Notes: The destruction of various UV stabilizers in polypropylene films during irradiation has been examined both in the presence and absence of air and oxidation products. Although some stabilizers were destroyed in the absence of oxygen, all were more sensitive when irradiated in the presence of air. Several stabilizers were highly effective but were destroyed so rapidly that photoprotection of the polymer must have resulted from stabilizer products. Work with preoxidized polymer and with model hydroperoxides and peroxides indicated that most stabilizers were destroyed by the attack of oxygen-centered radicals, with peroxy radical attack appearing to dominate.

Description: Continuum observations of 23 extragalactic objects have been made at a wavelength of 1 mm; nine of the 23 have been definitely detected at this wavelength. The sources detected include seven from which the 1-mm emission appears to be nonthermal, and two from which the emission appears to be thermal radiation from dust. Repeated observations of the brightest nonthermal sources permitted a search for 1-mm variations on time scales of one month to three years. BL Lac was observed to vary by more than a factor of 2 in flux, and 3C 84, 3C 120, and 3C 273 showed possible variations; 3C 279 showed no variations greater than + or - 30%. The 1-mm variations of BL Lac appear to be correlated with variations at radio wavelengths. The other sources with nonthermal spectra detected were 3C 111 and M87; the two thermal sources detected were NGC 253 and M82. Comparison of the latter two 1-mm measurements with 2.6-mm CO measurements suggests that the CO line in these galaxies is not heavily saturated.

Notes: Earlier studies by the authors showed that the collagen fibrils in rat-tail tendon have a bi-modal distribution of fibril diameters from a time shortly after birth through to the onset of maturity at about 3-4 months. Present work has extended those observations for rats up to the age of 2 years. Histograms of the fibril diameter distributions for mature tail tendon and direct electron microscope observations show that the fibrils break down as the tendon ages. Further work on the constant diameter subfibrils of diameter 140 Å described previously, has confirmed that these are part of the elastic fibers present in tendon at all ages. It has been shown that there is relatively little variation in the collagen fibril diameter distribution as a function of the position of the specimen in the tail, and as the measured percentage of the area taken by the collagen fibrils present at any particular point. Estimation of the fibrillar collagen content of rat-tail tendon as a function of age indicates that it increases steadily from birth and reaches a maximum at the onset of maturity, beyond which the fibrillar collagen content appears to remain constant.

Notes: 1H and 13C n.m.r. Spectroscopical Measurements and HMO-π-Calculations to the π-Electronic Structure of p,p′-Disubstituted trans-Stilbenes and Substituted trans-β-Styryl-methyl-SulfonesThe 1H and 13C n.m.r. spectra have been measured for a series of donator-acceptor-disubstituted trans-stilbenes 1 and monosubstituted β-styryl-methyl-sulfones 2. The 13C n.m.r.-signals have been identified by comparison with empirical calculated chemical shifts and quantum chemical determined HMO-π-electrons densities. The influence of substitutents to the experimental chemical shifts are discussed in detail. For 1 and 2 exist linear correlations between chemical shifts and HAMMETT values, and π-electron densities, respectively.

Notes: Cellulose II and cellulose I samples were treated respectively with a strong solution of sodium deuteroxide in D2O and a 99% solution of ethylenediamine. The complexes were washed with D2O and the nature of the OD bands were studied after rehydrogenating the sample. The infrared spectra of these rehydrogenated samples showed well-resolved OD bands which are resistant to reexchange. The Characteristic bands are discussed and compared with the predicted OH-OD frequency shifts.

Notes: Urease from Jack bean was immobilized on nonporous glass beads by covalent bonding and its kinetics were studied in a packed-column differential reactor. To facilitate comparison, the urease was immobilized by both diazo and glutaraldehyde coupling. The kinetic properties of immobilized urease were similar to those of the soluble enzyme and different immobilization methods did not appreciably alter the kinetic properties. The affects of three different amino acid activators appear to follow predictions obtained from a relatively simple competitive model, except at very low substrate levels.

Notes: Glucose oxidase from Aspergillus niger was immobilized on nonporous glass beads by covalent bonding and its kinetics were studied in a packed-column recycle reactor. The optimum pH of the immobilized enzyme was the same as that of soluble enzyme; however, immobilized glucose oxidase showed a sharper pH-activity profile than that of the soluble enzyme. The kinetic behavior of immobilized glucose oxidase at optimum pH and 25°C was similar to that of the soluble enzyme, but the immobilized material showed increased temperature sensitivity. Immobilized glucose oxidase showed no loss in activity on storage at 4°C for nearly ten weeks. On continuous use for 60 hr, the immobilized enzyme showed about a 40% loss in activity but no change in the kinetic constant.

Notes: A 1000 liter fermentor has been used to produce a continuous feed of Escherichia coli containing a high level of β-galactosidase. We have investigated the individual unit operations for the isolation of the enzyme: cell disruption, nucleic acid removal, protein precipitation, and solid-liquid separation after each stage. Using the information obtained we have been able to operate a semicontinuous process which when fully continuous would yield 100 g protein/hr, comprising 23% β-galactosidase.
