Notes: Summary In the measurement of arsenic by the method ofVašák andŠedivec, different wavelengths have been used to measure the concentration of the arsenic diethyldithiocarbamate. Such differences may be explained by the possible presence of silver nitrate in the silver salt solution. The absorbance should be measured at 522 nm when the salt is properly purified. The authors also measured the interference of antimony on the arsenic determination in the concentration range of 50–450μg.

Description / Table of Contents: Zusammenfassung Ein Verfahren zur Bestimmung von Schwefelsäure als Verunreinigung der Atmosphäre wurde beschrieben. Die Abtrennung von der Luftprobe erfolgt durch Mikrodiffusion bei 200° C. Die Schwefelsäure wird in Natronlauge absorbiert und mit Bariumperchlorat titriert.
Notes: Summary A new method for the measurement of sulphuric acid in airborne particulates is described. Sulphuric acid present is separated from the sample matrix by microdiffusion at 200°. The separated sulphuric acid is absorbed in sodium hydroxide, and titrated by barium perchlorate.

Notes: Summary A method is described for the detection and spectral examination of traces of porphyrin complexes by controlled demetallation and spectrofluorometry. By carrying out the demetallation with methanesulfonic acid in the optical cell, difficulties connected with handling losses and introduction of contamination are minimized. The method is sensitive to quantities as low as 0.05 ng.

Notes: Summary The flux of water, $$f_{{\text{H}}_{\text{2}} {\text{O}}}$$ , to the xylem of exuding root systems of Ricinus communis was controlled using a range of mannitol concentrations permitting the influence of this water flux on the potassium flux, f K, to be studied. The relationship between $$f_{{\text{H}}_{\text{2}} {\text{O}}}$$ and f K thus obtained was investigated, for a number of external concentrations of potassium, Cm, supplied as potassium nitrate. An analysis of these data indicated the presence of a water dependent and a water independent f K both of which varied with Cm. The water dependent f K shows a parabolic relationship with Cm for Cm values 〈1 mM followed by a sharp inflection and decline at higher Cm values whereas the water independent f K shows an hyperbolic relationship over the same range of Cm values. Uptake of potassium by exuding root systems was measured and shown to be dependent on the solute potential of the medium. The uptake was also shown to exhibit a dual absorption isotherm the kinetics of which indicate a low Km system (system 1) and a high Km system (system 2). The Km value obtained for system 1 is very similar to that obtained for the water independent f K. It is postulated that the water independent f K is contributed by that portion of f K arriving in the stele via the cortical symplast and is directly dependent on Cm. The water dependent f K is contributed by those ions moved across the root in response to centripetal water movement through the cortical cell walls.

Notes: Summary A technique for maintaining mammalian ovaries in organ culture is described. Ovaries of rats, mice, monkeys and humans have been maintained in a histologically normal condition for up to 20 days in culture, during which many were X-irradiated. The doses of X-rays required to destroy all the primordial oocytes in organ cultures are similar to those reported forin vivo studies. The human foetal ovary is resistant to X-irradiation. Some oocytes survive for 7 days following exposure to 4000 R in organ culture. The radiation sensitivity of the human ovary is discussed: it would appear that it is amongst the most resistant to oocyte-killing known.

Notes: Abstract The NADP-dependent malate dehydrogenase (MDH) in the supernatant fraction of mouse tissues is known to occur in two allelic forms which are electrophoretically distinguishable; each produces a single band in starch gel. We have investigated the subunit structure and synthesis of NADP—MDH through electrophoretic patterns obtained from several experimental sources. (1) Heterozygotes containing both alleles yield a five-banded pattern. The bands are in an approximate frequency of 1:4:6:4:1; the two extremes correspond to the pure types and the three intermediates are presumably hybrid enzymes. The NADP—MDH molecule therefore appears to be a tetramer. (2) In muscle heterokaryons of allophenic mice (with homozygous nuclei of each genotype within a common cytoplasm), hybrid enzymes are formed; they are not formed in other allophenic tissues. Therefore the gene at this locus codes only for monomeric subunits and the tetramer is assembled in a second step in the cytoplasm. Also, both genes must function in a nucleus when the locus is active (e.g., in F1 uninucleated cells). (3) Dissociation in vitro of mixtures of both pure types of enzymes, followed by reassociation among fragments, leads to a three-banded pattern, even after repeated cycles. Thus the tetramer must cleave in a fixed plane, to form dimers, which reassociate, rather than in a random fashion to form monomers. The most likely interpretation is that mouse NADP—MDH is an example of the type of tetramer postulated by Monod et al. (1965) and termed “isologous.” The dimers are held symmetrically in the tetrameric conformation by relatively weak forces; the monomeric subunits comprising the dimer are held together by stronger forces.
