Beschreibung: We report measurements of eight primary volatiles (H2O, HCN, CH4, C2H6, CH3OH, C2H2, H2CO, and NH3) and two product species (OH and NH2) in comet 103P/Hartley-2 using high dispersion infrared spectroscopy. We present production rates for individual volatiles species, their mixing ratios relative to water, and their spatial distributions in the coma on multiple dates that span the interval Sept. - Dec. 2010. The production rates vary strongly with nucleus rotation, but the mixing ratios remain constant throughout the campaign. The released primary volatiles exhibit diverse spatial properties which favor the presence of separate polar and apolar ice phases in the nucleus, establish dust and gas release from icy clumps (and also, directly from the nucleus), and provide insights into the driver for the cyanogen (CN) polar jet.

Beschreibung: A highly noise-suppressed TF 34 engine was used to investigate the noise of several powered lift configurations involving upper-surface-blown (USB) flaps. The configuration variables were nozzle type (i.e. slot and circular with deflector), flap chord-length, and flap angle. The results of velocity surveys at both the nozzle exit and the flap trailing edge are used for correlation of the noise data. Configurations using a long flap design were 4 dB quieter than a short flap typical of current trends in USB flap design. The lower noise for the long flap is attributed primarily to the greater velocity decay of the jet at the flap trailing edge. The full-scale data revealed substantially more quadrupole noise in the region near the deflected jet than observed in previous sub-scale tests.

Beschreibung: Flight tests were performed to evaluate the vortex wake characteristics of a Boeing 727 aircraft during conventional and two-segment instrument landing approaches. Smoke generators were used for vortex marking. The vortex was intentionally intercepted by a Lear Jet and a Piper Comanche aircraft. The vortex location during landing approach was measured using a system of phototheodolites. The tests showed that at a given separation distance there are no readily apparent differences in the upsets resulting from deliberate vortex encounters during the two types of approaches. The effect of the aircraft configuration on the extent and severity of the vortices is discussed.

Beschreibung: The NASA Planetary Science Summer School (PSSS) at JPL offers graduate students and young professionals a unique opportunity to learn about the mission design process. Program participants select and design a mission based on a recent NASA Science Mission Directorate Announcement of Opportunity (AO). Starting with the AO, in this case the 2009 New Frontiers AO, participants generate a set of science goals and develop a early mission concept to accomplish those goals within the constraints provided. As part of the 2010 NASA PSSS, the Ganymede Interior, Surface, and Magnetosphere Observer (GISMO) team developed a preliminary satellite design for a science mission to Jupiter's moon Ganymede. The science goals for this design focused on studying the icy moon's magnetosphere, internal structure, surface composition, geological processes, and atmosphere. By the completion of the summer school an instrument payload was selected and the necessary mission requirements were developed to deliver a spacecraft to Ganymede that would accomplish the defined science goals. This poster will discuss those science goals, the proposed spacecraft and the proposed mission design of this New Frontiers class Ganymede observer.

Beschreibung: We report measurements of eight primary volatiles (H2O, HCN, CH4, C2H6, CH3OH, C2H2, H2CO, and NH3) and two product species (OH and NH2) in comet lO3P/Hartley-2 using high dispersion infrared spectroscopy. We quantified the long- and short-term behavior of volatile release over a three-month interval that encompassed the comet's close approach to Earth, its perihelion passage, and flyby of the comet by the Deep Impact spacecraft during the EPOXI mission. We present production rates for individual species, their mixing ratios relative to water, and their spatial distributions in the coma on multiple dates. The production rates for water, ethane, HCN, and methanol vary in a manner consistent with independent measures of nucleus rotation, but mixing ratios for HCN, C2H6, & CH3OH are independent of rotational phase. Our results demonstrate that the ensemble average composition of gas released from the nucleus is well defined, and relatively constant over the three-month interval (September 18 through December 1,7). If individual vents vary in composition, enough diverse vents must be active simultaneously to approximate (in sum) the bulk composition of the nucleus. The released primary volatiles exhibit diverse spatial properties which favor the presence of separate polar and apolar ice phases in the nucleus, establish dust and gas release from icy clumps, and from the nucleus, and provide insights into the driver for the cyanogen (CN) polar jet. The spatial distributions of C2H6 & HCN along the near-polar jet (UT 19.5 October) and nearly orthogonal to it (UT 22.5 October) are discussed relative to the origin of CN. The ortho-para ratio (OPR) of water was 2.85 +/- 0.20; the lower bound (2.65) defines T(sub spin) 〉 32 K. These values are consistent with results returned from ISO in 1997 .

