    Notes: Abstract The characteristics of slab waveguide tapers with finite-thickness cladding are investigated by approximating the tapered section as a series of uniform five-layer waveguides with successive changes in the core and cladding thickness. The behaviour of multimode coupling and transmission characteristics for typical tapers are studied in detail by computer simulation, employing parameter values appropriate for practical systems. Strong coupling and efficient conversion between core and cladding modes are demonstrated. In addition, it is shown that for tapers of moderate slopes, the nearest-neighbour coupling approximation is quite adequate for the calculation of the individual mode amplitudes. For steeper tapers, however, additional neighbouring modes have to be taken into account, and the modal energy tends to spread further towards the higher-order modes. It is also demonstrated that relatively large step-size can be used in the iterative numerical calculations. This, in conjunction with the choice of an appropriate coupling scheme (e.g., the nearest-neighbour approximation for mild tapers), makes it possible to reduce the cost of computer simulation. The relevance of the present analysis to fiber tapers is also discussed.

    Notes: Abstract A short description of a rigorous electromagnetic theory based on the numerical integration of a differential system is given. It allows the study of the stop-bands of a corrugated waveguide. Dispersion curves in the first and the second-order stop-bands are computed for various periodic waveguides. A selection rule due to the symmetry of the corrugation is found. The reflection of a plane wave by the guide is examined. It is demonstrated that under peculiar circumstances, the reflection coefficient has a modulus equal to unity.

    Notes: Abstract Kinetin, applied as a dispersion in aqueous lanolin to the stumps of decapitated stems of P. vulgaris plants with their roots removed, was found to promote the transport of 14C- and 32P-labelled assimilates to the site of hormone application. Measurement of photosynthetic rate of, and assimilate export rate from the source leaves, indicated that kinetin was not acting to promote assimilate transport by stimulating these processes. Moreover, it was found that the time between kinetin application and detection of an enhanced transport flux was independent of the distance over which kinetin would need to move to be present throughout the length of the transport pathway. These observations, together with the finding that lateral applications of kinetin to the stems resulted in an enhanced localized accumulation of assimilates, provided evidence that kinetin acted locally at its point of application to stimulate assimilate transfer.

    Notes: Abstract Isolated cotyledons of Phaseolus vulgaris L. cv. Canadian Wonder accumulated 14C2H4 (0.7–1 μl l-1) from air to give partition coefficients of 1 to 4, which greatly exceeded the value obtained with steam killed cotyledons (0.05) and with water (0.11). After 14C2H4 treatment, 98% of the 14C in the tissue remained as 14C2H4. The labelled ethylene accumulated by cotyledons was released only slowly (1–10% h-1) either in an air stream or into toluene. Heating to 60°C for 2 h, but not freezing and thawing, caused the immediate release of 14C2H4 from the tissue. Propylene and vinyl chloride competitively inhibited the accumulation of 14C2H4. Cotyledons emanated endogenous ethylene at a very low rate but after heating (although not freezing and thawing) 13 nl of ethylene per g fresh mass were released within minutes. It was concluded that french bean cotyledons hold ethylene in a compartmented form in sufficient amount to account for at least 200 h of emanation.

    Notes: Abstract Phaseolus coccineus embryos at the heartshaped and the middle cotyledonary stages were cultured in vitro on media added with different concentrations of zeatin (Z) or zeatin riboside (Zr). Growth of early embryos was clearly favored by concentrations of Z from 10-8 M to 10-5 M, lower concentrations having no effect. Zr also promoted in vitro growth of early embryos, but in concentrations from 10-12 M to 10-10 M, higher concentrations being inhibitory. More developed embryos were scarcely sensitive to the presence in the culture medium of either Z or Zr at any concentration.

    Notes: Abstract A particulate preparation from developing cotyledons of Phaseolus vulgaris L. was incubated with uridine-5′-diphospho-N-acetyl-D-glucosamine (UDP-GlcNAc; [6-3H]glucosamine), and by polyacrylamide gel electrophoretic analysis it was shown that the labeled (N-acetyl)glucosamine (GlcNAc) was incorporated into the principal reserve protein of the cotyledons, vicilin, and also into phytohemagglutinin. Some of the labeled product also reacted with antiserum to vicilin from mature seeds. In contrast it was not possible to detect the incorporation of labeled mannose from guanosine-5′-diphospho-D-mannose (GDP-mannose; [U-14C]mannose) into either of these proteins by gel-electrophoretic analysis of the mannose-labeled products, but we did observe a low incorporation of mannose into material which reacted with antiserum to vicillin. The predominant glycosylation reaction in vitro was therefore probably a transfer of GlcNAc alone, rather than in combination with mannose as preformed oligosaccharide.

    Notes: Abstract We have studied the effects on the PDA of modifying intracellular and extracellular concentrations of Ca2+ and Mn2+. The effect of decreased Ca2+ concentration or addition of EGTA is mainly an increase in the PDA amplitude and length. Raising Ca2+ concentration using ruthenium red or high external Ca2+ has the opposite effect. The effect of Mn2+ is much more striking: In the presence of 50–100 mM Mn2+ the PDA is initially greatly depressed but can rise slowly for up to 20 or 30 s (in the dark) until it approaches its original amplitude and time course. Bridge measurements showed that the depression of the PDA corresponds to a depressed conductance and so is not due to an increase in K+ conductance. The Mn2+ effect is potentiated by decreased Ca2+. Appropriate stimulation suppresses the rising PDA as promptly as it does a normal PDA, suggesting that if lateral diffusion is the source of the slow rise, the PDA and PDA-depressing processes must be spatially linked. The action of the anti-PDA is apparently prolonged by both Ca2+ and Mn2+.

    Notes: Abstract We show that the effect of an adapting light on the sensitivity of barnacle photoreceptors depends on the direction of net pigment transfer [rhodopsin (R) to metarhodopsin (M) or reverse] occasioned by the adapting light. For stimuli giving no net pigment transfer the state of the pigment appears irrelevant, R → R having the same effect as M → M. With respect to these, R → M gives enhanced facilitation and M → R depressed facilitation. This suggests a correlation with the prolonged depolarising after-potential (PDA) and the anti-PDA, which follow R → M and M → R stimuli respectively. These effects appear mainly in less sensitive cells and for higher amounts of conditioning light — but still well within the physiological range and well below the threshold for PDA and anti-PDA induction. The special interest of these results is that they appear to be interpretable only by assuming that absorption of light by metarhodopsin exerts an effect on the stimulus coincident response (LRP), the first demonstration of such an effect.
