    Notes: The effects of the male antifertility agent, α-chlorohydrin, six of its derivatives, and glycidol were studied on the metabolism of washed ram spermatozoa in vitro with fructose as substrate. The α-chlorohydrin derivatives were the amino, the phosphorylated, and four glycol-bridge (ketal) compounds. All compounds except glycidol, in a concentration between 0.1 and 100 mM, reduced the aerobic glycolsis and/or oxidation of fructose. However, there was not a high correlation between the ability of these compounds to inhibit the metabolism of ram spermatozoa in vitro and their antifertility activity when administered to male rats. Other factors are clearly involved in their antifertility activity, eg, the concentration of the compounds in the epididymis and their conversion of either more or less spermicidal compounds in the body.

    Description: Measurements of forbidden line Ar II 6.99 micron and Pf alpha 7.45 micron made from the Kuiper Airborne Observatory are presented for Sgr A, the H II region at the center of the Galaxy. These line strengths, when combined with ground-based measurements, suggest a factor of enhancement of two in the Ar/H ratio in the galactic center region relative to that in the sun and in the solar neighborhood. The accuracy of the determination is presently limited by the uncertainty in the collision strength for Ar(+).

    Description: The J = 6 - 5 rotational transition of carbon monoxide has been detected in emission from the KL 'plateau source' in the Orion molecular cloud. The corrected peak antenna temperature is 100 K, and the FWHM line width is 26 km/sec. These observations were carried out using the 3 m telescope of the NASA IRTF (Infrared Telescope Facility) on Mauna Kea, Hawaii, and constitute the first astronomical data obtained at submillimeter wavelengths with a heterodyne system using a laser local oscillator. The data support the idea that the high-velocity dispersion CO in Orion is optically thin and set a lower limit to its temperature of approximately 180 K.

    Description: During the three months of orbital operations, the satellite returned data from the world's oceans. Dozens of tropical storms, hurricanes and typhoons were observed, and two planned major intensive surface truth experiments were conducted. The utility of the Seasat-A microwave sensors as oceanographic tools was determined. Sensor and geophysical evaluations are discussed, including surface observations, and evaluation summaries of an altimeter, a scatterometer, a scanning multichannel microwave radiometer, a synthetic aperture radar, and a visible and infrared radiometer.

    Description: The magnetic disturbance fields caused by global external current systems are considered with particular emphasis on improving the understanding of the physical processes which control high latitude current systems. Following processing the MAGSAT data were routinely plotted in the Universal Time (UT) format as well as in a polar plot format. The H'D'U' coordinate system, was adopted as the standard for representing the MAGSAT residual magnetic field vectors. A data file was generated and the TPOLAR computer code was developed to determine from the orbital elements, the time, latitude, and MLT of the extremum latitude of each transpolar segment of orbit. The precision of the vector data set from MAGSAT prompted an extended exploratory phase for data analysis procedures, modeling techniques and phenomenology.

    Notes: Standard histological and SEM techniques have been used to examine the pair of statocyst organs located in the telson of the isopod, Cyathara polita. Each organ is formed as an invagination of the dorsal cuticle of the telson. The invagination narrows to form a stalk between the statocyst and dorsal surface. A canal courses longitudinally through this stalk and forms a continuous channel between the lumen of the cyst and the external environment. On the luminal floor of each statocyst, there are three pits; each correlates with a nodule protruding from the ventro-medial wall. From each pit, a single, bifurcating hair projects dorsally to contact the single concretion within the statocyst lumen. No other static organs have been found in this animal. Thus, maintenance of equilibrium in this species appears to be under the control of but six hairs, three in each statocyst. Innervation of each statocyst is provided by a branch of a nerve which connects anteriorly with the last abdominal ganglion.

