    Notes: Summary A method has been developed for the dual staining of neutral complex carbohydrates in light microscopy. It combines an alkaline phosphatase-labeled concanavalin A-5-bromo-3-indolyl phosphate, p-toluidine salt (Con A-ALP-BIPT) method with periodic acid-Schiff (PAS) sequence. With the present dual staining method, it is possible to color α-d-glucosyl and α-d-mannosyl residues blue and 1,2-glycol groups of neutral complex carbohydrates magenta. The validity of this method has been confirmed with appropriate histochemical controls and enzyme digestions on test tissues.

    Notes: Summary In the epithelium lining the ventral prostate of the rat, complex carbohydrate-containing structures have been studied by means of both light and electron microscopic histochemical methods. According to light microscopy, the free surface and granules of different sizes in the distal cytoplasm of the epithelial cells were found to exhibit positive reactions for complex carbohydrates with 1,2-glycol and acidic groups and sialic acid residues. In addition, secretory substances within the glandular lumen were shown to exhibit positive reactions for similar groups and saccharide residues of complex carbohydrates. In electron microscopy, the surface coat of the plasma membrane, certain elements of the Golgi apparatus and lysosomal dense bodies were found to exhibit positive reactions for glycoproteins with 1,2-glycol groupings. The histophysiological significances of the carbohydrate-containing structures have been discussed with special reference to the known physiological functions of the prostate in the rat.

    Notes: Summary In the scrotal skin of the boar, the histochemistry of complex carbohydrates has been studied by means of a series of selected methods of light microscopy. The epidermis of the scrotal skin was found to contain neutral and acidic complex carbohydrates with different saccharide residues. The secretory epithelial cells and secretory substances of the saccular apocrine sweat glands contained sulfated, other acidic and neutral complex carbohydrates, whereas the secretory epithelial cells and secretory substances of the tubular apocrine sweat glands involved largely neutral complex carbohydrates. The two types of complex carbohydrates from the both glands were shown to contain commonly substantial amounts of various saccharide residues but were devoid of notable amounts of sialic acid residues. In addition, complex carbohydrates in the smooth muscle cells were reacted for relatively small amounts of saccharide residues. From the present results, the histophysiological significanses of complex carbohydrates in the particular histologic structures of the scrotum have been discussed with special reference to the functions of the skin in the boar.

    Notes: Summary A correlation between Y chromosome length and stature was statistically analyzed in a normal male population of 142 Japanese students with a mean age of 24.0 years. Evidence was obtained that increased length of the heterochromatic band Yq12 may be associated with increased height: The correlation coefficient between band Yq12 length and height was 0.17, statistically significant at the 5% level. And, taller males had longer Y chromosomes, in which the mean length of band Yq12 was significantly longer than that of shorter males. No correlation was seen between length of the euchromatic band Yq11 and stature. The present study reveals a possible effect of Yq heterochromatin on the development of body height in man.

    Notes: Summary The purpose of this investigation was to determine whether fibronectin preparations from both chicken plasma and cell surface of fibroblasts can promote phagocytosis of gelatin-coated latex particles. Chicken plasma fibronectin was isolated (a) by ammonium sulfate fractionation, chromatography on Sepharose-4B followed by purification on a Sepharose-4B-heparin column; (b) by affinity chromatography on a Sepharose-4B-rat-antifibronectin column; (c) by affinity chromatography on Sepharose-4B-gelatin followed by molecular sieve separation on Sepharose-CL4B; (d) by a dual affinity chromatographic method using a Sepharose-4B-gelatin column and a Sepharose-4B-heparin column. Chicken cell surface fibronectin from fibroblast cultures was purified by ammonium sulfate precipitation followed by chromatography on Sepharose-CL4B. The purity of preparations was examined by polyacrylamide gel electrophoresis in the presence of sodium dodecylsulfate; all samples showing high purity. The opsonic activities of the preparations were measured by the uptake of125I-labeled gelatin coated latex particles in conjunction with rat liver slice, and peritoneal macrophage monolayer systems. Both the plasma fibronectin and cell surface fibronectin preparations showed substantial opsonic activities in the test systems. Fresh chicken plasma did not reveal any phagocytosis promoting activity due to the presence of some unidentified inhibitor(s). The results showed that an opsonically active protein can be isolated from chicken plasma or serum and this protein is identical to plasma fibronectin. Furthermore, it could be concluded that cell surface fibronectin from chicken fibroblasts also can serve as an opsonin for geletin coated particles.

    Description / Table of Contents: Résumé Dans les aciers à faible ductilité et à haute résistance, le comportement à la fatigue au cours des premiers stades du phénomène est associé aux inclusions non métalliques et présente un caractère hautement localisé au voisinage de l'inclusion. Cependant, la nature du processus d'initiation d'une fissure de fatigue n'est pas claire. Dans ce mémoire, l'accent à été placé sur des différences que l'on peut rencontrer dans le mécanisme d'amorçage et dans le début de la croissance des fissures de fatigue entre un acier martensitique et des matériaux de ductilité normale. La faible liaison existant entre la matrice et une inclusion d'oxyde d'aluminium conduit à la formation d'une singularité associée à l'inclusion qui détermine une croissance de la contrainte. La fissure de fatigue prend naissance à la périphérie de cette singularité à un angle de 45° par rapport à la direction de la contrainte principale. Les observations métallurgiques et micro-fractographiques ont révélé que l'amorçage et le début de croissance des fissures de fatigue à partir d'inclusions métalliques relèvent du cisaillement plutôt que de la traction simple. On conclut donc que, bien que le processus de fatigue est entièrement localisé au voisinage d'une inclusion, le mécanisme d'amorçage et de début de croissance d'une fissure de fatigue est essentiellement le même que dans un matériau ductile. L'effet d'hétérogénéités métallurgiques dans la martensite, tel que les frontières du grain austénitique primaire, les empilements et les plaquettes, peuvent être considérés sur l'amorçage et le début de croissance des fissures de fatigue; ce point est également discuté au point de vue mécanique et métallurgique.
    Notes: Abstract In low ductility and high strength steels, the early stage fatigue behavior associated with non-metallic inclusions is a highly localized phenomenon near the inclusions. However, the nature of the fatigue crack initiation process is not clear. In this paper, a special emphasis is placed on the possible differences in the mechanism of initiation and the early growth of fatigue cracks between a martensitic steel and ordinary ductile materials. The poor adhesion between matrix and aluminum oxide inclusion leads to the formation of an inclusion pit which serves as a simple stress raiser. The fatigue crack originates at the periphery of this inclusion pit at an angle of 45 degrees to the principal stress direction. Metallurgical and micro-fractographical observations revealed that the initiation and early growth of fatigue cracks from non-metallic inclusion are of the shear rather than the tensile mode. Thus, it is concluded that, though the fatigue process is quite localized in the vicinity of inclusion, the mechanism for an initiation and early growth of fatigue cracks is essentially the same as that for ductile materials. The effects of metallurgical heterogeneities in the martensite, such as the prior austenite grain boundaries, packets, and plates on the initiation and early growth of the cracks, are also discussed from the mechanical-metallurgical viewpoint.