Beschreibung: In the lunar magma ocean model, ferroan anorthosites (FANs) are expected to be derived from a relatively undifferentiated source with chondritic REE abundances and yield the oldest radiometric ages of any lunar rocks. Samarium-neodymium-isotopic analyses of 60025 and 67016 yielded ages of 4.44 +/- 0.02 Ga and 4.53 +/- 0.12 Ga and initial Epsilon(sub Nd)(sup 143) values in excess of -0.5. These ages are within error of ages determined on some Mg-suite rocks by researchers. Thus, the existing data are not completely consistent with the standard magma ocean model. We have completed isotopic analysis on the pristine FAN 62236 in order to place further constraints on lunar crust-forming processes.

Notizen: The multifunctional transcription factor YY1 is associated with the nuclear matrix. In osteoblasts, the interaction of several nuclear matrix-associated transcription factors with the bone specific osteocalcin gene contributes to tissue-specific and steroid hormone-mediated transcription. A canonical nuclear matrix targeting signal (NMTS) is present in all members of the AML/CBFβ transcription factor family, but not in other transcription factors. Therefore, we defined sequences that direct YY1 (414 amino acids) to the nuclear matrix. A series of epitope tagged deletion constructs were expressed in HeLa S3 and in human Saos-2 osteosarcoma cells. Subcellular distribution was determined in whole cells and nuclear matrices in situ by immunofluorescence. We demonstrated that amino acids 257-341 in the C-terminal domain of YY1 are necessary for nuclear matrix association. We also observed that sequences within the N-terminal domain of YY1 permit weak nuclear matrix binding. Our data further suggest that the Gal4 epitope tag contains sequences that affect subcellular localization, but not targeting to the nuclear matrix. The targeted association of YY1 with the nuclear matrix provides an additional level of functional regulation for this transcription factor that can exhibit positive and negative control. J. Cell. Notizen: Various types of cells from the testes of mice and hamsters were separated according to differences in sedimentation velocity by centrifugal elutriation, a counterflow centrifugation technique. Approximately 3 × 108 cells, prepared from six mouse testes or from one hamster testis, were separated into 11 fractions in less than two hours as compared to the 4-5 hours required for sedimentation at unit gravity (“Staput”). Fractions enriched in elongated spermatids and spermatozoa (100%), stages 1-8 spermatids (69%) and pachytene spermatocytes (58%) were obtained from mouse testis dispersions. Similarly enriched fractions were obtained from hamster cells. A single fraction enriched in stages 1-8 spermatids (mouse) was prepared in less than 30 minutes. As many as 2 × 109 cells were separated in a single procedure. Spermatogenic cells exhibited no evidence of structural damage with trypan blue and phase microscopy, and recovery was essentially 100%. Centrifugal elutriation had no effect on sperm motility or on the plating efficiency of CHO cells.

Notizen: Potassium (K+) conductances are known to be involved in cell proliferation of a number of nonexcitable cell types. The nature of the mechanism by which K+ channel inhibition reduces cell proliferation has remained elusive despite intensive search. We investigated whether such a phenomenon could be demonstrated in excitable cells, using the GH3 pituitary cell line as a cell model. Our aims were: (1) to study the effect of K+ channel inhibition on the proliferation of GH3 cells; and (2) to investigate the putative intracellular signals involved in this inhibition. Tetraethylammonium chloride (TEA), a blocker of the calcium (Ca2+)-dependent K+ conductances of GH3, was found to reversibly inhibit cell proliferation, as measured by 3H-thymidine incorporation. Cell cycle block specifically occurred at the G1/S phase of the cell cycle. This inhibition of proliferation was observed for 1-4 mM TEA, which suppressed most of the Ca2+-activated K+ current and part of the inward rectifying K+ current, as shown by electrophysiological experiments. Increasing extracellular K+ concentrations with KCl also inhibited cell proliferation in a dose-dependent manner. Both TEA and KCl depolarized the cells and increased intracellular Ca2+ levels ([Ca2+]i), showing that, in this type of excitable cell, inhibition of cell proliferation can be associated with elevated Ca2+ levels. Ca2+ and membrane resting potential (MRP) were considered as possible messengers of this inhibition. Our results suggest that cell cycle arrest of GH3 cells by K+ channel block probably involves an additional pathway, distinct from those of Ca2+ and MRP. J. Cell.
