Notes: Synthetic cyclic octapeptides of general structure cyclo[Glu(γOBzl)-Sar-Gly-(N-R)Gly]2 (R = n-hexyl and cyclohexyl) transport calcium ions selectively across organic phases and phospholipid membranes. We have now used proton nmr spectroscopy (360 MHz) to study the solution conformation(s) of their calcium complexes. When Ca(ClO4)2 was added to solutions of these peptides in CDCl3, nmr spectra of the resulting calcium complexes were characteristic of a single C2-symmetric conformer. From a Karplus-Bystrov analysis of vicinal coupling constants in both the peptide backbone and Glu side chain (treated as an ABCC′MX spin system), in conjuction with model-building studies, a structure was proposed in which the calcium ion is bound in an octahedral-type complex by the four (coplanar) carbonyl groups of the (all-trans) Glu-Sar and Gly-(N-R)Gly peptide bonds. Occurrence of preferred rotamers about Glu side chain Cα-Cβ bonds indicated that restricted rotation in peptide side chains arises upon calcium binding.

Notes: Copolymerization of acrylamide with allyl glycidyl ether yielded water soluble polymers containing an epoxy group; addition of a crosslinking agent, N,N′-methylenebis(acrylamide), to the copolymerization mixtures resulted in the formation of similar though water insoluble polymers. Condensation of a potent beta-adrenergic antagonist Alm—H, which contains a reactive amino group, with a polymer containing an epoxy group yeilded a pharmacologically active polymer. The polymer containing the epoxy group was also converted, by reaction with sodium thiosulfate and subsequent reduction, into a mecapto groups containing polymer which was also water soluble. The latter polymer was converted by condensation with a beta-adrenergic antagonist Alm—COCH2Br, which contains a reactive bromoacetamido group, into a pharmacologically active polymer.

Notes: Infeasible path optimization algorithms have been effective with equation-solving or simultaneous modular process simulators. Using these simulators, successive quadratic programming is applied to large nonlinear programs. However, the algorithms are not directly applicable to commercial simulators of the sequential modular type. Here an infeasible path algorithm is presented which can easily be integrated with most commercial process simulators. The optimization problem remains small (10 or 20 variables) and the user merely replaces the convergence block with an “optimization” block.A simple process example demonstrates the operation, effectiveness, and potential of this algorithm.

Notes: This study is based on the use of integral, hybrid thermal barrier to protect the core of the composite structure. Thermal barrier treatments evaluated in this study include ceramic fabric, ceramic coating, intumescent coating, hybrid of ceramic and intumescent coating, silicone foam, and phenolic skin. The composite systems evaluated in combination with thermal barrier treatments included glass/vinyl ester, graphite/epoxy, graphite/bismaleimide, and graphite/phenolic. All configurations were tested for flammability characteristics. These included smoke density and combustion gas generation (ASTM E-662), residual flexural strength (ASTM D-790), heat release rate, and ignitability (ASTM E-1354). ASTM E-662 test method covers the determination of specific optical density of smoke generated by solid materials. ASTM D-790 test method covers the determination of flexural properties of composite materials in the form of rectangular bars. ASTM E-1354 (cone calorimeter) covers the measurement of the response of materials exposed to controlled levels of radiant heating with or without an external ignitor, and is used to determine the ignitability, heat release rates, mass loss rates, effective heat of combustion, and visible smoke development. Without any fire barrier treatments, all composite systems evaluated in this study failed to meet ignitability and peak heat release requirements of MIL-STD-2031 (SH) at radiant heat fluxes of 75 and 100 kW m-2, respectively. Intumescent coating and a hybrid system consisting of intumescent coating over ceramic coating were the most effective fire barrier treatments for composite systems evaluated in this study. Using either of these treatments, all composite systems met the ignitability requirements of 90 and 60 at 75 and 100 kW m-2, respectively. Except for glass/vinyl ester, all systems also met the peak and average heat release requirements of MIL-STD-2031 (SH) at radiant heat fluxes of 25, 75, and 100 kW m-2, respectively.

Notes: Samples of polyethylene and polytetrafluorethylene were sintered at 200°C. The samples were cleaved and cut, and the two types of surfaces were studied by ESCA. The possibility of chemical interaction between the two polymers and the formation of a transitional layer is postulated on the basis of the results. A morphological model of the structure of these samples is given.

Notes: The electron energy analyser transmission (peak area/incident current) of a VG Scientific ESCALAB has been measured as a function of kinetic energy, analyser energy and source position for a point source of electrons in the sample plane (produced using a specially designed mini electron gun). Results have been obtained in both CAE (constant analyser energy) and CRR (constant retard ratio) modes. The results are extrapolated to give the energy dependence of the analyser transmission for an extended current source, such as encountered in routine XPS analysis applications. A detailed theoretical study explains the rationale behind this data extrapolation and predicts an analyser transmission in fair agreement with experimental results, possible causes for the discrepancies between theory and experiment are discussed.

Notes: N-Terminal sequence analysis of proteins separated by two-dimensional polyacrylamide gel electrophoresis and transferred onto polyvinylidene difluoride (PVDF) membranes has become the method for molecular characterization of proteins contained in biological samples. However, the proteins of lower abundance cannot be sequenced directly, without improving the technique. We have studied a drying method on several PVDF membranes including Trans-Blott, Immobilon P and Problott. Using Amido Black, Coomassie Brilliant Blue R-250 and Ponceau S, we have obtained, in comparison with the non-dried membranes, an enormous increase in the number of detectable proteins.

Notes: This publication establishes a reference human liver protein map obtained with immobilized pH gradients. By microsequencing, 57 spots or 42 polypeptide chains were identified. By protein map comparison and matching (liver, red blood cell and plasma sample maps), 8 additional proteins were identified. The new polypeptides and previously known proteins are listed in a table and/or labeled on the protein map, thus providing a human liver two-dimensional gel database. This reference map can be used to identify protein spots on other samples such as rectal cancer biopsies.

Notes: Normal human skin fibroblast primary cell lines secrete over 50 proteins into culture medium. These have been mapped previously using two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) and this technique has now been used to investigate extracellular protein secretion by human osteoblasts in vitro. We report the mapping of a number of consistent markers specific to the osteoblast. In particular, one protein chain with posttranslational modifications was found to be unique to the osteoblast extracellular protein map. The absence of the N- and O-glycoforms of collagenase from the osteoblast profile in this study concurs with findings reported using the immunoprecipitation functional assay and Northern blot analysis. The use of 2-D PAGE in phenotypic assessment provides a more complete analysis than the standard range of single-parameter tests for osteobiasts. Mapping of extracellular and cellular proteins in addition to bone matrix protein analysis will allow a comprehensive analysis of normal osteoblast function. This technique may also be applied to the study of osteoblasts in relation to bone disease and in assessing the phenotypic shift within a normal osteoblast culture.

Notes: The reference plasma protein map, obtained with immobilized pH gradients in the first dimension of two-dimensional electrophoresis, is presented. By microsequencing, more than 40 polypeptide chains were identified. The new polypeptides and previously known proteins are listed in a table and labeled on the protein map, thus providing an update of the human plasma two-dimensional gel database.