    Notes: Assemblies of protein molecules represent a fundamental level of biological organization. The dynamic behavior of these systems-including both the assembly process and functional rearrangements-may be accounted for by the specificity of the protein interactions, which depend on environmental conditions. Analysis of the self-assembly of virus particles has established that the design of an ordered structure can be built into the specific bonding properties of the constituent proteins. Any structure which can change its state of organization is, by definition, polymorphic. The distinctive aspect of polymorphism in protein structures, contrasted with nonliving states of matter, is that the molecular design has been selected to carry out a function and that this function is part of an integrated system. The differences in molecular conformation and arrangement in all polymorphic structures-for example, allosteric enzymes or ice crystals-depend on the intrinsic interaction properties of the molecules themselves. The structures of ice and water illustrate relations between specificity and polymorphism which are relevant to the form and function of protein assemblies.Two types of polymorphism can be distinguished: modal polymorphism, which is externally moderated, as in phase transitions between different crystals forms; and positional polymorphism, which is internally moderated, as in the different disposition of identical molecules within a single crystal lattice. Positional polymorphism, exemplified by the quasi-equivalent bonding of icosahedral virus coat proteins and the different arrangement of myosin and paramyosin at the center and polar portions of the bipolar filaments, results from specific interactions that are not compatible with a strictly equivalent packing of identical molecules. The structural rearrangements in muscle contraction and the switching between the oxy and deoxy forms of hemoglobin represent the formation of different structures in response to altered external conditions. The different structural states of many protein assemblies are characterized by conserved connections which may be regarded as providing the framework for functional rearrangements. The types of polymorphism displayed by hemoglobin, virus, and muscle proteins demonstrate the relevance of the simple view that the function of a protein is determined by the potential structures it can form.

    Notes: Fifteen alcohol-soluble carbohydrates have been isolated from logphase cultures of Tetrahymena. Preliminary results obtained by parallel chromatography of impure extracts indicate that some of the soluble components were 2-D-deoxyribose, ribose, mannose, fructose, arabinose, xylose, and glucose. Another elution gradient with extracts from cells washed several times, indicates that maltose and glucose were the only identifiable components as compared with 18 known standards. Differences were observed in the alcohol-soluble carbohydrate content in log-phase versus stationary-phase cells. For example, it is clearly indicated that component 13 is missing in stationary-phase cells while component 15 is missing in log-phase cells. Fifteen alcohol-soluble carbohydrates have been chromatographed from synchronized cells. All of these compounds show cyclic variation in relation to cell division. Component 13 accumulates in cells blocked in morphogenesis and decreases as oral development proceeds in synchronized cells.

    Notes: Experiments will be described which demonstrate that the product of purified “Qβ-replicase” is fully competent to serve both as a template and as a program for the synthesis of complete virus particles. The use of mutant RNA has permitted the demonstration that nucleic acid is the instructive agent in the replication process and hence satisfies the definition of a self-duplicating entity. Methods have been devised to examine the product both physically and biologically with a minimum of manipulation and with complete recovery of product and input template. A detailed analysis of every interval of synthesis has thus become possible. These technical advances have permitted us to demonstrate the existence of a latent period prior to the appearance of the first new complete infectious RNA molecules. Further, this latent period is accompanied by an eclipse of the input templates as infectious agents. The use of electrophoretic separation on acrylamide gels has yielded a detailed account of both templates and early product during the latent period, with the consequent identification of the intermediate stages. The resulting data and their implications for the mechanism of RNA replication will be discussed.

    Notes: As a component of studies to search for effects of 60-Hz electric field exposure on mammalian endocrine function, concentrations of melatonin, 5-methoxytryptophol, and serotonin-Nacetyl transferase activity were measured in the pineal glands of rats exposed or sham-exposed at 65 kV/m for 30 days. In two replicate experiments there were statistically significant differences between exposed and control rats in that the normal nocturnal increase in pineal melatonin content was depressed in the exposed animals. Concentrations of 5-methoxytryptophol were increased in the pineal glands of the exposed groups when compared to shamexposed controls. An alteration was also observed in serotonin-N-acetyl transferase activity, with lower levels measured in pineal glands from exposed animals.
