    Notes: The morphological and ultrastructural changes that occur during preparation of porcine, bovine, and murine spermatozoa for flow cytometric quantification of the relative DNA content of the X- and Y-chromosome-bearing sperm populations were examined. Ejaculated spermatozoa from the boar and bull were washed using a series of dimethyl sulfoxide (DMSO) solutions prior to fixation, whereas the epididymal mouse spermatozoa were washed only in phosphate-buffered saline (PBS). Spermatozoa from all three species were then fixed in ethanol and processed for fluorochrome staining by a treatment regimen consisting of sulfhydryl reduction and proteolysis. The processed sperm nuclei were stained for DNA with the fluorochrome, 4′-6-diamidino-2-phenylindole (DAPI) before quantification by flow cytometry. Scanning and transmission electron micrographs of sperm heads taken at various steps of the preparation and staining procedures show 1) that the rigorous washing procedure disrupted the plasma and outer acrosomal membranes, 2) that ethanol fixation resulted in removal of the outer membranes and disintegration of the nuclear envelope, and 3) that thiol and proteolysis treatment removed the remaining cellular organelles including the tail and rapidly induced partial decondensation of the tightly packed chromatin. Sequential micrographs showed that the nuclear matrix of all three species increased in thickness about twofold during the preparation and staining. Consequently, the harsh procedures currently used for quantitative staining of DNA for high-resolution flow cytometric analyses destroy most cellular organelles and thereby prevent simultaneous characterization of DNA content and other sperm cell constituents.

    Description: The detection of the J = 10 manifold of the pure rotational band of PH3 on Saturn is reported. The observations were made from the far-infrared cooled grating spectrometer. The wavelengths and observed brightness temperatures for the full disk plus rings are 89 + or - 3 K at 97.04 micrometer, 77 + or - 3 K at 102.72 micrometer, 77 + or - 3 K at 102.94 micrometer, and 83 + or - 3 K at 105.12 micrometers. The points of 97.04 and 105.12 micrometers establish the continuum level and the two points near 103 micrometers measure the depth of the PH3 manifold. After the flux due to the rings is subtracted, the depth of the feature is 16 + or - 6 K relative to the nearby 102 K continuum. These results are compared to theoretical models which parameterize the PH3 mixing ratio as x = x sub zero (P/P sub zero)(alpha) for P P sub zero and as x = x sub zero for P or = P, where P is the total pressure and alpha = H/h is the ratio of the dynamical scale height (H) and the scale height for decreasing the PH3 mixing ratio (h). The parameters x sub zero, P sub zero, and h were varied, as well as the H/He mixing ratio and the pressure-temperature profile. The data are well fitted using pressure-temperature profiles. The preferred values of h, P sub zero, and x sub zero imply that there is little or no PH3 above the thermal inversion and that the mixing ratio below the inversion is consistent with PH3 being 1 to 4 times overabundant relative to the solar P/H ratio.

    Description: Multitemporal Thematic Mapper, Thematic Mapper Simulator, and detailed ground truth data were collected for a 9- by 11-km sample segment in Webster County, IA, in the summer of 1982. Three dates were acquired each with Thematic Mapper Simulator (June 7, June 23, and July 31) and Thematic Mapper (August 2, September 3, and October 21). The Thematic Mapper Simulator data were converted to equivalent TM count values using TM and TMS calibration data and model based estimates of atmospheric effects. The July 31, TMS image was compared to the August 2, TM image to verify the conversion process. A quantitative measure of proportion estimation variance (Fisher information) was used to evaluate the corn/soybeans separability for each TM band as a function of time during the growing season. The additional bands in the middle infrared allowed corn and soybeans to be separated much earlier than was possible with the visible and near-infrared bands alone. Using the TM and TMS data, temporal profiles of the TM principal components were developed. The greenness and brightness exhibited behavior similar to MSS greenness and brightness for corn and soybeans.

    Description: It is pointed out that Titan, which is the second largest satellite in the solar system, is considerably larger than Mercury. It is made unique by its dense atmosphere, which consists mainly of nitrogen, although a substantial component of methane is present. The basic properties of Titan are summarized in a table. Many of the data were obtained during the close pass of Voyager 1 in November 1980. The atmospheric temperature decreases from its surface value of 94 K at a pressure of 1500 mbar to a minimum of 71 K at a height of 42 km and a pressure of 128 mbar. Details of atmospheric composition and thermal structure are discussed, taking into account chemical identifications and abundances, the vertical temperature structure, the horizontal temperature and opacity structure, and the radiative equilibrium. The upper atmosphere composition and temperature is considered along with the properties of aerosols, and meteorology and atmospheric dynamics. Titan's interior has an average density of 1.88 g per cu cm. Attention is given to Titan's surface and interior, and its formation.

    Description: The use of an image registration program in the studies of urban growth is described. This program permits a quick identification of growing areas with the overlap of the same scene in different periods, and with the use of adequate filters. The city of Brasilia, Brazil, is selected for the test area. The dynamics of Brasilia urban growth are analyzed with the overlap of scenes dated June 1973, 1978 and 1983. The results showed the utilization of the image registration technique for the monitoring of dynamic urban growth.

    Description: The urban growth of Brasilia within the last ten years is analyzed with special emphasis on the utilization of remote sensing orbital data and automatic image processing. The urban spatial structure and the monitoring of its temporal changes were examined in a whole and dynamic way by the utilization of MSS-LANDSAT images for June (1973, 1978 and 1983). In order to aid data interpretation, a registration algorithm implemented in the Interactive Multispectral Image Analysis System (IMAGE-100) was utilized aiming at the overlap of multitemporal images. The utilization of suitable digital filters, combined with the images overlap, allowed a rapid identification of areas of possible urban growth and oriented the field work. The results obtained in this work permitted an evaluation of the urban growth of Brasilia, taking as reference the proposal stated for the construction of the city in the Pilot Plan elaborated by Lucio Costa.

    Description: To estimate the one-sided leaf area index (LAI) of temperate coniferous forests using data acquired from the Daedalus Airborne Thematic Mapper, an empirical model is developed. The study area follows an environmental gradient across west-central Oregon, where leaf development varies in response to temperature and moisture. The relationship between the ratio of thematic-mapper simulator channels four and three and the leaf area index for selected closed canopy or fully stocked forest stands along the gradient is analyzed. Results show that a good relationship exists between the LAI and the IR/red ratio for conifers and that a conifer species-independent asymptotic relationship is observed between LAI and near IR/red reflectance, with near radiometric saturation occurring at an LAI of about 7-8.

    Notes: This study describes intercellular bridges in the ovaries of neonatal gerbils. Electron microscopy has revealed the presence of true intercellular bridges, connecting oogonia or oocytes, in ovaries of newborn gerbils. The cytoplasm of the intercellular channels is similar to that of the connected cells, with mitochondria, smooth and rough endoplasmic reticulum, and free ribosomes present. Lysosomes are also occasionally present in the intercellular bridges and they may be involved in early waves of oocyte atresia. An electrondense substance, 350-500 Å thick, is located immediately beneath the unit membrane of the intercellular bridges. Accumulation of electron-dense material increases the thickness of the walls of the intercellular bridges, supporting and maintaining the patency of the channels. It is suggested that the intercellular channels probably allow the interchange of nutrients, organelles, and possibly regulatory materials as well.
