    Inhibition of the metabolism of ram spermatozoa by derivatives of α-chlorohydrin (1981)

    Notes: The effects of the male antifertility agent, α-chlorohydrin, six of its derivatives, and glycidol were studied on the metabolism of washed ram spermatozoa in vitro with fructose as substrate. The α-chlorohydrin derivatives were the amino, the phosphorylated, and four glycol-bridge (ketal) compounds. All compounds except glycidol, in a concentration between 0.1 and 100 mM, reduced the aerobic glycolsis and/or oxidation of fructose. However, there was not a high correlation between the ability of these compounds to inhibit the metabolism of ram spermatozoa in vitro and their antifertility activity when administered to male rats. Other factors are clearly involved in their antifertility activity, eg, the concentration of the compounds in the epididymis and their conversion of either more or less spermicidal compounds in the body.

    Notes: The morphological and ultrastructural changes that occur during preparation of porcine, bovine, and murine spermatozoa for flow cytometric quantification of the relative DNA content of the X- and Y-chromosome-bearing sperm populations were examined. Ejaculated spermatozoa from the boar and bull were washed using a series of dimethyl sulfoxide (DMSO) solutions prior to fixation, whereas the epididymal mouse spermatozoa were washed only in phosphate-buffered saline (PBS). Spermatozoa from all three species were then fixed in ethanol and processed for fluorochrome staining by a treatment regimen consisting of sulfhydryl reduction and proteolysis. The processed sperm nuclei were stained for DNA with the fluorochrome, 4′-6-diamidino-2-phenylindole (DAPI) before quantification by flow cytometry. Scanning and transmission electron micrographs of sperm heads taken at various steps of the preparation and staining procedures show 1) that the rigorous washing procedure disrupted the plasma and outer acrosomal membranes, 2) that ethanol fixation resulted in removal of the outer membranes and disintegration of the nuclear envelope, and 3) that thiol and proteolysis treatment removed the remaining cellular organelles including the tail and rapidly induced partial decondensation of the tightly packed chromatin. Sequential micrographs showed that the nuclear matrix of all three species increased in thickness about twofold during the preparation and staining. Consequently, the harsh procedures currently used for quantitative staining of DNA for high-resolution flow cytometric analyses destroy most cellular organelles and thereby prevent simultaneous characterization of DNA content and other sperm cell constituents.

    Regulation of leucine metabolism in man: a stable isotope study (1981)

    Description: Leucine catabolism is regulated by either of the first two degradative steps: (reversible) transamination to the keto acid or subsequent decarboxylation. A method is described to measure rates of leucine transamination, reamination, and keto acid oxidation. The method is applied directly to humans by infusing the nonradioactive tracer, L-[15N,1-13C]leucine. Leucine transamination was found to be operating several times faster than the keto acid decarboxylation and to be of equal magnitude in adult human males under two different dietary conditions, postabsorptive and fed.     Lymphadenopathy associated virus infection of a blood donor--recipient pair with acquired immunodeficiency syndrome (1984)

    Description: A retrovirus isolated from three patients with the acquired immunodeficiency syndrome (AIDS) in the United States was morphologically and antigenically identical to lymphadenopathy associated virus isolated in France. Two of these isolates were from a blood donor-recipient pair, each of whom developed AIDS. Lymphadenopathy associated virus was isolated from the blood donor's lymphocytes 12 months after his onset of AIDS symptoms and from the blood recipient's lymphocytes 1 month after her onset of AIDS symptoms. Two isolates from the blood donor-recipient pair and an isolate from an epidemiologically unrelated homosexual man were examined by competitive radioimmunoassay to determine their antigenic relatedness to each other and to other human retroviruses.     Neuroleptic-induced decrease in plasma homovanillic acid and antipsychotic activity in schizophrenic patients (1984)

