    Notes: Summary We report two male cousins with Duchenne muscular dystrophy (DMD) in whom cytogenetic studies have shown a small interstitial deletion at Xp21. The lesion is readily detectable in patients and carriers by flow cytometry which indicates that approximately 6000 kb of DNA are deleted in each case. The DNA markers OTC, C7, and B24 are present in the deleted X chromosome but 87-8, 87-1, and 754 are absent. Despite apparently identical deletions one affected boy has profound mental handicap while the other is only mildly retarded. The results confirm the assignment of familial DMD to Xp21 and illustrate the value of flow cytometry in improving the precision of chromosome analysis. We have also undertaken flow cytometry in a cell line from a previously reported DMD patient with a de novo Xp21 deletion who had, in addition, chronic granulomatous disease, retinitis pigmentosa, and the McLeod syndrome. The results indicate that the amount of DNA deleted from the X is similar in both families despite the striking differences in phenotype.

    Notes: Abstract Perovskite (CaTiO3) has been exposed to hydrothermal chemical attack in aqueous solution. Dependences on temperature (150 to 250°C) and on duration of attack (1 to 35 days) have been investigated. It was found that a precipitate surface layer was formed. The thickness of this layer ranged from a few monolayers to several hundred nanometres for the least and the most severe hydrothermal conditions, respectively. The composition of the layer, as deduced by surface analytical techniques, suggests that (i) there is congruent dissolution of the perov skite surface, (ii) a TiO2 precipitate layer is formed, and (iii) the presence of silica and CO2 in solution do not appear to affect substantially the mechanism or the rate of dissolution.

    Notes: Summary Changes in the abundance and form of endoplasmic reticulum in the three major cell types of the maize root cap were investigated by stereological and stereometric techniques. Quantification from thin sections was by the modification and application of standard morphometric procedures. This revealed dramatic increases in both the volume fraction and surface densities of endoplasmic reticulum as the meristem cells differentiate into starch and secretory cells. A stereometric technique for analysing thick sections was used to assess changes in the types of endoplasmic reticulum as cells differentiate through the root cap. This procedure showed that the proportions of cisternal endoplasmic reticulum to tubular endoplasmic reticulum was highest in the peripheral secretory cells. Electron opacity of the endomembrane system was enhanced by selective staining with zinc iodide and osmium tetroxide (ZIO).

    Notes: Abstract Similarities between simple shear and pure shear or planar extension are exploited to derive equations relating stress in pure shear at constant extension rate to the stress in simple shear at constant shear rate. For the class of materials considered it follows that there are only two independent material functions required to describe simple shear. The relationships derived may also be used to estimate the ratio of first to second normal-stress differences in simple shear using experimental results from pure shear experiments.

    Notes: Abstract We compared the structure of P.II proteins of gonococcal strain FA1090 by N-terminal sequence analysis of purified proteins and by DNA sequencing of cloned P.II genes. Regulation of P.II gene expression does not involve major DNA rearrangements, but may involve generation of frame-shifts in unexpressed P.II genes. There are probably 8 or 9 P.II genes, each possessing a common leader sequence, in the gonococcal chromosome.

    Notes: Abstract The murine locus corresponding to the human Duchenne/Becker muscular dystrophy (DMD) gene has been regionally mapped on the mouse X chromosome by hybridizing DNA from interspecies mouse crosses with a cDNA clone for the mouse Dmd gene. The results demonstrate that the relative organization of genes on the murine and human X chromosomes is more divergent than has previously been postulated. Furthermore, the mouse Dmd gene maps to a similar region of the X chromosome as does the mouse muscular dystrophy mutation mdx, providing further evidence that the mdx mutant may be a murine equivalent of human DMD. However, Southern analysis of portions of the mouse Dmd gene has not yet revealed any differences between mdx and wild-type mice.

    Notes: Abstract The removal of nitrilotriacetic acid (NTA) dosed to two full-scale septic tanks each connected to a single dwelling was monitored weekly over a period of 75 weeks. Nitrilotriacetic acid in the septic tank effluent was detected in samples diluted at least 25 times by a differential pulse polarographic technique. After an initial period of stabilization, concentrations of NTA in the effluent at both sites were always 〉 10 mg L−1. Mean five-weekly estimated removals of NTA during the last 50 weeks of the study ranged from 33 to 52%. Removal of NTA was probably by solids adsorption only as no acclimatization period or overall increase in removal during the investigation was apparent.
