    Notes: The karl Fischer reagent method has been used to study the drying of whale skeletal muscle myoglobin. Drying curves for metmyglobin were obtained by mechanical pumping (moderate vacuum) and diffusion pumping (high vacum) at 20, 10, 0, -10, and -20°C. Strongly held water is present in about the same amounts as were previously observed with lysozyme. Nearly all of this strongly held water is still present during the drying of acid denatured metmyoglobin. This suggest that a core structure containing strongly held water is still present at pH 4.0. Comparison of the drying of metmyoglobin, deoxymyoglobin, and oxymyoglobin indicates that the iron-coordinated water of methyoglobin is the second most strongly held water of the molocule.

    Notes: Possible conformations of X-D-alanyl4-D-alanine5 and its analogs X-L-analyl4-D-alanine5 X-D-alanyl4-L-alanine5, X-D-butyl4-D-alanine5, X-D-alanyl4-D-butyric acid5, X-D-valyl4-D-alanine5, and X-D-alanyl4-D-valine5 have been analyzed by theoretical methods. These studies suggest that L-alanine and D-valine at the 4 or 5 postion of the pentapeptide moiety of peptidoglycan will drastically reduce the cross-linking in peptidoglycan biosynthesis, whereas the effect of D- butyric acid will be marked at the 4 postition and modeate at the 5 position. This is in good agreement with experimental results. The cross-linking enzyme transpeptidase requires a specific confroamation for the 4th and 5th residues for optimal binding.

    Notes: A method for the measurement of water that is strongly held by lysozyme is described. This water is slowly removed by vaccum drying of lyophilized and can be titrated with the Karl Fischer reagent. Drying curves were obtained by mechanical pumping (moderate vacuum) and diffusion pumping (high vacuum) at 20°, 10°, 0°, -10° and -20°C. About 23 water molecules per lysozyme molecule are at least moderately held by the protein. These water molecules fall into several classes. Three to four of them are quite strongly held and may correspond to the three buried water molecules observed in the x-ray analysis of lysozyme structure. The presence of tri-N-acetylglycosamine in the lysozyme active cleft has no effect on drying at 0°C. The method shows promise of being generally applicable to the measurement of small amounts of water which are strongly held by biological structures.

    Notes: The conformation of (Pro-Gly-Phe)n in trifluoroethanol was investigated using CD, nmr and ir techniques. After making appropriate correction for the contribution of the phenylalanine chromophore to the observed CD spectra of the polytripeptide at several temperatures, it is found that (Pro-Gly-Phe)n can exist in a partially triple-helical conformation in this solvent a t low temperatures. The nmr and ir data support this conclusion. In conjunction with recent theoretical sutdies, our data offer an explanation for the preferential occurrence of the Phe residue in position 2 of the tripeptide sequence Gly-R2-R3, in collagen.

    Notes: Ultrafiltration membranes were prepared from cellulose nitrate under different conditions and were characterized using a solution of 1.0% dextran-150 having a salinity of 1500 ppm. The role of various parameters such as polymer concentration, ratio of ether to alcohol, methanol as the solvent, and other additives is systematically studied and presented. Similarly, the effect of operational and hydraulic parameters on the membrane performance is evaluated and the minimum activation energy for the solvent permeability is experimentally deduced.

    Notes: Copolymers of methacrylic acid (MAA) and a nonionic hydrophilic monomer N-vinylpyrrolidone (NVP) were synthesized by polymerization in aqueous solution in the absence of metal ions. The NVP content of the copolymers ranged from 2 to 36 mole % with sequences of MAA interrupted at random by a single unit of NVP at all compositions. The pH-induced conformational transition of these copolymers was followed by potentiometric titration and viscosity studies and the results were compared with those of pure poly(methacrylic acid) (PMAA). The negative free energy of transition from the un-ionized compact from to expanded structure showed a gradual decrease with increasing NVP content, and the collapsed conformation observable for PMAA at low degrees of ionization (0 〈 α 〈 0.3) disappeared at NVP contents greater than 15 mole%. These findings are supported by viscosity data. The results suggest that long-range methyl-methyl hydrophobic contacts still possible in higher NVP content copolymers are not sufficient to bring about the collapse of the molecule and a minimum average sequence length of about 20 MAA units is required to compact the molecule. Hydrophilic “shielding” of MAA chains by NVP segments could also partly destabilize the collapsed structure.

    Notes: The reactivity of the mono-, di- and trimethyl-ammonium nitrates to heat has been studied using differential thermal analysis, thermogravimetry and mass spectral analysis. The decomposition temperature of these compounds increases in the order CH3NH3NO3 〉 (CH3)2NH2NO3 〉 (CH3)3NHNO3. The activation energies have been determined employing isothermal decomposition technique. Mass spectrometric investigation shows that the extent of decomposition increases with increasing substitution on the nitrogen atom. Possible reasons for such behaviour have been explored.
