    Notes: Monoclonal antibodies are being manufactured for clinical trials in suspension culture at the 1300-L scale. Suspension culture offers some advantages relative to high-density mammalian cell culture methods; in particular, the ability to closely monitor the behavior of cells in a homogeneous environment. Computer control and on-line mass spectrography of exit gases provide instantaneous information about the culture metabolic activity. Air sparging and agitation by marine impeller provide aeration sufficient to maintain a constant dissolved oxygen tension at cell concentrations up to 5.0 × 106 cells/mL without causing apparent cell damage.

    Notes: A combination of ionic strength reduction and diafiltration of Trichoderma reesei cellulate complex through a hollow fiber apparatus of 5000 molecular weight (MW) cutoff and subsequent passage of filtrate over a Spherogel-TSK 3000-SW column provided extracts that had the ability to generate microfibrils in filter paper and to disrupt filter paper and corn leaf tissue. Milligram quantities of material obtained from these extracts released small amounts of soluble carbohydrate from filter paper, required ferric iron for increased activity, and contained amino acids. Short fiber formation and disruption of filter paper during interaction with these extracts was enhanced by prior acid treatment and eliminated by prior base treatment. The amount of soluble carbohydrate hydrolyzed in 24 h from filter paper by whole cellulase complex was not changed by first disrupting the substrate with the extracts.

    Notes: The Raman and infrared spectra of glutaric acid, (CH2)3[COOH(D)]2, and its potassium salts, CH2)3(COO2KHD and CH23COO2K2, in aqueous solution have been investigated in the range 3500-200 cm-1. The analysis of the polarization ratio of the Raman lines indicated that glutaric acid belongs to point group C1 whereas its ions belong to point group Cs. An assignment of the fundamental vibrations is proposed.

    Notes: A fast atom bombardment/collisional activation/linked-scan at constant B/E (tandern mass spectrometric) method is described which can distinguish between α- and β-aspartyl and α- and μ-glutamyl underivatized peptides. The method is based upon differences in loss of CO from aspartyl or glutamyl B-fragment ions (IB) in these isomers which are rationalized from the stability of the resultant A-fragment ions (IA). It was observed that the ratio of IB:IA which was used in this determination was dependent upon the collision cell pressure. The higher the collision cell pressure, the larger the difference between the IB: IA ratios for these linkages.

    Notes: Analysis of doxylamine N-oxide and pyrilamine N-oxide as synthetic standards and biologically derived metabolites by thermospray mass spectrometry (TSP/MS) provided [M + H]+ ions for each metabolite. TSP/tandem mass spectrometry (TSP/MS/MS) of the [M + H]+ ions provided fragment ions characteristic of these metabolites. In addition, TSP mass spectrometry and TSP/MS/MS analysis of ring-hydroxylated N-desmethyldoxylamine, N-desmethylpyrilamine and O-dealkylated pyrilamine is also reported. A fragmentation pathway for analysis by MS/MS of pyrilamine and its metabolites is also proposed. The results demonstrate the utility of TSP/MS for biologically derived metabolites of pyrilamine and doxylamine.

    Notes: Techniques for the immobilization of bovine carbonic anhydrase (BCA) on porous silica beads and graphite are presented. Surface coverage on porous silica beads was found to be 1.5 × 10-5 mmol BCA/m2, and on graphite it was 1.7 × 10-3 mmol BCA/m2 nominal surface area. Greater than 97% (silica support) and 85% (graphite support) enzyme activity was maintained upon storage of the immobilized enzyme for 50 days in pH 8 buffer at 4°C. After 500 days storage, the porous silica bead immobilized enzyme exhibited over 70% activity. Operational stability of the enzyme on silica at 23°C and pH 8 was found to be 50% after 30 days. Catalytic activity expressed as an apparent second-order rate constant K′Enz for the hydrolysis of p-nitrophenyl acetate (p-NPA) catalyzed by BCA immobilized on silica beads and graphite at pH 8 and 25°C is 2.6 × 102 and 5.6 × 102 M-1s-1 respectively. The corresponding KENZ value for the free enzyme is 9.1 × 102 M-1s-1. Activity of the immobilized enzyme was found to vary with pH in such a manner that the active site pK, on the porous silica bead support is 6.75, and on graphite it is 7.41. Possible reasons for a microenvironmental influence on carbonic anhydrase pKa, are discussed. Comparison with literature data shows that the enzyme surface coverage on silica beads reported here is superior to previously reported data on silica beads and polyacrylamide gels and is comparable to an organic matrix support. Shifts in BCA-active site pKa values with support material, a lack of pH dependent activity studies in the literature, and differing criteria for reporting enzyme activity complicate literature comparisons of activity; however, immobilized BCA reported here generally exhibits comparable or greater activity than previous reports for immobilized BCA.

    Notes: Static coating of phenyl and biphenyl polysiloxane stationary phases on 50 μm i.d. open tubular capillaries was studied. The influence of coating solvents and coating temperatures on the viscosities and surface tensions of the polymer stationary phases and their coating solutions was determined. A measure of the Rayleigh instability paralled the observed coating efficiencies.

    Notes: This study describes the use of high performance liquid chromatography (HPLC) and capillary gas chromatography/mass spectrometry (GC/MS) in the characterization of polar glucuronide conjugates of doxylamine and their subsequent aglycones following deconjugation. Rat urinary extracts which contained doxylamine and both nonconjugated and conjugated doxylamine metabolites, were examined by HPLC before and after incubation with rat intestinal microflora. The subsequent deconjugated urinary metabolites and the nonconjugated products remaining in the urinary extracts were then isolated, acetylated, and assayed by GC/MS. Incubation with the intestinal microflora indicated that anaerobic bacteria were capable of effecting hydrolytic cleavage of these polar O-glucuronide metabolites of doxylamine and its demethylated products to their subsequent aglycones. GC/MS analysis was performed using a fused silica DB-5 GC column and was utilized for the identification of these deconjugated metabolites.

    Notes: A method has been developed for fabricating fused silica capillary columns which have specific surface properties but still retain the excellent strength, flexibility, and resilience of pure fused silica. By using the Modified Chemical Vapor Deposition process (MCVD), typically used for the production of optical fiber lightguides, inorganic dopants such as Al, Nd, Ge, and P can be introduced into the preform-tube by MCVD. Doped columns have a wide range of specific surface properties, and columns with undoped fused silica prepared by MCVD are more chemically inert and less acidic than columns prepared by conventional methods. This paper describes the method for fabricating capillaries and the initial studies to characterize them.

    Notes: Cyclic, high molecular weight perfluorocarbons (PFC's) are very electronegative, and can be detected by methane negative ion chemical ionization (NICI); however only molecular ions are detected. Electron ionization (EI) of PFC's, on the other hand, yields little or no molecular ion but very intense low mass fragment ions such as CF3+ (m/z 69), C3F5+ (m/z 131) and C4F7+ (m/z 181), which are not structurally characteristic. High-energy collisionally activated decomposition (CAD) of positive, molecular ions of perfluoro-1,3-dimethyladamantane yields important structural information by providing more intense high mass fragment ions. Fragments of the following formulae and abundances (in brackets): C5F11+ (100), C11F17+ (73), C7F9+ (67), C8F11+ (43), C4F7+ (26), C7F8+. (18), and CF3+ (14) are detected by this method.
