    Notes: Human polymorphonuclear leukocyte elastase (PMN elastase) is inhibited by L-659, 286 (7α-methoxy-8-oxo-3-[[(1,2,5,6-tetrahydro-2-methyl-5,6-dioxo-1,2,4-triaz-in-3-yl)thio]methyl]-5-thia-1-aza-6R-bicyclo [4.2.O]oct-2-ene-2-pyrrolidine carboxamide-5,-dioxide) with a Ki of 0.4 μM. This inhibition is time-dependent, rapid, and only slowly reversible, with a t1/2 of 〉 3 days at 25°C. L-659, 286 is also highly selective for PMN elastase, as it does not inhibit thrombin, trypsin, papain, plasmin, chymotrypsin, or cathepsin G. L-659, 286 administered intratracheally inhibits lung damage caused by administration via the same route of human PMN elastase into hamsters. In marmosets, L-659, 286 is cleared from blood very rapidly after an intravenous injection but is recovered in bronchoalveolar lavage fluid for several hours after intratracheal administration.

    Description: Spectra of SN 1987A showing the Ni II 6.634-micron and Ar II 6.983-micron fine-structure lines were obtained from the Kuiper Airborne Observatory in April 1988. The signal-to-noise ratio of 100 near the peaks and resolving power of 200 are sufficient to show the average velocity of expansion from the core of about 1400 km/s and to indicate the range of velocities. An asymmetry in the profiles of both lines and a redshift of the line centroids of about 440 km/s above the 280-km/s recessional velocity of the LMC can be explained in terms of scattering of the photons by electrons in the expanding hydrogen envelope of the supernova. A mass of 0.0030 solar masses of Ni II can be deduced from the line strength of the Ni II line and a mass of 0.0009 solar masses of Ar II from the Ar II line strength.

    Description: Results are reported from a search to determine the correlation, if any, between the temporal behaviors of 0.2-7 MeV or higher electrons at GEO (6.6 earth radii) and 6-10 MeV electrons in the interplanetary region near earth at the period of the Jovian synodic year (about 13 months). The 13-month intensity variation results from the synodic interplanetary magnetic field conection of earth to Jupiter. Direct compariosn of intensity-time flux profiles for the years 1976-1984, about 7 synodic Jovian electron seasons, shows that the intensity envelope of peak electron flux at GEO does not appear to be correlated to the observed 13 month intensity envelope of relativistic electron flux in the interplanetary region near earth. A persistent 13-month variation of GEO flux is not obvious, thus indicating that the intensity of electron flux at GEO is not directly and soley related to the intensity of Jovian electron flux near earth. It is concluded that dynamic erergization and redistribution processes in earth's magnetosphere must be invoked to produce the intensity variations of relativistic electron flux at GEO and not interplanetary magnetic field connection to Jupiter.

    Description: The Einstein Observatory and the VLA were used to scan the radio halo of the galaxy M87 to detect the presence, if any, of inverse Compton X-ray (ICX) emissions. The scans revealed a coincidence of an asymmetrical X-ray structure with a luminosity near 4 x 10 to the 41st ergs/sec and dominant symmetrical emission from the hot interstellar gas. The emission can be attributed to ICX if the magnetic field of the proposed halo varies between 3-8 times below equipartition levels of 2-5 micro-gauss, which would imply that the confinement is produced by an outside medium. The necessary characteristics of the confining mechanism are discussed.

    Description: Spectrophotometry of comet Halley from 5-13 microns was obtained from the Kuiper Airborne Observatory and from the Lick Observatory Nickel Telescope, revealing a strong broad emission band at 10 microns and a weak feature at 6.8 microns. The 10-micron band is identified with silicate materials, and the primary component of the silicate emission is suggested to be due to olivine. The 6.8 micron feature may be due either to carbonates or the C-H deformation mode in organic molecules. The data indicate that small particles are abundant in the coma and that the dust contains at least two physically separate components. Significant spatial and temporal variations are also noted in the spectrum.

    Notes: Development and innervation of the lymph heart musculature of chicken, emu, rhea, and duck was studied by electron microscopy at posthatch ages from 3 days to adulthood. Development of innervation was monitored by acetylcholinesterase staining. Horseradish peroxidase was used to determine the extent of the transverse tubule network. Chickens were unusual among these birds in that lymph heart myocytes had already undergone a definitive differentiation and degeneration by 3 days. In ducks and ratite birds, lymph heart myocytes more slowly but progressively differentiate a cytomorphology that does not conform in all characteristics to cardiac or skeletal muscle and even resembles in some aspects, smooth muscle. Myofibrils become the dominant cytoplasmic structure, transverse tubules form ‘internal couplings’ with agranular reticulum cisternae, and ‘external couplings’ are formed between myocytes at myomyal junctions. The myomyal junctions also contain AChE-positive reaction product and some subplasmalemmal vesicles that lack a dense core. The lymph heart myocardium of ducks of 2 weeks demonstrated mitotic figures. In adult ducks the myosatellite cell numbers diminish and a characteristic pattern of myocyte degeneration appears. In juvenile ducks and ratites some myocytes differentiate to conductile cells, much as the conductile myocytes and myofibers of the blood heart. The lymph heart innervation is described, and the role of nerve in differentiation and maintenance of myocyte morphology in the lymph heart is discussed.

