    Notes: Acronycine  -  an alkaloid with antineoplastic activity against a wide range of experimental tumors  -  at concentrations of 0.5-12 μg/ml rapidly inhibits RNA synthesis in L5178Y mouse lymphoma and IRC rat monocytic leukemia cultures. Culture growth is arrested only at acronycine concentrations which markedly inhibit RNA synthesis. DNA synthesis is inhibited at rather higher concentrations but this is not a prerequisite of the arrest of growth. It is suggested that the arrest of growth may be a consequence of the inhibition of RNA synthesis.In both cultures arrest of growth coincides with the appearance of many cells with two apparently normal nuclei. Cells are not arrested in mitosis. It is shown these binucleated cells very probably arise from an inhibition of cell cleavage. Studies with synchronized cultures show that at low drug concentrations, more than one cell cycle may elapse before growth is arrested and binucleated cells appear, indicating the effect on cytokinesis is not immediate. The results suggest that the arrest of growth may be a result of a slow depletion of a component essential for cell cleavage. The disturbance at division is a major factor in arresting growth at low drug concentrations. At higher acronycine concentrations, when RNA synthesis may be inhibited by 80-90%, the cytotoxic effects appear earlier and are less specifically directed at cytokinesis; DNA synthesis is then also rapidly and markedly inhibited.

    Notes: The epidermal covering of the tail scales of the gekkonid lizard Lygodactylus bears three distinct types of specialization: sense organs, pilose pads whose function is either sensory or scansorial or both, and holocrine secretory organs (β-glands) which are only found in males. The same specializations are found on regenerated tails, and although the morphological form and patterning of the scales do not resemble the original, the structure, distribution and sexual specificity of the specialized units is perfectly restored. These structures, and similar units in other lacertilian genera have certain resemblances to mammalian and avian epidermal specializations. Perfection of replacement of integumentary specializations can be correlated with functional demands, although the developmental mechanism underlying the phenomenon is unknown.

    Notes: The fine structural morphology of the male squirrel monkey adrenal cortex has been examined. When compared to other laboratory animals, the squirrel monkey adrenal cortex secretes large amounts of cortisol and maintains extraordinarily high plasma cortisol levels for prolonged periods of time. The normal cortical cells have numerous mitochondria with either a tubulo-vesicular or lamellar internal membrane arrangement, a well-developed agranular endoplasmic reticulum which is arranged in juxtaposition to mitochondria and lipid droplets, several lysosomes, and numerous thin-walled blood vessels of large caliber, suggestive of a rich blood flow through the gland. These characteristics have heretofore been associated with hypersecretion. Their presence in the squirrel monkey cortex, known to have high secretory activity, lends credence to the correlation of hyperdevelopment of the agranular reticulum with increased rates of secretion of corticoids.During chair restraint, the plasma cortisol levels rise two to three fold. Adrenocortical cells thus stressed exhibit a depletion and disorientation of membranes both of the agranular endoplasmic reticulum and mitochondria and a loss of ribosomes, lysosomes and, to some degree, intracellular lipid. The animal appears to be responding maximally to the stress of chair restraint. These fine structural characteristics are interpreted as an example of an adrenal cortex in the process of becoming functionally exhausted, since these animals sometimes do not survive the stress of chair restraint.

    Notes: Electron microscopic observations on the mechanically undisturbed guinea pig bone marrow show that the sinusoidal lining is continuous. There are neither intercellular nor intracellular apertures allowing free communication between the extravascular and intravascular compartments. A transient migration pore is only formed during the diapedetic transit of blood cells. Serial sections show that this aperture is transcellular. A functional continuity of the sinusoidal lining appears to be maintained during the diapedesis of blood cells, which is evident from the absence of a significant extravascular leakage of plasma during this process.

    Notes: Mice (C57Bl) were placed for 35 days in a room with reversed light cycle; lights came on at 1800 hours and off at 0600 hours. At six hour intervals throughout the day three mice were injected with tritiated thymidine and sacrificed 30 minutes later. Crypts were dissected for radioautography and determination of disintegrations per minute per crypt, the number of cells in mitosis and the number of cells synthesizing DNA was determined. The results reported clearly demonstrate reversal of the circadian rhythm in the parameters tested.

    Notes: Establishment of monolayer cell-cultures from adult normal rat liver is described using cells isolated by a collagenase-hyaluronidase perfusion technique. The cells plated originally were polygonal in shape and contained a large number of mitochondria. However, cells with less granules were also observed in the primary culture, after a few days of plating. In subsequent subcultures, the latter type of cells could be grown continuously. After an initial lag, the cells attained a doubling time of 55 hours and a plating efficiency of 20%. The cells are “normal” with respect to their karyology, morphology and growth pattern. Difficulties encountered during the culture and the possible uses of these cultured cells in studies on carcinogenesis in vitro are discussed.

    Notes: The rate of protein synthesis per cell in cultured hamster embryo fibroblasts in the stationary growth phase falls to about one third of the rate in the exponential growth phase. This reduction can be entirely accounted for by the following observations: (1) the average cell in stationary phase contains about one-half the number of ribosomes per cell compared to the average cell in exponential phase; (2) only two thirds of the ribosomes are bound to polysomes in stationary phase, while nearly all of the ribosomes are polysome-bound in exponential phase.In stationary phase, ribosomes which are polysome-bound function with the same efficiency and produce proteins of approximately the same average length as in exponential phase. Experimental findings are presented which suggest that the generation of a higher proportion of free ribosomes in stationary phase in not due to a limitation in messenger RNA, but to a decreased attachment probability of ribosomes to messenger RNA.

    Notes: Intracellular microelectrode studies of passive membrane properties and action potential generation were carried out on cloned and uncloned mouse neuroblastoma cells in tissue culture. The cloned cells were studied between the eighth and tenth months and the uncloned cells between the third and fifth weeks after primary dissociation. Electrophysiologic measurements of cell membrane properties were made by passing stimulating current pulses across the cell membrane from an intracellular microelectrode and recording simultaneously from the same electrode, by means of a bridge circuit, the changes in membrane potential. The range of responses to electrical stimulation varied from passive increases in membrane potential to repetitive firing of action potentials. A 20 fold range in spike generating capability was found. Passive membrane properties (membrane potential, specific membrane resistivity, and specific membrane capacitance) were similar to those of sympathetic neurons in intact preparations. Seventy-nine percent of the cloned cell line compared to 94% of the uncloned line were capable of generating action potentials. Less than 2% of the cloned cells showed repetitive firing whereas 23% of the uncloned cells had this property. As in several types of normal neurons, the action potential mechanism was largely, although not completely, blocked by iontophoretic and bath applied tetrodotoxin.

    Notes: Using a new, sensitive and quantitative technique for determining the ribosomal-RNA content of a measured number of cells, the cellular ribosome complement was compared for cultured hamster embryo cells in the stationary growth phase and in the early G1 phase of the cell cycle. Cells from stationary phase cultures were found to contain less than 70% of the ribosome complement of the early G1 phase cells, though the volumes of the two cell types were similar. This would imply that the stationary phase cell is physiologically different from a cell merely arrested at some point in the cell cycle.

    Notes: Cultures of BHK2l cells continuously release into serum-free medium, active materials which stimulate the incorporation of thymidine in non-confluent, but not in confluent cultures of homotypic cells. The activity is not removed by centrifugation at 30,000 g for four hours, but is non-dialysable and retained by membranes with a nominal limitation for MW 25,000. Activity is stable at 4° for several weeks, but destroyed in 30 minutes at 56°. BHK2l cells also release active material capable of enhancing the effect of added serum. This is also heat labile and its action is density-dependent.
