    Notes: Abstract An experimental method is described for the phase determination of x-ray reflections from noncentrosymmetric crystals. This method results from considerations on the phase dependence of line profiles in three-beam reflection-type diffractions for wavelength λ above and below a critical absorption edgeλ E of the heaviest constituent atom in the crystals. A relationS p =S L ·S R for phase determination is derived theoretically and verified experimentally forλ〈λ E ,S p being the sign of the sine of invariant phase,S L the sign defined from the line profile, andS R determined by the rotation of the crystal lattice.

    Notes: Summary Although the presence of magnetite in their tissues is correlated with the ability of different species to detect magnetic fields, proof that the magnetite is involved in magnetoreception has not yet been provided. Using the approach employed to localize and isolate magnetic particles in the yellowfin tuna, we found that single-domain magnetite occurs in chains of particles in tissue contained within the dermethmoid cartilage of adult chinook salmon,Oncorhynchus tshawytscha. The particles are present in sufficient numbers to provide the adult fish with a very sensitive magnetoreceptor system. Magnetite in the chinook can be correlated with responses to magnetic fields in a congeneric species, the sockeye salmon. Based on the presence of the chains of particles, we propose behavioral experiments that exploit the responses of sockeye salmon fry to magnetic fields to test explicit predictions of the ferromagnetic magnetoreception hypothesis.

    Notes: Abstract A new general method is proposed for determining the orientation of a parent crystal and the habit plane normals of a plate-shaped product phase without the aid of X-ray experiments. The determination starts with measuring angles between traces formed by the intersection of product phase variants with the surface of a specimen. The results reported are obtained by applying the method to three specific cases. As one of the applications, the unknown orientation of the habit plane of a 2H-stacking ordered martensite in a Ni3Sn alloy is determined as {3 3 1}.

    Description / Table of Contents: Summary The spontaneity of metal atom formation from metal oxides, both in the condensed phase, via thermal decomposition or reduction by carbon in a graphite furnace, depends on the instantaneous partial pressure of oxygen inside the furnace. Since the partial pressure of the gaseous carbon species is very low, the reaction between carbon and oxygen is determined by heterogeneous, gas-solid reaction. The factors affecting the instantaneous partial pressure of oxygen inside a pulse-heated graphite furnace have been studied by computer simulation. The results suggest that the instantaneous oxygen partial pressure inside the furnace is determined by the initial amount of oxygen and the heating rate of the furnace. The effect of oxygen on the shift of the analyte atomic absorption pulse is also discussed.
    Notes: Zusammenfassung Die spontane Bildung von Metallatomen aus Metalloxiden in der kondensierten Phase durch thermische Zersetzung oder Reduktion durch Kohlenstoff im Graphitofen ist abhängig vom Sauerstoff-Partialdruck innerhalb des Ofens. Da der Partialdruck des gasförmigen Kohlenstoffs sehr niedrig ist, wird die Reaktion zwischen Kohlenstoff und Sauerstoff durch eine heterogene Gas-festReaktion bestimmt. Die Faktoren, die den Sauerstoff-Partialdruck in einem impuls-erhitzten Graphitofen beeinflussen, wurden mit Hilfe einer Computer-Simulation untersucht. Die Ergebnisse zeigten, daß dieser Druck von der anfänglichen Sauerstoffmenge und der Aufheizgeschwindigkeit des Ofens abhängt. Die Wirkung des Sauerstoffs auf die Verschiebung des analytischen AAS-Impulses wird ebenfalls diskutiert.

    Notes: Abstract The effect of valproic acid on carbamazepine epoxidation in the perfused liver was investigated in two separate studies. In study I, significant decreases were observed both in the intrinsic clearance of carbamazepine and the intrinsic formation clearance of carbamazepineepoxide in the presence of therapeutic concentrations of valproate. The same inhibitory effect of valproate was also observed in liver preparations from a group of animals pretreated with carbamazepine. Study II focused on the effect of valproate and carbamazepine on the apparent Michaelis-Menten parameters (V max,m,Km,m)associated with the intrinsic formation clearance of carbamazepineepoxide in the perfused liver. Valproate had no statistically significant effect on either the V max,morthe Km,m of epoxidation, although the Km,m value was 43% higher in the presence of valproate. However, the ratio of V max,m and Km,m (intrinsic formation clearance) was significantly reduced by valproate. The V max,m and Km,m values obtained in study II predicted a significant decrease in the intrinsic formation clearance of carbamazepineepoxide, consistent with the results of study I. Carbamazepine pretreatment was associated with significant increases in apparent V max,m and Km,m of epoxide formation.

