    Notes: Large between-year variation was observed in spring air and soil temperatures at Pant-y-dwr Hill Centre (305 m altitude) from 1967 to 1984 and at Bronydd Mawr Research Centre (330 m) from 1985 to 1986 in Powys. The mean date of attaining T-sum 200°C accumulated air temperature was 13 March (range 9 February to 23 April) and that of soil temperature at 100 mm depth permanently above 5 5°C was 9 April (range 7 March to 4 May). Net herbage accumulation and response to applied N from Aberystwyth S23 perennial ryegrass (Lolium perenne L.) swards during April were also highly variable and were correlated with the date of reaching target soil temperatures of above 5 5°C for 5 consecutive days (r=−0·68, P 〈 0·001 for net herbage accumulation and r=−0·70, P 〈 0·001 for response to N).The use of early flowering varieties of perennial ryegrass gave a significant increase of herbage growth in spring compared with late varieties. From 1979 to 1980 net herbage accumulation during April from the early variety Frances was 94% more than from Perma (late) and 55% more than from Talbot (intermediate) varieties. Under conservation management more winter hardy and persistent varieties of Italian and hybrid ryegrasses (L. multiflorum L. and L. multiflorum X L. perenne) gave higher quantities of firstcut silage crops in early June than RvP Italian ryegrass.After the severe winters of 1978–79 and 1985–86, subsequent spring production from a wide range of ryegrasses was shown to be affected by sward survival, highlighting the value of winter hardy varieties when resowing in the uplands

    Notes: Summary The interpretation of data from scanning electron microscopy (SEM) combined with energy dispersive analysis of X-rays (EDAX) is dependent on the size of the excitation volume, the magnitude of which for organic materials such as wood has been little studied. From the analysis of a model system comprised of a thin layer of gold sandwiched between two layers of an epoxy plastic, it was shown that about 90% of all excitations arise from a volume about 6 μm wide and 4 μm deep. However, these dimensions vary with the orientation of the gold relative to the specimen surface, and the specimen tilt. Theoretical predictions based on these data support, but not conclusively, the previously published SEM/EDAX evidence of the penetration of urea formaldehyde resins into the wood cell wall in particleboard manufacture.

    Notes: Abstract In order to detect low copy number sequences in pea using biotin-labelled probes we optimised some aspects of the in situ hybridization technique. We found protoplast preparations to be superior to standard squashes in terms of their signal: noise ratio. Heat and alkali denaturation of chromosomal DNA were both more effective than acid denaturation. A comparison of antibody-fluorochrome and streptavidin-enzyme conjugates showed the streptavidin-alkaline phosphatase conjugate to be the most sensitive detection system. Using the optimised method, we were able to detect a single site for a 13.5 kb legumin gene clone.

We also show that the heterogeneity at the Vc-5 locus is less among r brb lines than among pea genotypes as a whole. The relevance of this RFLP is discussed in relation to the construction of the double recessive rr r brb genotypes.

    Notes: A gas chromatographic/mass spectormetric assay has been developed for the simultaneous measurement of 2-amino-3,8-dimethylimidazo[4,5-ƒ]quinoxaline (MeIQx) and 2-amino-3,4,8-trimethylimidazo[4,5-ƒ]quinoxaline (DiMeIQx) in fried beef. The method employs capillary column gas chromatography, electron capture negative ion chemical ionization mass spectrometry and a stable isotope labelled analogue of MeIQx as common internal standard. Two patties of lean minced beef which had been cooked separately were analysed and found to contain both compounds (patty 1:2.4 ng MeIQx g-1 meat, 1.2 ng DiMeIQx g-1 meat; patty 2: 1.3 ng MeIQx g-1 meat, 0.5 ng DiMeIQx g-1 meat). Neither compound was present in the meat prior to cooking.

    Notes: Summary Immature stages of the ichneumonid parasitoid, Campoletis sonorensis, develop within the haemocoel of its noctuid host, Heliothis virescens. The host cannot encapsulate the parasitoid egg owing to the suppressive effect of the polydnavirus-laden calyx fluid injected by the female parasitoid during oviposition. We have examined the effects of injection of calyx fluid on the following haemocytic manifestations of the immune system of 5th-instar larvae of H. virescens: encapsulation, nodulation, phagocytosis, erythrocyte rosetting and coagulation. Of these phenomena, only those requiring the formation of a multicellular sheath of plasmatocytes were affected. In general, encapsulation was fully suppressed; all of the C. sonorensis eggs and most of the glass rods implanted as targets were devoid of attached haemocytes 3 days after implantation although a few of the latter were coated by a sparsely distributed layer of granulocytes. Plasmatocytes also appeared to be present in thicker depositions of haemocytes. In nodulation, only the second, encapsulation-like phase was inhibited. The resistant first stage, involving the entrapment of particles by haemocytes, only resulted in the formation of amorphous, disorganized nodules. Granulocyte-dependent aspects of the immune system (phagocytosis, rosetting and possibly coagulation and the first stage of encapsulation and nodulation) occurred normally. The data suggest that in 5th-instar hosts injection of calyx fluid acts specifically on plasmatocyte function.

    Notes: Campoletis sonorensis calyx fluid arrests the development of last-instar Heliothis virescens larvae and is associated with the gross degeneration of the host's prothoracic glands. Through manipulations of ovary supernatant, Campoletis sonorensis polydnavirus (CsV) was found to be the only component of calyx fluid responsible for causing host developmental arrest. Venom from C. sonorensis had no effect on host development. Suspensions of CsV were quantified, and various doses were injected into last-instar hosts. The percentage of larvae developmentally arrested was dose dependent. In addition, larvae not arrested by injection with CsV suspensions were developmentally delayed in a dose-dependent manner. Hosts were delayed in the stage in which they were injected and, after recovery, developed at normal rates. Measurements by radioimmunoassay indicated that developmental delay was due to a suppression of ecdysteroid titers. After a dose-dependent period of suppression, hemolymph ecdysteroid titers recovered and reached titers comparable to those observed in saline-injected controls. Examination of prothoracic glands from developmentally delayed larvae revealed that partial degeneration occurred. Comparisons of the number and mean size of surviving gland cells and the length of developmental delay suggested that surviving gland cells may compensate for degenerated cells by increasing their ecdysone production.

    Notes: Maltitol was assigned unequivocally without any need for reference to model compounds by a combination of 2D, SIMPLE (secondary isotope multiplets of partially labelled entities) and DIS (differential isotope shift) NMR techniques. The combination of these three techniques is very powerful for the assignment of molecules with many exchangeable hydrogen atoms, such as carbohydrates. In the case of maltitol a complete 13C NMR assignment was achieved in three solvents [H2O, D2O and (CD3)2SO] using three experiments. A 1H-13C 2D correlation experiment allowed the 1H NMR spectrum to be assigned unequivocally for the first time.

    Notes: Chemical shifts of secondary references for D2O and (CD3)2SO solutions were measured as a function of temperature. The 1H and 13C chemical shifts of dioxane, sodium 4,4-dimethyl-4-silapentanesulphonate, HOD, tetramethylammonium chloride and sodium 3-(trimethylsilyl)propionate-d4 in D2O solution were measured relative to external TMS and the chemical shifts of (CD3)2SO and H2O were measured in (CD3)2SO solution relative to internal TMS. For accurate comparison of chemical shifts it is necessary to take into account the sample temperature and, therefore, the temperature dependences of 1H and 13C chemical shifts of a number of secondary references have been determined accurately and fitted to a parabola or straight line, as appropriate.
