    Notes: A new method to estimate the oxygen transfer coefficient (KLa) from the experimental dynamic response data is presented. Employing a linear model which allows for gas phase, diffusion film, and oxygen electrode dynamics, the first moment of the response curve is simply related to the sum of the model parameters. Two separate experiments are used to characterize the measurement dynamics and to measure the unknown KLa parameter. The simple calculation procedure involves only measuring the area above the response curves.

    Notes: Mouse teratocarcinoma cells derived from embryoid bodies of 129SVsl mice were cultured in vitro to permit their differentiation. These cells were then infected with simiam virus 40 (SV40) and 31 cloned cell lines (SVTER) were derived from these cultures. All 31 SVTER cell lines contained the SV40 tumor (T) antigen and grew as permanent lines in culture. Mock-infected embryoid body cultures did not give rise to permanent cell lines. The morphology of each SVTER cell line was distinct and did not change during successive subclonings.The growth properties and tumorigenic potential of all 31 SVTER cell lines were investigated. None of these lines produced tumors in 129SVsl mice. Each cell line was tested for its ability to (1) grow in medium containing 1% serum, (2) plate on a cell monolayer, and (3) form clones in methocel suspension. Only three of the SVTER cell lines were transformed with respect to all three of these criteria. Most of these cell lines were minimal transformation.The SVTER cell lines were tested for creatine phosphokinase (CPK), an enzyme activity characteristic of mouse brain and muscle tissue, and the protease, plasminogen activator (PA) which is found in embryoid bodies and several differentiated cell types. Some of the SVTER cell lines contained high levels of CPK, while others had high levels of PA and a third group of cells contained neither enzyme activity. No SVTER cell line was found with high levels of both these enzyme activities. This result suggests that mutually exclusive sets of genes are expressed in these cells as might be expected from the distinct tissue distribution of the two enzyme activities studied. These SVTER cell lines may be useful in reconstructing developmental pathways of differentiating teratomas in vitro.

    Notes: The mechanical properties of the whole muscle and fast-twitch muscle units of the cat hindlimb pretibial flexors have been explored and related to normal locomotion. Tibialis anterior (TA) is parallel-fibered and functionally crosses a single joint, the ankle, whereas extensor digitorum longus (EDL) is pinnate and spans the ankle, knee, metatarsophalangeal and interphalangeal joints. The active tetanic tension of TA remains near its peak value over a range of muscle lengths associated with normal ankle movement. In contrast, the length-tension curve of EDL is sharply peaked. However, normal corollary action of the knee, ankle and metatarsophalangeal joints during stepping minimizes EDL's excursion and maintains it at or near a length optimal for peak tension development. EDL is capable of producing synchronous but sterotyped digit and ankle movements while TA provides for independent ankle flexion at all relevant joint angles.The mechanical properties of 84 TA and 98 EDL fast-twitch muscle units were studied by measuring twitch contraction time (≤45 msec), peak tetanic tension, response to repetitive stimulation, and contractile fatigue resistance during electrical stimulation of single alpha axons, functionally isolated from ventral root filaments. These mechanical properties were essentially similar for both muscles with the exception of mean peak tetanic tension which was 30% lower for TA units (14 gm-wt) than for EDL units (20 gm-wt). A high proportion of units in both muscles demonstrated fatigue resistance which is reflective of the repetitive, phasic demand upon these muscles during locomotion.

    Notes: A survey is made of some ultrastructural features of the developing cornea of Macaca mulatta. The observations are confined to the anterior central area, starting with the lens vesicle stage and progressing through midgestation, when the morphologic characteristics of the cornea are fully established. Subepithelial filaments and some partially aggregated collagen fibrils are present in the earliest embryo and are of a size and appearance similar to those in the future vitreous cavity. Epithelial secretory activity points to, but does not prove direct contribution to the deposition of the acellular matrix components beneath it. No trace of a structured, orthogonal collagenous stroma can be visualized. The primitive endothelium forms prior to the fibroblast invasion of the distended filamentous matrix. Bowman's layer has undoubted epithelial contributions. Its aggregated collagen fibrils have approximately the same diameter as those of the anterior stroma. Intraepithelial appearance of single nerve fibers and fascicles takes place during the first trimester of gestation, as soon as the two continuous epithelial layers are formed. Terminal areas approach closely to the basal cell's nucleus, without touching it. The plasmalemma of the invaginating nerve fiber is surrounded by that of the epithelial cell in a mesaxon-like manner, with occasional gap junctions uniting adjoining neural and epithelial cell membranes. The fetal neurites contain microtubules, some clear vesicles and dense vacuoles resembling those of mature monamine and non-monamine neurons. Mitochondria are small and compact, their presence indicating a high rate of metabolic activity in the immature terminal area.

    Notes: The origin, distribution and structure of the blood vessels of the female reproductive tract and the testis of the brush possum (Trichosurus vulpecula) were studied using latex and silicone rubber casting and histological techniques. Latex casts of the vessels of the female tract were also studied in five macropod species - Macropus giganteus, M. eugenii, M. agilis, Megaleia rufa and Thylogale billardierii, and in the common wombat (Vombatus ursinus).The female reproductive tract in the brush possum was supplied and drained by four major sets of paired vessels - ovarian, cranial urogenital, caudal urogenital, and internal pudendal arteries and veins. These vessels formed substantial anastomoses with one another on each side of the midline, and also across-the-midline anastomoses. The proximal part of the ovarian artery ran in close apposition to the ovarian vein, which received one or more large uterine branches. In its distal protion the ovarian artery gave rise to a leash of small, tortuous ovarian branches, which wound around and between the plexiform ovarian veins.The testicular arteries and veins in this species also ran in close apposition to one another. Both arteries and veins branched into many smaller, mildly tortuous, parallel vessels in the spermatic cord, which reunited before entering the testis.The blood vessels of the reproductive tract in all of the macropod species studied, and in the common wombat, were basically similar to those of the brush possum.The intimate structural relationships between ovarian arteries and veins, and their ovarian branches, in these marsupials are suggestive of specializations for counter-current exchange between venous and arterial blood. However, in contrast to those of the testicular vessels where heat exchange is a demonstrated function, their physiological significance remains unknown.

    Notes: Continuous culture studies have been carried out growing Trichoderma viride QM 9123 in a 10 liter stirred fermentor on a medium containing commercial glucose as the carbon source. Experiments were carried out at 30°C and at three controlled pH values of 2.5, 3.0, and 4.0, over a range of dilution rates from 0.01 to 0.11 hr-1. Steady-state values of cell, glucose, and cellulase concentration oxygen tension, and outlet gas oxygen partial pressure were recorded. Values of maximum specific growth rate, endogenous metabolism coefficient, Michaelis-Menten coefficient, yield and maintenance coefficient for glucose were derived and correlated the effect of the hydrogen ion concentration. Specific oxygen uptake rates were correlated with specific growth rates and absorption coefficients were shown to be a function of dilution rate independent of pH. Some data on cellulase biosynthesis were examined and correlated in terms of a maturation time model.
