Beschreibung: Phase 2 (development) which was concluded for the ultrasonic Doppler device and the light-pipe-reflectance device is reported. An ultrasonic Doppler breadboard system was assembled which accurately measured runout in the J-2 LOX pump impeller during operation. The transducer was mounted on the outside of the pump volute using a C-clamp. Vibration was measured by conducting the ultrasonic wave through the volute housing and through the fluid in the volute to the impeller surface. The impeller vibration was also measured accurately using the light-pipe probe mounted in an elastomeric-gland fitting in the pump case. A special epoxy resin developed for cryogenic applications was forced into the end of the fiber-optic probe to retain the fibers. Subsequently, the probe suffered no damage after simultaneous exposure to 2150 psi and 77 F. Preliminary flash X-radiographs were taken of the turbine wheel and the shaft-bearing-seal assembly, using a 2-megavolt X-ray unit. Reasonable resolution and contrast was obtained. A fast-neutron detector was fabricated and sensitivity was measured. The results demonstrated that the technique is feasible for integrated-time measurements requiring, perhaps, 240 revolutions to obtain sufficient exposure at 35,000 rpm. The experimental verification plans are included.

Beschreibung: The problems of developing large-area, gas-scintillation proportional counters with high resolution are considered. It is found that simple large-area, parallel-grid proportional counters suffer from a variation in gain over the counter window. Some success has been achieved in overcoming this problem by focusing the charge cloud as it drifts into the multiplication region. Measurements are reported for various mixtures of argon and xenon as well as pure xenon.

Notizen: The relative proliferative capacity of haematopoietic cell populations derived from 22-week-old adult bone marrow and 14-18 day foetal liver has been studied in lethally irradiated syngeneic recipients by means of chromosome markers. Although starting at a disadvantage in terms of the number of colony-forming units (stem cells) injected, the foetal liver-derived populations steadily increased their relative numbers in the myeloid and lymphoid tissues over a period of several weeks until a plateau was reached. It is suggested that stem cells in foetal liver have, on average, a higher intrinsic capacity for self-renewal than do those in bone marrow, and that this capacity falls to the adult level within about ten weeks of transfer.

Notizen: Lymphopoiesis with respect to recirculating and non-recirculating small lymphocytes was measured simultaneously in rats thymectomized as adults. Removal of the thymus at four to five weeks of age had a profound inhibitory effect upon the production of recirculating cells, whereas the formation of non-recirculating lymphocytes was only slightly depressed. Thymectomy had approximately the same impact of lymphopoiesis as thymectomy and exposure of the animal to a large dose of whole body X- and γ-irradiation. The latter finding, and the failure of a thoracic duct cell transfusion to augment lymphocyte production, accord with the view that the thymus is the principle intermediate source of recirculating small lymphocytes in the normal, unstimulated animal.

Notizen: A method is described for measuring lymphopoiesis that enables the production of recirculating and non-recirculating small lymphocytes to be estimated simultaneously. Using this technique, experiments were undertaken to determine whether the production of recirculating cells is influenced by the number present in the recirculating lymphocyte pool. The results suggest that neither a massive lymphocyte transfusion nor depletion of the pool by whole body irradiation or chronic lymph drainage affect the rate at which recirculating small lymphocytes are generated.

