of molecular weight 42200. A putative KdgR-binding site was identified in the region 5’to the peh gene. The Peh protein showed significant homology with Peh from tomato. In addition, we have found homologies between pectin methylesterase and pectate lyase from Erwinia and their counterparts in tomato. These homologies are described, and their significance discussed.

    Description: This paper presents fission-track ages and confined track-length measurements from detrital apatites recovered from Ocean Drilling Program Leg 116 Site 717 and 718 cores. We interpret these data in terms of the post-depositional thermal history at these two sites and the thermotectonic history of apatite source areas. Composite apatite samples were derived by combining fine-grained sand samples from Sites 717 and 718 cores over 70- to 120-m intervals over the total depth penetrated at Sites 717 (T.D. = 820 mbsf) and 718 (T.D. = 960 mbsf). Thirty apatite grains per composite sample from ten samples (at least every other sampled interval) were dated and track-length measurements (20-50 per sample) were obtained for all samples. Mean track lengths from Site 717 samples are statistically identical, ranging from 14.4 ± 0.4 to 14.8 ± 0.3 ?m (all errors are the 95% confidence interval), and mean fission-track ages increase monotonically downhole from 4.8 ± 1.1 to 14.3 ± 2.3 Ma. For Site 718, located approximately 7 km to the south of Site 717 on an adjacent fault block, mean track lengths to 560 mbsf are equivalent to those measured from Site 717 samples. A decrease in mean track length (14.6 ± 0.3 to 13.2 ± 0.4 µm) and a corresponding decrease in mean fission-track age (21.1 ± 2.9 to 15.8 ± 2.4 Ma) with depth for samples between 560 and 960 mbsf from Site 718 indicates that post-depositional downhole shortening of fission tracks at elevated temperatures has taken place. Track-length shortening, based on mean track lengths relative to an unannealed mean track length of 16.3 µm, is approximately 10% for all Site 717 samples and for samples from the upper 560 m of Site 718. The total amount of shortening of the lowermost sample from Site 718 is approximately 20%. Based on extrapolation of published laboratory annealing experiments, maximum isothermal time-temperature condition extremes that could produce this degree of annealing at the base of Site 718 are estimated to range from 50°C for a duration of 17 m.y. (since deposition) to 55°C for a duration of 7.5 m.y. (since the onset of deformation). These estimates argue against regional thermal conduction as the only mechanism for post-depositional heating and support seafloor heat flow and shipboard geochemical evidence for local convective heat transfer in the vicinity of Site 718. In terms of source-area implications, dated samples have mean apatite fission-track ages that are only 0 to 10 m.y. older than depositional ages. These young ages imply rapid transport of sediment to the distal Bengal Fan and source areas characterized by high denundation rates (〈300 m/m.y.). These rates suggest that source areas similar to parts of the present-day Himalayas supplied sediment to the distal Bengal Fan since at least 17 Ma.
