    An Organofluorophosphate-Hydrolyzing Activity in Tetrahymena thermophila (1985)

    Notes: An enzymatic activity that hydrolyzes O,O-diisopropylphosphofluoridate (DFP) and O-1,2,2–trimethylpropylmethyl-phosphonofluoridate (Soman) was discovered in the ciliate protozoan Tetrahymena thermophila. The enzymatic activity classifies the protein as Mazur-type similar to that found in hog kidney and Escherichia coli. The rate of hydrolysis of Soman by the Tetrahymena-extract is the highest, on a per gram of extract basis, of any eucaryote. The molecular weight is approximately 75,400 as determined by Sephacryl column chromatography. A maximum fifteen-fold purification has been achieved.Potential exists for the detoxification and one-step detection of common organofluorophosphate pollutants. Additionally, Tetrahymena should prove an easier subject for manipulation than mammalian or squid sources. Protozoa may be a potentially important source of detoxification and degradation enzymes for other environmental contaminants.

    Notes: Human erythrocytes infected with five strains of Plasmodium falciparum and Aotus erythrocytes infected with three strains of P. falciparum were studied by thin-section and freeze-fracture electron microscopy. All strains of P. falciparum we studied induced electron-dense conical knobs, measuring 30–40 nm in height and 90–100 nm in diameter on erythrocyte membranes. Freeze-fracture demonstrated that the knobs were distributed over the membrane of both human and Aotus erythrocytes. A distinct difference was seen between the intramembrane particle (IMP) distribution over the knobs of human and Aotus erythrocyte membranes. There was no change in IMP distribution in infected human erythrocyte membranes, but infected Aotus erythrocytes showed an aggregation of IMP over the P face of the knobs with a clear zone at the base. This difference in IMP distribution was related only to the host species and not to parasite strains. Biochemical analysis demonstrated that a higher proportion of band 3 was bound to the cytoskeleton of uninfected Aotus erythrocytes than uninfected human erythrocytes after Triton X-100 extraction. This may account for the different effects of P. falciparum infection on IMP distribution in the two different cell types.

    Notes: Research relating to sequencing rules in simple job shops has proliferated, but there has not been a corresponding proliferation of research evaluating similar sequencing rules in more complex assembly job shops. In a simple job shop, all operations are performed serially; but an assembly shop encompasses both serial and parallel operations. As a result of the increased complexity of assembly shops, the results associated with the performance of sequencing rules in simple job shops cannot be expected for an assembly shop. In this paper, 11 sequencing rules (some of which are common to simple job shops and some decigned specifically for assembly shops) are evaluated using a simulation analysis of a hypothetical assembly shop. The simulation results are analyzed using an ANOVA procedure that identifies significant differences in the results of several performance measures. Sensitivity analysis also is performed to determine the effect of job structure on the performance of the sequencing rules.

    Notes: Linear programming and sensitivity analysis in linear programming have long been integral parts of business school curricula, both at the undergraduate and graduate levels. Most business school students are required to take an introductory course in management science emphasizing linear programming and sensitivity analysis. This note is educational and presents a non-dual approach to restoring the optimal solution to a linear program when changes in the right-hand-side vector render the current solution infeasible and thus nonoptimal.

    Notes: The effects of the addition and withdrawal of gibberellic acid (GA3) and Ca2+ on enzyme synthesis and secretion by barley (Hordeum vulgare L. cv. Himalaya) aleurone layers were studied. Incubation of layers in GA3 plus Ca2+ affects the total amount of secreted α-amylase (EC 3.2.1.1) and acid phosphatase (EC 3.1.3.2) by promoting the appearance of different isoenzymic forms of these enzymes. The release of α-amylase isoenzymes 1–4 in response to GA3 plus Ca2+ has a lag of 6 h. When layers are incubated in GA3 alone for 6 h prior to the addition of Ca2+, isoenzymes 1–4 appear in the medium after only 30 min. When the addition of Ca2+ to layers pretreated in GA3 is delayed beyond 12 h, its effectiveness in stimulating the synthesis and release of isoenzymes 3 and 4 is diminished. After 35 h of preincubation in GA3, addition of Ca2+ will not stimulate synthesis of α-amylase isoenzymes 3 and 4. Aleurone layers preincubated for 6 h in GA3 will respond to Ca2+ when the GA3 is withdrawn from the incubation medium by producing α-amylase isoenzymes 1–4. The converse is not the case, however, since layers preincubated in Ca2+ for 6 h will not produce all isoenzymes of α-amylase when subsequently incubated in GA3. The Ca2+-stimulated release of α-amylase from GA3 pre-treated layers is dependent on the time of incubation in Ca2+ and the concentration of the ion. The response to Ca2+ is temperature-dependent, and other divalent cations such as Mg2+ cannot substitute for Ca2+. We conclude that Ca2+ influences α-amylase release by influencing events at the biochemical level.

    Notes: Abstract Chromosomal DNA from Streptococcus mutans strain Ingbritt (serotype c) was cloned into the bacteriophage λ replacement vector L47.1. The bank of recombinant phage was screened for the presence of plaques in which sucrose-hydrolysing (sucrase) activity was expressed. Two distinct sucrase-expressing recombinants were identified. In one type, designated scr, the product is an invertase-like enzyme. This enzyme was purified from lysates of recombinants and shown to have an apparent Mr of 59000. The second class of recombinants was found to express a glucosyltransferase, identical in size, as well as in enzymatic and antigenic properties, to the previously described product of gtfA.

    Notes: Abstract In comparison with the wild-type, mutants of Rhodopseudomonas sphaeroides defective in bacteriochlorophyll synthesis fail to alter their lipid composition on shifting from non-photosynthetic to photosynthetic growth conditions. The earlier the lesion in the bacteriochlorophyll synthetic pathway, the more severe the effect on membrane lipid composition, indicating that acyl lipid and pigment syntheses are co-ordinated and linked to pigment-protein complex assembly.
