    Notes: Abstract An electronic circuit is described which automatically rezeroes a physiologic signal whenever it reaches the limit of a display. This modified signal can be used for polygraph monitoring while the raw signal is processed by a wide-range high-resolution a.d. convertor. The device is currently used in conjunction with the barometric method for measuring tidal volume, but should find general use in plethysmography.

    Notes: Summary The inactivation (desensitization) of the acetylcholine receptor by carbanylcholine, a stable analogue of acetylcholine, has been investigated in eel Ringer's solution, pH 7.0, 0°C, by measurements of (i) ion flux and (ii) the kinetics of the reaction of [125I]-α-bungarotoxin with the receptor. The effect of preincubation with carbamylcholine is significantly different in the two types of measurement. In both the receptor-controlled flux of inorganic ions and the toxin-binding kinetics a biphasic process has been observed (Hess, G.P., Lipkowitz, S., Struve, G.E., 1978,Proc. Nat. Acad. Sci. USA 75:1703; Hess, G.P. et al., 1975,Biochem. Biophys. Res. Commun. 64: 1018; Bulger, J.E. et al., 1977,Biochemistry 16: 684), only the initial fast phase of which is inhibited and the subsequent slow phase persists. However, preincubation with carbamylcholineper se has no effect on the toxin reaction. The results obtained are consistent with the proposal of Katz and Thesleff (Katz, B., Thesleff, S., 1957,J. Physiol. (London) 138: 65) that the active form of the receptor is converted to an inactive form in the presence of acetylcholine receptor ligands, and with our previous experiments (Hess et al., 1978) which indicated that one receptor form is responsible for the initial fast phase of both the receptor-controlled ion flux and the toxin binding reaction, and that its conversion to the other form results in the slow phases in these two measurements.

    Notes: Summary In the stimulated state, with glucose as substrate, oxygen uptake by the isolated perfused rectal gland is directly related to the rate of chloride secretion. Lactate production is negligible under aerobic conditions in the stimulated gland. A stoichiometric relationship exists between chloride transport and oxygen consumption, with a Cl/O2 ratio of about 30∶1, resembling that reported for sodium in mammalian kidneys. This ratio remains constant under varying degrees and modes of stimulation. The ratio does not change when the gland is induced to secrete chloride against varying electrochemical gradients by altering the concentration of urea in the perfusate.

    Notes: Summary Secondary mutations which increase the efficiency of suppression of nonsense mutations in the rHB cistron of bacteriophage T4 have been isolated. These secondary mutations, called context mutations, map at sites very close to the nonsense codon, possibly on the promotor distal side. In context-nonsense double mutants, the amount of suppressed gene product is increased approximately 10-fold. The context mutations examined can act on the UAA (ochre) nonsense allele as well as on the UAG (amber) nonsense allele at a given site. These context mutations affect all suppression mechanisms analyzed (genetic suppressors. 5-fluorouracil suppression and spontaneous suppression). We suggest that context mutations affect information which is significant to the termination of polypeptide chains. According to our view, context mutations change the immediate neighborhood of nonsense mutations and so reduce the degree of resemblance to the sequences normally used for the termination of translation.
