    Notes: An immobilized lipase suitable for fat interesterification has been prepared by precipitation with acetone of a commercial lipase from Rhizopus arrhizus onto diatomaceous earth. As observed previously with a less active enzyme from Aspergillus sp., the interesterification activity was enhanced by addition of purified lipase or by high loadings of commercial enzyme. The interesterification activities reached maximum values in both cases. For immobilized preparations with purified enzyme, interesterification activity was also enhanced by the presence of a precoat of glutaraldehyde cross-linked commercial lipase. A 2.9-L column of immobilized lipase was used to interesterify batches of shea oleine (67 kg) and shea oil (40 kg). Little activity was lost processing shea oleine, but slow poisoning of the bed occurred when shea oil was fed to the column.

    Notes: A procedure is described for the analysis of the estrogens equilin and estrone in human plasma following oral administration of conjugated estrogen preparations. After enzymatic hydrolysis of the sulfate conjugates, plasma proteins are precipitated with methanol and the estrogens extracted into ethyl acetate. Derivatization with the reagent flophemesylamine converts equilin and estrone into volatile pentafluorophenyldimethylsilyl ethers ideally suited to capillary gas chromatography-negative ion chemical ionization mass spectrometry. Using a 15 meter dimethyl silicone bonded phase fused silica capillary column separation of the estrone and equilin derivatives is achieved within 9 minutes. Selected ion monitoring of the intense negative molecular ions enables levels of 1 ng.ml-1 to be measured with coefficients of variation of 9.3 % and 14.2 % for estrone and equilin respectively. Plasma levels of the compounds are reported in two male volunteers up to 24 hours after dosing with 5 milligrams of Premarin™. (™ Ayerst Laboratories Inc., New York, USA.).

    Notes: Glass capillaries were leached, dehydrated, persilylated with 1, 3-bis (3-cyanopropyl) tetramethyldisiloxane, and coated with OV-240-OH. After crosslinking and binding the phase to the glass surface the columns showed high separation efficiency, high temperature stability, and inertness comparable to persilylated apolar columns. Column performance is shown to be superior to liquid phase cyanopropyl columns such as SP 2330. The excellent separation capabilities together with the selectivity of the phase makes OV-240-OH coated columns a valuable tool for the determination of toxic isomers in complex mixtures of polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs). The order of elution of individual TCDD isomers was found to be similar to that described for SP 2330 or Silar 10c. The detection of PCDDs and PCDFs in a fly ash extract further illustrates the utility of OV-240-OH coated columns. The high temperature limit of these columns opens the way for the analysis of high boiling compounds such as mixed brominated/chlorinated dibenzo-p-dioxins and dibenzofurans.

    Notes: The ions produced by 252Cf plasma desorption are detected by Fourier transform ion cyclotron resonance techniques. Results for CsI and N,N,N-trimethyl-p-aminophenolate iodide show the feasibility of the technique with high resolution (up to 11 000 at m/z 23) and good signal-to-noise ratio (up to 726).