Notizen: The calcitonin (SCT) from salmon ultimobranchial bodies which (like mammalian calcitonins) lowers the plasma calcium concentration in mammals can also affect cyclic AMP (cyclic adenosine 3′,5′-monophosphate) metabolism and proliferation of lymphoblasts in normal and prostaglandin E1 (PGE1)-treated rat thymocyte populations in three different ways. In the first case, low concentrations (0.5-5.0 ng per milliliter) of SCT lower (by a calcium-mediated process) the ability of PGE1 to transiently increase cyclic AMP synthesis, but the reduced surge of cyclic AMP production is still ample to stimulate lymphoblasts in the cell population to initiate deoxyribonucleic acid (DNA) synthesis. Secondly, these low SCT concentrations affect the eventual progression of the PGE1-stimulated, DNA-synthesizing lymphoblasts into mitosis by a calcium-mediated process. Depending on the extracellular calcium concentration and the magnitude of the initial increment in the intracellular cyclic AMP content, SCT can either promote or inhibit the progression of the stimulated cells into mitosis. SCT's third action is a rapid (within 5 minutes), calcium-independent elevation of the cellular cyclic AMP content in otherwise untreated thymic lymphocyte populations exposed to a very high concentration (100 ng per milliliter) of the hormone. This early, transient rise in the cyclic AMP level is followed by a calcium-dependent increase in lymphoblast proliferation. An attempt is made to interrelate and explain the different actions of SCT on cyclic AMP metabolism and mitogenesis.

Notizen: Mouse teratocarcinoma cells derived from embryoid bodies of 129SVsl mice were cultured in vitro to permit their differentiation. These cells were then infected with simiam virus 40 (SV40) and 31 cloned cell lines (SVTER) were derived from these cultures. All 31 SVTER cell lines contained the SV40 tumor (T) antigen and grew as permanent lines in culture. Mock-infected embryoid body cultures did not give rise to permanent cell lines. The morphology of each SVTER cell line was distinct and did not change during successive subclonings. The growth properties and tumorigenic potential of all 31 SVTER cell lines were investigated. None of these lines produced tumors in 129SVsl mice. Each cell line was tested for its ability to (1) grow in medium containing 1% serum, (2) plate on a cell monolayer, and (3) form clones in methocel suspension. Only three of the SVTER cell lines were transformed with respect to all three of these criteria. Most of these cell lines were minimal transformation. The SVTER cell lines were tested for creatine phosphokinase (CPK), an enzyme activity characteristic of mouse brain and muscle tissue, and the protease, plasminogen activator (PA) which is found in embryoid bodies and several differentiated cell types. Some of the SVTER cell lines contained high levels of CPK, while others had high levels of PA and a third group of cells contained neither enzyme activity. No SVTER cell line was found with high levels of both these enzyme activities. This result suggests that mutually exclusive sets of genes are expressed in these cells as might be expected from the distinct tissue distribution of the two enzyme activities studied. These SVTER cell lines may be useful in reconstructing developmental pathways of differentiating teratomas in vitro.

Beschreibung: The first photographs of the moon taken with earthshine illumination by the Apollo 15 crew were examined. Emphasis in the analysis was placed on a comparison of predicted exposures and obtained exposures. The results of the investigation indicate the following: (1) The predicted exposure of the maria areas and the obtained exposures were in reasonably close agreement. (2) The apparent-albedo values obtained for the floor of Aristarchus Crater are seven times greater than those for the maria.

Beschreibung: The advantages resulting from the use of near-terminator photography in lunar surface investigations are discussed. It is pointed out that, under near-terminator conditions, small changes in slope produce greater contrast changes than at high sun elevation angles. This desirable phenomenon is confirmed by an examination of the near-terminator photography taken during the Apollo 15 mission. Many of the photographs obtained show lunar surface areas within a few degrees of the terminator and are therefore of significant geologic interest. In addition, many geologic features stand out in a distinct manner not normal in conventional lunar photography, thus providing additional data on the surface morphology and the configuration of a large number of lunar surface structures.

Beschreibung: The pressure-modulated carbon dioxide radiometer is a new kind of instrument capable of making temperature soundings in the 40-80-km region of the earth's atmosphere. It is intended to be mounted on a polar-orbiting satellite, where it will give global coverage of the upper atmosphere in a region that is not well understood at present but that is, as rocket soundings show, clearly the seat of many interesting and vigorous phenomena. The new technique employs a cell containing carbon dioxide as a filter. The pressure and hence transmission of this cell is periodically modulated, resulting in the selection of thermal radiation from the strong lines in the spectrum of atmospheric carbon dioxide. This radiation originates at levels in the atmosphere where the pressure is low. The energy grasp of the device is large enough to give high sensitivity.
