    Notes: Chymotrypsin has been immobilized to several nonporous magnetic materials. Nickel particles were considered to be most suitable as immobilized enzyme supports. Chymotrypsin immobilized to nonporous magnetic supports was not fouled significantly by6 either whole milk or clarified yeast homogenate. AE-cellulose-chymotrypsin was rapidly fouled by both these materials and chymotrypsin immobilized to acrylic-based ion exchangers was slowly fouled. Immobilized enzyme activity was found to be inverted proportional to particle diameter for nonporous rock magnetite particles. Immobilization by adsorption and then glutaraldehyde crosslinking was used to produce controlled amounts of chymotrypsin on the particles. Esterolytic activity increased with enzyme loading but caseinolytic activity did not increase. Chymotrypsin is inhibited by metal ions from the magnetic supports. It is partially protected by use of a preliminary protein coating and may be reactivated by incubation with EDTA or BSA.

    Notes: Biological kinetic behaviors of the oxygenated and aerated activated sludge process were studied and compared in both once-through and constant sludge recycle systems. The models derived by Herbert, Elsworth, and Telling [J. Gen. Microbiol., 14, 601 (1956)] and Ramanathan and Gaudy [Biotechnol. Bioeng., 11, 207 (1969)] were used for the studies of once-through and constant sludge recycle systems, respectively. Soft drink waste water was used for the growth limiting substrate. Temperature was controlled within 30 ± 2°C. The influent substrate concentration was maintained at 1,000 mg/liter. The experiments were conducted at various dilution rates (from \documentclass{article}\pagestyle{empty}\begin{document}$ \frac{1}{9} $\end{document} to 1/1.0 hr-1), and recycle solids concentration values (from 5,000 to 10,000 mg/liter), with hydraulic recycle ratio, α, at 0.3.Biological kinetic constants were evaluated and compared. It was found that these constants were different for the aerated and oxygenated systems within a certain range of dilution rates studied. The critical dilution rates for diluting out effluent chemical oxygen demand (COD) occurred at 0.1 and 0.2 hr-1in the once-through operation, and 0.2 and 0.4 hr-1in the sludge recycle operation for aerated and oxygenated systems, respectively. Observed sludge yield values and specific growth rate were varied with the type of aeration and with and without constant sludge recycle concentration applied. Sludge carbohydrates and proteins content in the oxygenation system (cell recycle) were 10.1-21.6% and 35.6-52.2%. Sludge volume index in the air and oxygenation systems varied from 41.4 to 354 and 31.9 to 58.5, respectively.

From the present study it may be specifically found that trypsin and pappain can be immobilized by molecular sieve type 4A following a very simple technique.     Notes: Entomophthora virulenta has been grown using inexpensive, commercial grade nutrient sources in order to produce a resting spore formulation available to agriculture. Solid media and liquid media in shake flasks and fermentors were examined. The best carbon sources for sporulation were dextrose and corn syrup, while the best nitrogen sources were yeast extract, soybean, and cottonseed flours. The optimal nutrient concentration (12%) produced 3 × 106spores/ml with a 70% germination rate within five days.

    Notes: The reliability of the process of Ramanathan and Gaudy (Biotechnol Bioeng., 13, 125 (1971)) for the completely mixed activated-sludge process holding the recycle cell concentration, XR, as a system constant with respect to step changes in hydraulic retention time was investigated. The experiments were run at initial dilution rates of ⅛, ⅙, ¼, and ½ hr-1 treating a soft drink bottling wastewater. The influent substrate concentration was maintained at 1000 mg/liter chemical oxygen demand and the hydraulic recycle ratio at 0.3. The recycle sludge concentration was maintained at about 7000 mg/liter.It was found that the system could accommodate hydraulic shock loads up to 200% positive changes and down to 50%negative changes without disruption of the effluent quality. Shorter retention time of the range studied, from 2 to 8 hr, has the advantage of shorter response time with respect to the response of the concentration of biological solids in the reactor.

    Notes: A new approach to preparative organic synthesis in aqueous-organic systems is suggested. It is based on the idea that the enzymatic process is carried out in a biphasic system “water-water-immiscible organic solvent.” Thereby the enzyme is localized in the aqueous phase - this eliminates the traditional problem of stabilizing the enzyme against inactivation by a nonaqueous solvent. Hence, in contrast to the commonly used combinations “water-water-miscible organic solvent,” in the suggested system the content of water may be infinitely low. This allows one to dramatically shift the equilibrium of the reactions forming water as a reaction product (synthesis of esters and amides, polymerization of amino acids, sugars and nucleotides, dehydration reactions, etc.) toward the products. The fact that the system consists of two phases provides another very important source for an equilibrium shift, i.e., free energies of the transfer of a reagent from one phase to the other. Equations are derived describing the dependence of the equilibrium constant in a biphasic system on the ratio of the volumes of the aqueous and nonaqueous phases and the partition coefficients of the reagents between the phases. The approach has been experimentally verified with the synthesis of N-acetyl-L-tryptophan ethyl ester from the respective alcohol and acid. Porous glass was impregnated with aqueous buffer solution of chymotrypsin and suspended in chloroform containing N-acetyl-L-tryptophan and ethanol. In water (no organic phase) the yield of the ester is about 0.01%, whereas in this biphasic system it is practically 100%. The idea is applicable to a great number of preparative enzymatic reactions.
