Benzylpenicilline has a very marked absorption at 240 mμ, as compared with benzylpenicilloic acid, its degradation product by penicillinase. The diminution of the extinction, which a solution of benzylpenicilline presents progressively at this wave length in the presence of penicillinase, is proportional to the amount of penicilline transformed. The final value of the extinction at 240 mμ after complete hydrolysis of the penicilline primitively present can be determined immediately by measuring the not varying extinction at 262,5 mμ, as at this wave length the extinction of benzylpenicilloic acid is equal to its extinction at 240 mμ, and the extinctions of equivalent quantitites of benzylpenicilline and benzylpenicilloic acid are the same. A method of determination of penicillinase based on these facts is described.
