Notes: Summary The three-dimensional structure of the mitochondria and sarcoplasmic reticulum (SR) in the three types of twitch fibers, i.e., the red, white and intermediate skeletal muscle fibers, of the vastus lateralis muscle of the Japanese meadow frog (Rana nigromaculata nigromaculata Hallowell) was examined by high resolution scanning electron microscopy, after removal of the cytoplasmic matrices. The small red fibers have numerous mitochondrial columns of large diameter, while the large white fibers have a small number of mitochondrial columns of small diameter. In the medium-size intermediate fibers, the number and diameter of the mitochondrial columns are intermediate between those of the red and white fibers. In all three types of fibers, the terminal cisternae and transverse tubules form triads at the level of each Z-line. The thick terminal cisternae continue into much thinner flat intermediate cisternae, through a transitional part where a row of tiny indentations can be observed. Numerous slender longitudinal tubules originating from the intermediate cisternae, extend longitudinally or obliquely and form elongated oval networks of various sizes in front of the A-band, then fuse to form the H-band collar (fenestrated collar) around the myofibrils. On the surface of the H-band collar, small fenestrations as well as tiny hollows are seen. The three-dimensional structure of SR is basically the same in all three muscle fiber-types. However, the SR is sparse on the surface of mitochondria, so the mitochondria-rich red fiber has a smaller total volume of SR than the mitochondria-poor white fiber. The volume of SR of the intermediate fiber is intermediate between other the two.

Notes: A 9% cross-linked porous polystyrene resin (10-15 μm) incorporating poly(γ-methyl L-glutamate) (PMLG) was synthesized, and the grafted PMLG was then converted into poly(N5-benzyl-L-glutamine) (PBLGN). In this newly developed procedure, almost quantitative conversion of PMLG to PBLGN is achieved. The obtained adsorbent for liquid chromatography demonstrated a total optical resolution of (RS)-5-isopropylhydantion. Experimental evidence demonstrated that the degree of polymerization (DP) of the incorporated poly(α-amino acid) is a critical variable. At DP = 2, no resolution was accomplished, whereas At DP = 14 or 36, base line resolutions were achieved. Other fundamental factors, such as the concentration of the immobilized PBLGN, the column temperature and the eluent composition, which affect the resolution efficiency, were also evaluated.

Notes: Polymer adsorbents for chromatographic optical resolution were synthesized by grafting poly(N5-benzyl-L-glutamine-co-γ-benzyl L-glutamyl) onto cross-linked polystyrene beads. The effects of the crosslinking degree (1,5%, 9%, 30%) and the porosity of the polystyrene matrix on the resolution efficiency of the adsorbent were investigated. Similar resolution efficiencies were observed on 1,5% cross-linked gel (non-porous) and 9% cross-linked porous adsorbents, while a decline in the efficiency was observed on 30% cross-linked porous adsorbent. Studies of transmission electron micrographs of these adsorbent clearly showed the effect of degree of crosslinking on the distribution manner of the incorporated poly(α-amino acid). At a degree of 30%, clustered incorporation occurs mainly on the pore surface and not within the cross-linked polymer matrix. Such incorporation manners are also suggested by the swelling properties of the adsorbents. Pore volumes, pore radii, and specific surface areas of the adsorbents were also measured. It was observed that uniform incorporation of poly(α-amino acid) derives a higher resolution efficiency of the adsorbent.

Notes: Polymer membranes having cationic charge site, poly(1-alkyl-4-vinylpyridinium iodide-co-acrylonitrile) (alkyl: methyl, butyl, or octyl) were prepared in terms of coulombic interaction for separation of water-ethanol mixtures. The incorporation of cationic charge site into the membrane led to improve not only separation factors (selectivity toward water) but also flux number in the separation of aqueous ethanol solution by pervaporation technique. Target values, which were requested to have from the viewpoint of industrial utilization for separation of aqueous ethanol solution were attained as follows: separation factor toward water over 50 and flux value over 500 g m-2 h-1, through poly(1-methyl-4-vinylpyridinium iodide-co-acrylonitrile) (membrane 3) (quarternized fraction of pyridinium moiety, 89.5%; mol fraction of pyridinium moiety, 0.034) and poly(1-butyl-4-vinylpyridinium iodide-co-acrylonitrile) (membrane 5) (quarternized fraction, 100%; mol fraction of pyridinium moiety, 0.038). It was found that the introduction of cationic charge site into the membrane was one of feasible methods to obtain suitable membranes for water permselective membranes in the separation of water-ethanol mixtures.

Notes: As a novel polymer adsorbent for optical resolution, cross-linked polystyrene gel incorporating poly(α-amino acids) was synthesized. The helicity of the incorporated poly(γ-benzyl L-glutamate) (PBLG) was demonstrated by Fourier-transform infrared spectroscopy. The immobilized PBLG (I) was converted to poly(L-glutamic acid) (II) and poly(N5-benzyl-L-glutamine) (III). The ability of I-III to resolve DL-mandelic acid was evaluated by liquid chromatography using toluene/dioxane as an eluent. Of the three resins, III resolves the recemate most effectively. In order to clarify the mechanism of chiral recognition, poly(N5-benzyl-D-glutamine) and poly(N4-benzyl-L-asparagine), with opposite helicity, was incorporated. In Contrast to III, these adsorbents demonstrated affinity for the L isomer. This result strongly indicates that the helical structure of the immobilized poly(α-amino acids) is essential for chiral recognition.

Notes: Various investigations have been carried out in order to further elucidate the chiral recognition mechanism of immobilized poly(N5-benzyl-L-glutamine) (PBLGN) for optical resolution. The shape and dimension of the chiral recognition site are determined by resolution of hydantoin derivatives, with substituents of varied bulkiness at the chiral center. The site of the hydrogen bonding association of the enantiomers responsible for chiral recognition is also elucidated. Several adsorbents with electron-donating/withdrawing substituents incorporated into the PBLGN side chain phenyl are synthesized and evaluated for the resolution of (RS)-5-isopropylhydantoin in order to elucidate the association site of PBLGN. Based on the experimental evidences obtained, a most plausible mechanism of chiral recognition is proposed. Additionally, adsorbents with several other poly(α-amino acid)s are also synthesized, and the effect of poly(α-amino acid) side chain length is discussed.

Notes: Two kinds of aromatic-aliphatic polyamide oligomers were newly prepared by the reactant pairs of 3,4′-oxydianiline-adipic acid and 3,4′-oxydianiline-azelaic acid. These oligomers were then condensed separately with α, ω-polybutadienedicarboxylic acid giving two series of polybutadiene-polyamide multiblock copolymers. Properties of four series of polybutadiene-polyamide multiblock copolymers, whose polyamide blocks consisted of not only newly prepared polyamides but also previously synthesized aromatic polyamides derived from 4,4′-oxydianiline-isophthalic acid and 3,4′-oxydianiline-isophthalic acid, were investigated on the view point of structure-property relationship. A larger extent of the Tg depression of polybutadiene phase, and higher tensile strength and modulus were observed in the block copolymers having aromatic polyamides compared with those having aliphatic ones.
