    Notes: Structure property relationship of four different diisocyanates is studied in detail. The isocyanates selected are TDI, MDI Crude, HDI, and IPDI. Physical properties such as mechanical, dynamic mechanical, electrical, and thermal are studied. Dynamic mechanical analyses are based on the compound resonance principle. Among all, IPDI-based polymer showed mediocre trend and TDI-based polymer showed best properties.

    Description / Table of Contents: Blown films of LD-PE prepared with varying processing conditions have been investigated using different X-ray diffraction methods. All films have a-texture. Differences in the degree of orientation are noticed. Arragement and size of lamellae are estimated by SAXS-investigations. A WAXS peak separation procedure using Pearson-VII-functions allows the determination of crystallite sizes. The orthorhombic modification as the dominant crystalline phase and some amount of monoclinic modification of PE have been detected from the resolved WAXS pattern.

Eine Peak-Separation der WAXS mittels Pearson-VII-Funktionen erlaubt Kristallitgrößenangaben und läßt monokline Anteile neben der hauptsächlich auftretenden orthorhombischen Modifikation erkennen.

    Description: The generally mild bleeding disorder of von Willebrand disease is associated with abnormalities of two distinct plasma proteins, the large multimeric von Willebrand factor (vWF), which mediates platelet adhesion, and von Willebrand antigen II (vW AgII), which is of unknown function. The two proteins were found to have a common biosynthetic origin in endothelial cells and megakaryocytes, which explains their simultaneous absence in the severe form of this hereditary disease. Shared amino acid sequences from a 100-kilodalton plasma glycoprotein and from vW AgII are identical to amino acid sequences predicted from a complementary DNA clone encoding the 5' end of vWF. In addition, these proteins have identical molecular weights and immunologic cross reactivities. Monoclonal antibodies prepared against both proteins recognize epitopes on the pro-vWF subunit and on a 100-kilodalton protein that are not present on the mature vWF subunit in endothelial cell lysates. In contrast, polyclonal antibodies against vWF recognize both pro-vWF and vWF subunits.     Description: A sensitive radioimmunoassay for atrial natriuretic peptide was used to examine the relation between circulating atrial natriuretic peptide and cardiac filling pressure in normal human subjects, in patients with cardiovascular disease and normal cardiac filling pressure, and in patients with cardiovascular disease and elevated cardiac filling pressure with and without congestive heart failure. The present studies establish a normal range for atrial natriuretic peptide in normal human subjects.     Description: Human probes identifying the cellular homologs of the v-ets gene, Hu-ets-1 and Hu-ets-2, and two panels of rodent-human cell hybrids were used to study specific translocations occurring in acute leukemias. The human ets-1 gene was found to translocate from chromosome 11 to 4 in the t(4;11)(q21;23), a translocation characteristic of a subtype of leukemia that represents the expansion of a myeloid/lymphoid precursor cell. Similarly, the human ets-2 gene was found to translocate from chromosome 21 to chromosome 8 in the t(8;21)(q22;q22), a nonrandom translocation commonly found in patients with acute myeloid leukemia with morphology M2 (AML-M2).     Description: The myb gene is the transforming oncogene of the avian myeloblastosis virus (AMV); its normal cellular homolog, c-myb, is conserved across a broad span of evolution. In humans, c-myb is expressed in malignant hematopoietic cell lines and in primary hematopoietic tumors. Partial complementary DNA clones were generated from blast cells of patients with acute myelogenous leukemia. The sequences of the clones were compared to the c-myb of other species, as well as the v-myb of AMV. In addition, the carboxyl terminal region of human c-myb was placed in an expression vector to obtain protein for the generation of antiserum, which was used to identify the human c-myb gene product. Like v-myb, this protein was found within the nucleus of leukemic cells where it was associated with the nuclear matrix.     Notes: This article presents a method for determining the rate constant for deactivation and the internal distribution of immobilized enzyme. This method makes use of the parallel deactivation process in a diffusion-controlled regime, in which the internal activity profile behaves like a penetration front. This front basically traces through the initial active enzymatic profile, and one can determine the internal profile and the rate constant for deactivation from the experimentally observable bulk concentration versus time. This method is applied to the experimental data of the system of hydrogen-peroxide-immobilized catalase on controlled pore glass and Si-Al particles.

    Notes: Xylose transport, xylose reductase, and xylitol dehydrogenase activities are demonstrated in Saccharomyces cerevisiae. The enzymes in the xylose catabolic pathway necessary for the conversion of xylose to xylulose are present, although S. cerevisiae cannot grow on xylose as a sole carbon source. Xylose transport is less efficient than glucose transport, and its rate is dependent upon aeration. Xylose reductase appears to be a xylose inducible enzyme and xylitol dehydrogenase activity is constitutive, although both are repressed by glucose. Both xylose reductase and xylitol dehydrogenase activities are five- to tenfold lower in S. cerevisiae as compared to Candida utilis. In vivo conversion of 14C-xylose in S. cerevisiae is demonstrated and xylitol is detected, although no significant levels of any other 14C-labeled metabolites (e. g., ethanol) are observed.

    Notes: A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system using celite as a support material. Hyphal growth through the pore matrices of the material showed strong anchorages and provided highly stable biofilm growth. With bioparticles developed in such a manner, both cell growth and penicillin production were observed to increase significantly compared to the conventional dispersed filamentous cultures. Maximum values of specific penicillin production rate were found to be constant regardless of the growth form. A three-phase fluidized-bed fermentor was designed and tested for penicillin production using the bioparticles. Two modes of operation, semicontinuous and repeated fed batch, of the fermentor were tried. It was noted that the overgrowth of free mycelia and the development of fluffy loose bioparticles caused poor mixing and made the fermentor operation quite difficult. Control of the bioparticle size and the extension of production phase were therefore considered important to maintain the reactor productivity at a desired level. From the results of repeated fed-batch operation it was found that the control of bioparticle size could be successfully achieved by phosphate-limiting culture condition. Penicillin production under this condition was also observed to be maintained at a high level (about 80% of the maximum) for at least 1 month.

    Notes: A study of molecular orientation in a series of five one-way-drawn sheets of poly(ethylene terephthalate) is reported. Five orientation averages Plmnr which characterize the benzene-ring orientation were determined directly by Raman spectroscopy. Two further averages P202r and P222r were determined from a combination of Raman spectroscopy and refractive index measurements on the basis of two different conformational models. As in a previous publication, the seven averages Plmnr were used to estimate all the Plmnr up to fourth order for the benzene rings on the assumption that in each case the actual distribution of orientations is close to the most probable distribution. The seven orientation averages Plmnr for each drawn sheet were also compared with those calculated on the basis of two simple models for the development of molecular orientation during drawing, the pseudoaffine deformation scheme and the rubber network model. It was shown that the orientation of the chain axes is closer to that predicted by the rubber network model, the pseudoaffine deformation scheme greatly overestimating the degree of chain biaxiality. The results also show that the benzene ring planes are preferentially oriented toward the planes of the sheets. The chain axis orientation is, however, nearly uniaxial and similar to that observed for uniaxially drawn samples, suggesting that there is no direct connection between the processes of chain axis orientation and preferential orientation of the benzene-ring planes.