    Notes: Conjugates of the single-chain ribosome-inactivating protein gelonin with ligands that bind to cell surface molecules vary greatly in their cytotoxicity. Conjugates that are not endocytosed after binding to cells exhibit low cytotoxicity similar to that of free gelonin, while conjugates that are endocytosed demonstrate enhanced cytotoxicity relative to free gelonin. However, the number of internalized gelonin molecules needed to intoxicate cells to the same degree has been found to be similar for all conjugates and for free gelonin. The intracellular concentration of gelonin has to be between 2,000-10,000 molecules/cell to achieve a surviving fraction of 0.37. Our studies revealed the presence of three distinct categories of cell surface molecules, those that are efficient in mediating endocytosis of im-munotoxins, those that are only moderately efficient, and those that seem not to cause internalization of bound immunotoxins.

    Notes: Marrow stroma has been shown to have osteogenic potential. Here we report the characterization of a unique stromal cell line derived from mouse bone marrow (MBA-15), which expresses osteoblastic phenotype in vitro and forms bone in vivo. More than 70% of cells in culture were histochemically positive for alkaline phosphatase. The enzyme levels were enhanced threefold when cultures were treated with dexamethasone. Gel electrophoresis of [3H]-proline-labeled cultures showed that MBA-15 cells produced only type I collagen. These cells were responsive to PTH, as indicated by a 50-fold increase in intracellular cAMP. Prostaglandin E2, but not calcitonin, stimulated cAMP up to 70-fold. When cultures were grown to confluence and fed daily with ascorbic acid and β-glycerophosphate, the cells formed a Von Kossa positive, thick extracellular matrix, shown to contain hydroxyapatite crystals. MBA-15 cells produced mineralized bone when implanted in diffusion chambers. These results indicate that the MBA-15 cell line possesses osteoblastic features in vitro and osteogenic capacity in vivo.

    Notes: Entry times for spherical (no pseudopods) polymorphonuclear leukocytes (PMNs) into a 4 μm micropipet have been measured as a function of pipet suction pressure (2,500-20,000 dyn/cm2) and concentration of the drug pentoxifylline (PTX, 0.1-10.0 mM). For control cells (0 mM PTX), entry rates (reciprocal entry times) increased almost linearly with increasing suction pressure, indicating a Newtonian-like behavior. With incubation in PTX solutions, entry rate vs. suction pressure became increasingly non-linear, suggesting a shear-thinning effect for the dissipative structure. At a given suction pressure the rate of entry showed a dose-dependent increase with increasing PTX concentration, the effect being most pronounced at high suction pressures (20,000 dyn/cm2). Also, with increasing PTX concentration two other effects were observed: (i) there was a decreased incidence of cells that displayed pseudopodia, and (ii) there was an increased incidence of cells forming hernias and an increased streaming of cell cytoplasm during aspiration. The first observation points to a down-regulation of the cell's functional ability to “activate” in response to surface/chemical stimuli, and the second indicates that both the cortical and cytoskeletal networks are weakened either by disruption and/or reduction in density of the protein polymers. These observations are in line with other recently published experiments which suggest that the rheological effects of pentoxifylline on PMNs may be associated with the state of actin.

    Notes: When visual fields of the primitive orb-weaver, Waitkera waitkerensis, are reconstructed using measurements taken from intact lenses and cross and longitudinal sections of the prosoma, they show that this species has complete visual surveillance, but that none of the visual fields of its eight eyes overlap. The more advanced orb-weaver, Uloborus glomosus, also has eight eyes, but each eye has a greater visual angle, giving this species a complex pattern of overlapping visual fields. Uloborids that spin reduced webs are characterized by reduction or loss of the four anterior eyes and other carapace modifications necessary for them to effectively monitor and manipulate their reduced webs. The eyes of these uloborids have greater visual angles than those of orb-weavers, resulting primarily from perimetric expansion of their retinal hemispheres. Additionally, the axes of their visual fields are more ventrally directed due to greater dorsal than ventral retinal expansion and to ventral redirection of the entire eye. Consequently, even though the anterior lateral eyes of the triangle-weaver Hyptiotes cavatus lack retinae, the species' six functional eyes permit complete visual surveillance and exhibit visual overlap. The single-line-weaver, Miagrammopes animotus, has lost its four anterior eyes, and with them much of the anterior vision and all of the visual overlap found in the other species. However, changes similar to those of H. cavatus permit this species to retain most if its dorsal and ventral visual surveillance. Thus, ocular changes act in consort to maintain relatively complete visual surveillance in the face of eye loss and other major carapace modifications necessary for the operation of reduced webs.