    Description: Plasma-free homovanillic acid, a major metabolite of dopamine, was measured in chronically ill schizophrenic patients both before and during treatment with the antipsychotic phenothiazine, fluphenazine. Neuroleptic treatment was associated with a significant time-dependent decrease in plasma homovanillic acid from pretreatment values, which were significantly elevated when compared with those of age- and sex-matched healthy control subjects. Further, both the absolute concentrations as well as the neuroleptic-induced reductions in plasma homovanillic acid determined over 5 weeks of neuroleptic treatment were statistically significantly correlated with ratings of psychosis and improvement in psychosis, respectively.     Description: Measurements of forbidden line Ar II 6.99 micron and Pf alpha 7.45 micron made from the Kuiper Airborne Observatory are presented for Sgr A, the H II region at the center of the Galaxy. These line strengths, when combined with ground-based measurements, suggest a factor of enhancement of two in the Ar/H ratio in the galactic center region relative to that in the sun and in the solar neighborhood. The accuracy of the determination is presently limited by the uncertainty in the collision strength for Ar(+).

    Detection of the J = 6 - 5 transition of carbon monoxide (1981)

    Description: The J = 6 - 5 rotational transition of carbon monoxide has been detected in emission from the KL 'plateau source' in the Orion molecular cloud. The corrected peak antenna temperature is 100 K, and the FWHM line width is 26 km/sec. These observations were carried out using the 3 m telescope of the NASA IRTF (Infrared Telescope Facility) on Mauna Kea, Hawaii, and constitute the first astronomical data obtained at submillimeter wavelengths with a heterodyne system using a laser local oscillator. The data support the idea that the high-velocity dispersion CO in Orion is optically thin and set a lower limit to its temperature of approximately 180 K.

    Evolution of the ultraviolet coma of comet Austin (1982g.) (1984)

    Description: Ultraviolet spectra of comet Austin (1982) obtained in July 1982 at heliocentric distances ranging from 1.10 to 0.81 AU preperihelion with the IUE reveal that the comet is very similar in chemical composition and appearance to comet Bradfield (1979X). In addition, the derived H2O production rate is found to vary with heliocentric distance as r to the -3.6, similar to the r to the -3.7 behavior found for comet Bradfield. It is pointed out, however, that the limited sample rate precludes the observation of the short-term variations which could be produced by a rotating nonuniform cometary nucleus.

    Far-infrared spectroscopy of the galactic center - Neutral and ionized gas in the central 10 parsecs of the Galaxy (1984)

    Description: The present mapping of the 3P1-3P2 fine structure line emission from neutral atomic oxygen near the galactic center shows the emission to be extended over more than 12 pc along the galactic plane, centered on the position of Sgr A West. The rotational velocity of the O I gas at R of about 1 corresponds to a mass within the central parsec of about 3 million solar masses. The forbidden O I line probably arises in a predominantly neutral atomic region immediately outside the ionized central parsec of the Galaxy. Gas temperatures are greater than 100 K, and the total integrated luminosity radiated in the line, which is about 100,000 solar luminosities, substantially contributes to the cooling of the gas. The 3P1-3P0 fine structure line of the O III forbidden line has also been detected at 88 microns toward Sgr A West, coming from high density ionized gas.

    Notes: This study describes intercellular bridges in the ovaries of neonatal gerbils. Electron microscopy has revealed the presence of true intercellular bridges, connecting oogonia or oocytes, in ovaries of newborn gerbils. The cytoplasm of the intercellular channels is similar to that of the connected cells, with mitochondria, smooth and rough endoplasmic reticulum, and free ribosomes present. Lysosomes are also occasionally present in the intercellular bridges and they may be involved in early waves of oocyte atresia. An electrondense substance, 350-500 Å thick, is located immediately beneath the unit membrane of the intercellular bridges. Accumulation of electron-dense material increases the thickness of the walls of the intercellular bridges, supporting and maintaining the patency of the channels. It is suggested that the intercellular channels probably allow the interchange of nutrients, organelles, and possibly regulatory materials as well.