    Description / Table of Contents: Résumé Du mycélium prélevé à la périphérie de colonies dévoloppées sur cellophane ou à partir de cultures liquides agitées a été utilizé pour la préparation de protoplastes d'Agaricusbisporus,Auricularia auricula, Lentinus edodes, Pleurotus sajor-caju, Volvariella bombycina etV. volvacea. Les cellules mycéliales ont été traitées par deux enzymes mycolytiques, Novozym 234 et lywallzyme. Par traitement avec le lywallzyme, des protoplastes ont été obtenus avec tous les champignons comestibles étudiés,P. sajor-caju donnant le rendement le plus élevé (3.84×107/ml), suivi parAu. auricula (7.46×106/ml,Ag. bisporus (2.16×106/ml etV. volvacea 1.92×106/ml). Avec le Novozym 234,Ag. bisporus a donné le rendement en protoplastes le moins élevé. Les effets de différentes molarités de stabilisateurs osmotiques ont également été étudiés. La méthode de la cellophane est simple et rapide et peut être employée comme procédé de degrossissage. Les rendements en protoplastes obtenus à partir des cultures liquides sont habituellement plus élevés que ceux à partir des cultures sur cellophane.
    Abstract: Resumen Para el aislamiento de protoplastos de cepas deAgaricus bisporus, Auricularia auricula,Lentinus edodes, Pleurotus sajor-caju, Volvariella bombycina, y V. volvacea se utilizo micelio periferico de colonias que estaban en crecimiento activo en celofan y en frascos de agitación. Las celulas del micelio se tratarón con dos enzymas mycoliticos: Novozym 234 y lywallzyme. Todas las setas comestibles ensayadas produjerón protoplastos. Mediante tratamiento con lywallzyme la mejor cosecha de protoplastos la proporcionoP. sajor-caju (3.84×107/ml) seguido porAu. auricula (7.46×106/ml),Ag. bisporus (2.16×106/ml) yV. volvacea (1.92×106/ml). Con Novozym 234Ag. bisporus fué la de menor rendimiento. También se estudiarón los efectos de distintos estabilizadores osmoticos a diferentes molaridades.
    Notes: Summary Mycelium from the periphery of actively growing colonies on cellophane and from shake flask cultures was used for the isolation of protoplasts from strains ofAgaricus bisporus,Auricularia auricula, Lentinus edodes, Pleurotus sajor-caju, Volvariella bombycina andV. volvacea. The mycelial cells were treated with two mycolytic enzymes, Novozym 234 or lywallzyme. Protoplasts were produced from all the edible fungi tested.Pleurotus sajor-caju gave the highest yield (3.84×107/ml), followed byAu. auricula (7.46×106/ml),Ag. bisporus (2.16×106/ml) andV. volvacea (1.92×106/ml), when treated with lywallzyme.Agaricus bisporus gave the smallest yield of protoplasts when Novozym 234 was used. The effects of different molarities of osmotic stabilizers were also studied. The cellophane method is simple and quick and can be used as a screening procedure. The yields of protoplasts obtained from liquid cultures were usually higher than those from cellophane cultures.

    Description / Table of Contents: Résumé Isolement et réversion des protoplastes de Pleurotus sajor-caju Les conditions pour l'isolement de protoplastes dePleurotus sajor-caju et pour leur réversion ont été déterminées. Des protoplastes sphériques et osmotiquement sensibles ont été préparés à partir du mycélium par action de Novozym 234 et de cellulase CP en milieu osmotiquement approprié. Le rendement maximum en protoplastes est obtenu à partir du mycélium jeune, en phase exponentielle précoce, dans un tampon phosphates 0,01 M à pH 5,0, contenant MgSO4 0,6 M. Pour la réversion des protoplastes, l'agent le plus favorable est le manitol. Deux modes de développement morphologique des protoplastes en milieu osmotique ont été constatés. Toutefois, la fréquence de réversion est faible (4 à 5%). Il ne semble pas que la fréquence de réversion soit significativement différente lorsque les protoplastes sont cultivés en milieu complet ou en milieu minimal.
    Abstract: Resumen Aislamiento y reversión de protoplastos en Pleurotus sajor-caju Se determinaron las condiciones para el aislamiento y reversión de protoplastos enPleurotus sajor-caju. Protoplastos esféricos y osmóticamente sensibles se obtuvieron a partir de micelio mediante la action de Novozym 234 y celulasa CP en un medio osmótico adecuado. El máximo numéro de protoplastos fueron obtenidos a partir de miclio joven en la primera parte de la fase exponencial de crecimiento utilizando 0.6 mol/l MgSO en un tampón fosfato (0.01 mol/l) a pH 5.0. Manitol parece ser adecuado para la reversion dePleurotus. Se observaron dos patrones de desarrollo morfológico de los protoplastos en el medio osmótico líquido. La frecuencia de reversion fue, sin embargo, baja (4–5%). No se observaron diferencias significativas entre los protoplastos obtenidos en el medio completo y los que lo fueron en un medio mínimo.
    Notes: Summary The conditions for protoplast isolation and reversion inPleuro tus sajor-caju were determined. Spherical, osmotically sensitive protoplasts were released from the mycelia through the action of Novozym 234 and cellulase CP in a suitable osmotic solution. A maximum protoplast yield was obtained from young mycelia at an early exponential growth phase by the use of 0.6 mol/l MgSC4 in a 0.01 mol/l phosphate buffer at pH 5.0. Mannitol appeared to be more suitable forPleurotus protoplast reversion. Two patterns of morphological development of the protoplasts were observed in the osmotic liquid medium. The reversion frequency was, however, low at 4–5%. There appeared to be no significant difference in the reversion frequencies of the protoplasts grown on the complete medium and on the minimal medium.
