    Notes: Pure pencillin N α-sulfoxide (1) and penicillin N β-sulfoxide (2) were obtained by HPLC and tested as substrates for deacetoxycephalosporin C synthetase (DXCS). Neither one of the sulfoxides was utilized under conditions of conversion of penicillin N (8) to deacetoxycephalosporin C (9). The cephalosporin C α and β-sulfoxides (3 and 4, resp.) were also prepared. Relative stabilities of the sulfoxides 3 and 4 are discussed by interpretation of the 13C-NMR spectra.

    Notes: In the crystal structure of the title compound at 98 K the dimethylamino group has pyramidal geometry. The nitrogenlone pair is not directed towards the triple bond; instead, one N—CH3 bond lies almost in the plane of the disubstituted ring, directed away from the triple bond, while the other N—CH3 bond and the lone pair are directed to opposite sides of the plane, nearer to the acetylene. There is a remarkably short intramolecular contact (2.39 Å) between a methyl H- and an acetylenic C-atom. The Taft σI parameter of the arylethynyl substituent appears to be similar to that of a carboxylic ester, judging from the bond-angle deformation at the ipso-C-atom.

    Notes: Attempting to explain the differences in the pharmacological profiles of the isomeric monohydroxy-and dihydroxy-2-aminotetralins (DHAT) which are potent dopaminergic agonists we have calculated the conformational energies of 2-aminotetralin and its N,N-dipropyl derivative using the QCFF/Pi and PCILO methods. Molecular electrostatic potential (MEP) maps based on ab initio (STO-3G) wave functions were computed for both dihydroxytetralins. Root-mean-square (rms) deviations from steric congruence between the enantiomeric 5,6- and 6,7-DHAT based either on atomic centers or on the minima in MEP near the putative points of attachment to the receptor are small, but may nevertheless be sufficient to cause differences in activity on subtypes of the dopamine receptor. N,N-dipropyl substitution influences the conformational energies of the skeleton and the preferences in the orientation of the propyl groups in the isomeric DHAT may be important for the interaction with the receptor. The HOMO energies of the isomeric HAT and DHAT do not correlate with their potencies.

Spectroscopical Investigations on the Conformational Behaviour of Some 2′ - and 3′-Halogen-substituted Pyrimidine NucleosidesA series of 2′- and 3′-halogenated pyrimidine nucleosides has been synthesized and investigated by 1H, 13C and 19F n.m.r. spectroscopy. The 1H and 13C chemical shifts for the positions 2′ and 3′ depend linearly but oppositely on the substituent electronegativities XR.The conformational equilibrium N ⇌ S of the nucleosides in solution is determined. An approximately linear correlation between the prefered gauche-gauche interaction of the exocyclic CH2OH groups and the ribose N conformation has been found.

    Notes: On the Pyrolysis of Pent-1-ene-3,3-d2The distribution of deuterium in the main products of the thermal decomposition of pent-1-ene-3,3-d2 at 550 to 650°C was studied and interpreted. The results include conclusions on kinetic isotope effects, on relative reactivities of different C—H-bonds, and on the importance of nonterminal radical addition. It is shown that butadiene-d2 detected in the isotopically labeled butadiene fraction is a reliable target molecule for the homoallylic rearrangement of the pent-1-ene-4-yl-3,3-d2 radical.

    Notes: Syntheses of 1-(β-D-Arbinofuranosyl)-pyrimidinesNew synthetic methods of 1-(β-D-arabinofuranosyl)-pyrimidines are described. 1-(β-D-arabinofuranosyl)-uracil, 1-(β-D-arabinofuranosyl)-5-fluorouracil, 1-(β-D-arabinofuranosyl)-thymine, and 1-(β-D-arabinofuranosyl)-cytosine can be obtained in a high yield by the splitting of the anhydrobond in 2,2′-anhydropyrimidines in dipolar aprotic solvents, such as HMPT, DMF, and DMSO, respectively, in the presence of activated elemental copper.Unlike the formation of cyclopyrimidiens ribonucleosides are directly transformed to the corresponding arabinofuranosyl derivatives by the reaction with diphenylcarbonate and NaHCO3 in the presence of copper. The reaction proceeds probably via an intermediate formation of the cycloproduct. Further aspects on the mechanism are described.

    Notes: The placement of ionic groups within the molecular structure of a polymer produces marked modification in physical properties. A large number of studies have been performed on these ion-containing polymers, but few have focused on the effects of anion-cation interactions (i.e., counterion binding or ionization) on hydrodynamic volume, especially as the molecular structure of the solvent and nature of counterion are varied. In this study changes in hydrodynamic volume are followed through reduced viscosity measurements as a function of the abovementioned molecular parameters.The dilute solution properties of various polyelectrolytes that contain sulfonate and carboxylate groups were investigated as a function of the counterion structure, charge density, molecular weight, and solvent structure. The polymeric materials were selected because of their specific chemical structure and physical properties. In the first instance a (2-acrylamide-2 methylpropanesulfonic acid)-acrylamide-sodium vinyl sulfonate terpolymer was synthesized and subsequently neutralized with a series of bases. Viscometric measurements on these materials indicate that the nature of the cation affects the ability of the polyelectrolyte to expand its hydrodynamic volume at low polymer levels. The magnitude of the molecular expansion is shown to be due in part to the ability of the counterion to dissociate from the backbone chain, which, in turn, is directly related to the solvent structure. The changes in solution behaviour of these inomers lend support for the existence of ion pairs (i.e., site binding) and ionized moieties on the polymer chains. Measurements performed in a variety of solvent systems further confirm this interpretation. In addition, and acrylamide-sodium vinyl sulfonate copolymer was partially hydrolyzed with sodium hydroxide to study the effect of varying the charge density at a constant degree of polymerization and counterion structure. The results show that the charge density has a significant effect on the magnitude of the reduced viscosity and dilute solution behaviour. These observations, made in aqueous and nonaqueous solvents, are related to the interrelation of hydrodynamic volume, counterion concentration, and site binding. Again the controlling factor is the degree of site binding of the counterion onto the polymer backbone. Finally, we observe that the increased hydrodynamic volume affects viscosity behavior beyond the polyelectrolyte effect regime. If the average charge density on the macromolecule is relative high and/or the molecular weight is large (≥ 106) sufficient intermolecular interactions will occur to produce rapid changes in reduced viscosity.

    Notes: Previous investigators have shown that statistical mechanical averages for configuration-dependent physical properties of long unperturbed polyoxyethylene chains are sensitive to the gauche-trans energy difference for rotation about C—C bonds. Agreement between theory and experiment could be obtained only by significant adjustment of this energy away from values predicted by semiempirical conformational energy computations. The present work examines the success of MM2 in evaluating conformational properties of long unperturbed polyoxyethylene chains. Calculations are performed which identify the rotational isomers, and their energies, for the indicated bonds in CH3OCH2CH2O—CH2—CH2—OCH2CH2OCH3. These energies are used to assign statistical weights utilized in the configuration partition function for a rotational isomeric state chain with symmetric threefold interdependent rotations. The customary generator matrix scheme is employed to evaluate the mean-square unperturbed end-to-end distance, mean-square unperturbed dipole moment, and their temperature coefficients. Contrary to computational schemes employed previously, MM2 is found to provide an estimate of the gauche-trans energy difference for rotation about C—C which is in harmony with the known dimensions and dipole moments of the unperturbed polymer. MM2 also provides good estimates for most of the other parameters required in the rotational isomeric state treatment. A notable exception is provided by the gauche-trans energy difference for rotation about the C—O bond. This energy difference is overestimated by MM2.
