    Notes: Splenocytes from young adult or old C57BL/6NNia mice were stimulated in vitro with the anti-CD3∊ mAb, 145-2C11, in either soluble (2C11s) or plate-bound (2C11i) form. In the young group, each mode of cell activation resulted in peak DNA synthesis at ∼48 h of culture; at this time point, the old group exhibited response levels to 2C11s or 2C11i that were ∼40% of those in the young group. However, in the presence of 2C11i, splenocytes from old donors showed a delayed peak response which approached the peak levels attained in the young group. To analyze the responsiveness of the CD4+ T cell subpopulation, this cell type was isolated from spleens of young or old mice and was stimulated in vitro with 2C11s or 2C11i, in the presence or absence of added accessory cells (T cell-depleted, irradiated splenocytes). The induction of DNA synthesis by 2C11s was accessory cell dependent, and the response in the old group were markedly reduced in comparison to those in the young group. In contrast, stimulation of DNA synthesis with 2C11i was relatively accessory cell independent, resulted in higher response levels in both age groups, and lessened the disparity between age groups. The analysis of IL-2 and IL-4 secretion by stimulated CD4+ cells revealed that, in response to 2C11s and accessory cells, only IL-2 accumulation was detectable and the levels in the young group were ∼10-fold higher than the IL-2 levels in the old group. However, stimulation of CD4+ cells with 2C11i and accessory cells yielded improved IL-2 production and a detectable IL-4 response in the old group, whereas the young group exhibited a response profile similar to that induced by 2C11s. Further analysis of the IL-2, IL-4, and IFNγ mRNA levels in 2C11i-stimulated CD4+ cells revealed that old donor cells accumulated similar levels of IL-2 transcripts, but higher levels of IL-4 and IFNγ transcripts, than young donor CD4+ cells. Finally, we analyzed splenic CD4+ cells for membrane expression of four molecules - 3G11, 6C10, CD45RB, and CD44 - thought to demarcate CD4+ cell subsets with restricted patterns of cytokine production. The CD4+ cell fraction of individual mice contained higher percentages of cell phenotypes associated with increased IL-4:IL-2 production ratios (i.e., 3G11lo, CD45RBlo) and with increased IFNγ synthesis (i.e., CD44hi). Taken together, these data show marked alterations in the CD4+ cell subset composition in old mice, detected at the levels of subset marker expression and profiles of cytokine production. Moreover, conclusions regarding CD4+ cell competency in old donors can differ depending on the choices of stimuli and readouts for cell function in the experimental design. Therefore, age-related differences in T cell reactivity in vitro may be partially explained by the shifts in the representation of individual CD4+ subsets, each with potentially unique activation requirements and functional attributes.

    Description: The neutral wind pattern over the summer polar cap can be driven by plasma convection to resemble the convection pattern. For a north-south component of the interplanetary magnetic field Bz directed southward, the wind speeds in the conducting E-region can become approximately 25 percent of the electric field drift speeds. If convection ceases, this neutral wind distribution can drive a significant polar cap current system for approximately 6 hours. The currents are reversed from those driven by the electric fields for southward Bz, and the Hall and field-aligned components of the current system resemble those observed during periods of northward Bz. The current magnitudes are similar to those observed during periods of small, northward Bz; however, observations indicate that electric fields often contribute to the currents as much as, or more than, the neutral winds.

    Description: Data from the Nimbus 7 Limb Infrared Monitor of the Stratosphere (LIMS) for the period October 25, 1978-May 28, 1979 are used in a descriptive study of ozone variations in the middle stratosphere. It is shown that the ozone distribution is strongly influenced by irreversible deformation associated with large amplitude planetary-scale waves. This process, which has been described by McIntyre and Palmer as planetary wave breaking, takes place throughout the 3-30 mb layer, and poleward transport of ozone within this layer occurs in narrow tongues drawn on the tropics and subtropics in association with major and minor warming events. These events complement the zonal mean diabatic circulation in producing significant changes in the total column amount of ozone.

    Description: Cross-spectral analysis of low-latitude average Nimbus 7 stratospheric and mesospheric sounder (SAMS) temperature deviations versus Nimbus 7 solar backscattered ultraviolet (SBUV) solar 205 nm flux measurements in the 1-3 mbar pressure range yields significant coherence at periods near 27 and 13 days. This supports earlier correlative evidence that the ozone response to solar ultraviolet variations is supplemented by a coupled temperature response in the upper stratosphere. Comparisons of improved one-dimensional radiative photochemical model calculations with ozone and temperature response measurements at low latitudes yields agreement only in the case of ozone at levels below 3 mbar. An additional, presumably dynamical, component of the upper stratospheric response is suggested. Cross-spectral analysis of low-latitude average SAMS temperature deviations versus higher-latitude temperature fluctuations of opposite sign in the winter hemisphere also yields significant coherency at periods near 27 and 13 days. Latitudinal temperature oscillations of this type result from interference between stationary and traveling wave components at stratospheric heights. It is therefore hypothesized that solar ultraviolet variations may force or modulate traveling waves with periods near 27 and 13 days in the upper stratosphere. The existence of such waves with higher amplitude near solar maximum than near solar minimum may help to explain observed solar cycle variations of zonally averaged winds and temperature in the upper stratosphere.

    Description: Moore et al. (1982) described a number of unexpected effects which were observed during the first Argon Release Controlled Study (ARCS 1, or rocket flight 29:014). The present paper provides a description of detailed analyses of the interaction of the argon beam with the ionosphere. An important feature of the considered test was that all detectors and the Ar(+) gun remained attached to the rocket throughout the flight. It is pointed out that the most dramatic effect of ion gun operation on ARCS 1 involved large changes in the fluxes of electrons with energies below about 600 eV. The observations are discussed, taking into account the distribution functions, azimuth dependence, and electron and ion trajectories. Attention is given to the perpendicular ion beam, the parallel ion beam, the acceleration of downgoing and upgoing electrons, and aspects of wave generation.

