    Notes: Abstract Oncocnemis chandleri, O. cibalis, andO. mackiei were attracted to chemically baited traps in the field. In all three cases, (5E,7Z)-dodecadienyl acetate was a key component for attraction. Attraction ofO. chandleri to traps baited with the (5E,7Z)-dodecadienyl acetate was inhibited by addition of (Z)-7-dodecenyl acetate.O. cibalis required both (5E,7Z)-dodecadienyl acetate and (Z)-7-dodecenyl acetate for attraction. Electroantennogram responses for the three species are also reported.

    Notes: Abstract Five hybrid myeloma cell lines that secrete antibodies to plant endomembrane-bound proteins have been prepared using rat myelomas and spleen cells from rats immunized against intact endoplasmic-reticulum and Golgi-membrane preparations from Phaseolus vulgaris. Four of these lines produced antibodies which all showed identical binding patterns in Western blots, recognising polypeptides of Mr 35 000, 58 000, 70 000, 91 000 and possibly 117 000 common to both membrane types, while the antibody produced by the fifth line bound to a polypeptide of Mr 57 000. This binding pattern persisted for the antibody produced by all positive clones derived by extensive subcloning of hybridomas 2B3 and 2C3 even with subsequent growth, so these polypeptides, therefore, probably have a common antigenic site. The antibody tested from the hybridoma 2C3 and two subsequent subclones inhibited the arabinosyl transferase involved in the synthesis of arabinan, a component of the primary cell-wall matrix, so that one of these polypeptides probably represents the enzyme. Comparison of the patterns of the changes in enzyme activity with the levels of each individual polypeptide in cells induced to divide and undergo primary growth tentatively identifies the 70 000-Mr polypeptide as the arabinan synthase. Interpretation of this and previous data indicates that the induction of this enzyme activity by plant growth regulators involves de novo synthesis of the protein.

    Notes: Abstract Mean catches of spruce budworm,Choristoneura fumiferana (Clemens), moths were not significantly different among four small-tree thinning treatments of young spruce-fir-hemlock regeneration. Significant inverse relationships were found between trap catches and distances to nearby spruce-fir-hemlock overstory. Prevailing wind directions indicated that moths were attracted anemotactically to upwind pheromone sources. No definite trends were detected between catches and temperature or precipitation.

    Notes: Summary Myeloid bodies (MBs) occur in the newt (Notophthalmus viridescens) retinal pigment epithelium (RPE) and are similar to areas of specialized endoplasmic reticulum found in a variety of other cell types. The function of these structures is unknown, although a role in lipid metabolism has been strongly suggested. Random samples from conventionally-fixed and sectioned newt RPE, obtained over a 24-hr cycle (LD 12∶12), were examined by electron microscopy. Myeloid bodies appear as stacks of flattened endoplasmic reticulum-associated saccules which increase in length and number as the RPE accumulates shed outer segment material, prior to increase in the amount of stored lipid. Associations of MBs with the nuclear envelope can be related to this increased length. Myeloid bodies decrease numerically in the cell as phagosomes are removed from the cytoplasm, but a decrease in mean sectional MB area, seen in the light phase, is counteracted in darkness where individual MBs are larger than those found in the light. The total sectional area of MBs within a cell and their mean length varied depending on the lighting condition; differences were also found between eyes after extended periods of continuous light and dark. Ribosomes were found in association with the surfaces of both flattened and circular MBs, but they were consistently more densely associated with the shorter concave surfaces of curved regions. A new hypothesis for MB function is presented, which is concerned with their role in isolating toxic lipids such as retinoids, which are accumulated during phagocytosis of shed outer segment tips, and which are capable of disrupting membrane-bound systems necessary for their eventual metabolism and safe storage.

    Notes: Summary Endoplasmic reticulum-polyribosome-Golgi apparatus associations were a general feature of cells of suspension cultures of wild carrot (Daucus carota L.). Free polyribosomes occurred within the Golgi apparatus zone for all dictyosomes and with equal frequency at all levels within the stack including the most mature or trans face. When evaluated and quantified from electron micrographs, approximately 60% of the dictyosome profiles were characterized by a system of transition elements consisting of part smooth-part rough endoplasmic reticulum. These were encountered most frequently in the immediate vicinity of the immature, forming or cis face, usually toward the periphery of the stacked cisternae. Analysis of serial sections showed that those dictyosome profiles not exhibiting this characteristic did so primarily because of an unfavorable plane of sectioning. All dictyosomes examined in 5 or more serial sections revealed some type of close association with endoplasmic reticulum. Some of the associations were so close that direct connections between Golgi apparatus and endoplasmic reticulum tubules could not be excluded. Also present, especially at the forming or cis face, were small 600 nm transition vesicles with nap-like surface coats on nearly 90% of the dictyosomes examined. More than 50% exhibited spiny (clathrin-)coated vesicles at the mature or trans face.

    Notes: Summary Wild carrot (Daucus carota L.) cells, grown in suspension culture, were labeled with radioactive precursors and fractionated into constituent membranes to be analyzed for specific radioactivity. Results show rapid incorporation of [3H] leucine into endoplasmic reticulum (ER)-, Golgi apparatus-, and plasma membrane/tonoplast-enriched fractions. The time lag between incorporation into ER and its appearance in Golgi apparatus or plasma membrane/tonoplast were less than 5 minutes. With an average time of 3–4 minutes for cisternal formation estimated from studies with monensin, and an average of 5 cisternae per dictyosome (total transit time of 15–20 minutes), it was not possible to account for early incorporation of radioactivity into plasma membranes by passage of proteins from ER to plasma membrane via the Golgi apparatus. To account for the findings, it would appear that at least some proteins were delivered to the plasma membrane via the first membranes that exited (i.e., mature face vesicles) from the Golgi apparatus post-pulse and that some of these proteins had been translated and inserted into membranes at or near the mature face of the Golgi apparatus.