    Notes: A cell surface UDP-galactose:N-acetylglucosamine galactosyltransferase (GT) has been directly localized on bovine cells in tissue culture by immunohistochemical techniques. A conventional rabbit heteroantiserum was prepared against an affinity-purified soluble form of GT from bovine milk, and a monospecific IgG fraction was isolated by affinity chromatography on a GT adsorbent. As demonstrated by indirect immunofluorescence, antigen to this antibody is present on the surface of all three bovine cell lines tested. It was uniformly distributed over the exposed membrane surface of fixed cells. Exposure of living cells to the anti-GT antibody resulted in its time-dependent aggregation in the plane of the membrane. Antigen (GT) was released from the membrane surface by trypsin digestion, and its reappearance required protein synthesis, since cycloheximide effectively prevented repopulation of the cell surface.

    Notes: Interleukin 4 (IL-4) is a product of activated T cells and mast cells with effects on immunologic and hematopoietic processes. We now report that IL-4 inhibits the formation of osteoclasts from murine bone marrow cells cocultured with stromal cells. Numerous (3,000-4,000 cells/2 cm2) tartrate-resistant acid-phosphatase-positive multinucleated cells with the capacity to generate cAMP in response to salmon calcitonin (ED50 = 10-10 M) developed within 10-12 days of culture. IL-4 (ID50 = 10 U/ml) inhibited osteoclast generation in doses similar to those that induce proliferation of IL-4-responsive T cells. Additionally, the rat antimurine IL-4 monoclonal antibody 11B11 antagonizes the IL-4-inhibitory effect on osteoclast formation. These results suggest that IL-4 impedes agonist-induced in vitro bone resorption by inhibiting osteoclastogenesis.

    Notes: The effect of 6-dimethylaminopurine (6-DMAP) on germinal vesicle breakdown (GVBD) and maturation in bovine oocytes was investigated in this study. This puromycin analog has been shown to be an inhibitor of phosphorylation. Whereas GVBD occurred in nearly all oocytes (96.8%; 120/124) in control medium, presence of 6-DMAP (2 mM) blocked this process almost completely, irrespective of the presence (98.3% GV, 349/355) or absence (97.1% GV, 165/170) of cumulus cells. When lower concentrations of 6-DMAP were used (100-500 μM), GVBD was observed in 87.9% of oocytes, but their maturation was arrested at late diakinesis-metaphase I stage. The inhibition of GVBD was fully reversible, but most of the metaphase II plates were abnormal (80%). To assess whether the action of 6-DMAP is different from the inhibitors of protein synthesis, metaphase II oocytes were exposed to either cycloheximide or 6-DMAP, respectively. Whereas in cycloheximide-supplemented medium approximately 80% of the oocytes were activated, parthenogenetic activation was much less frequent after incubation in 6-DMAP (14.5%). Fusion studies showed that, even if GVBD occurs in 6-DMAP supplemented medium, the level of the maturation-promoting factor (MPF) is decreased. These experiments may indicate the importance of phosphorylation for GVBD in cattle oocytes.

    Notes: The rate of cleavage and the onset of embryonic transcription of bovine embryos cultured in vitro (IVC) has been investigated. Embryos were derived from in vitro matured, in vitro fertilized oocytes (IVM/IVF) to improve developmental synchrony. The rate of cleavage was assessed by morphological evaluation between the one- and eight- to 16-cell stage. The rate of cleavage was found to be equivalent to that reported for in vivo recovered embryos. To assess the onset of embryonic transcription, embryos were cultured to the eight- to 16-cell stage in the presence of α-amanitin for various periods of time followed by two-dimensional polyacrylamide gel electrophoresis. Embryos readily cleaved to the eight- to 16-cell stage in the presence of inhibitor α-Amanitin-sensitive protein synthesis was first detected at 36-48 h post-insemination (hpi) and continued up to 84 hpi. We conclude that bovine embryos produced by IVM/IVF/IVC are competent to initiate embryonic transcription at 36-48 h post-insemination and suggest that in vitro-induced cleavage arrest is not due to failure of the embryonic genome to initiate transcription.

    Notes: Conversion from procollagen to collagen is a specific process that is a requirement for proper alignment of collagen molecules to form functional fibers. This process is catalyzed by at least three structurally and functionally distinct enzymes cleaving collagen types I-III. The cleavage processes possibly taking place in the more recently discovered collagen types are not known to any extent at this time.Two amino-terminal proteinases, one cleaving type I and type II procollagens and the other cleaving type III procollagen, have been purified close to homogeneity, and the more unspecific activity of carboxy-terminal proteinase has been isolated from several tissues. In our experimental model, however, cleavage of the carboxy-terminal propeptides of types I and III procollagen is differently affected by lysine. This suggests the presence of at least two distinct enzymes for the removal of carboxyl-terminal propeptides.The regulation of the reaction process from procollagen to collagen is not well known at present. The importance of the phenomenon in terms of fibril formation, however, is demonstrated by several elegant studies in vitro; and certain genetic disorders in which this process is defective demonstrate the significance in vivo. Moreover, the factors shown to effect the cleavage process may be potentially beneficial in the treatment of the pathological processes with abnormal collagen accumulation such as fibrosis.In this paper we briefly review the current knowledge of the converting enzymes, including some very recent findings of our laboratory as well as the evidence presented for the biological significance of the conversion process.
