    Notes: An in situ caged crayfish experiment was conducted in the Wabigoon River System, Ontario, Canada to determine the relative importance of food and water pathways for mercury accumulation in crayfish. Two groups of 160 crayfish were suspended in the water column; one at an ucontaminated site, the other at a location with highly elevated total mercury and methylmercury levels in water. Crayfish at each site were divided into two groups. Crayfish were fed either sucker flesh obtained from an uncontaminated lake (low mercury diet) or walleye flesh from contaminated Clay Lake, Ontario, Canada (high mercury diet). After 10 weeks crayfish at both sites fed the high mercury diet had over 20 times mercury accumulation compared to crayfish on the low mercury diet. There was no statistical effects due to sharply elevated water concentrations of total and methylmercury on mercury bodyburdens. This indicated that food was the dominant pathway for mercury accumulation in crayfish.

    Notes: Synthesis of 5-methyl-oxymorphone (3) was accomplished by oxidation of 5-methylthebaine (4) with performic acid, followed by catalytic hydrogenation and cleavage of the 3-MeO group. X-Ray analysis confirmed that the 14-OH group has, like the one in oxymorphone (1), β-orientation. Pharmacological studies in vivo and in vitro showed 3 to possess slightly less opioid agonistic properties than 1.

    Notes: The N,N-dimethyl-8-nitro-1-naphthaleneamine molecule has been observed in seven environments in three crystal modifications. A range of conformations, related by conrotatory motions about the two exocyclic C—N bonds, is observed. In all seven conformations, the lone pair of the amino N-atom is directed approximately towards the nitro N-atom, which is slightly pyramidalized towards the amino group. The NO2 group appears to be a poorer through-space electron acceptor than the carboxylic ester group, in contrast to their through-bond electron-accepting properties.

Spectroscopical Measurements and HMO-π-Calculations to the π-Electronic Structure of p,p′-Disubstituted trans-Stilbenes and Substituted trans-β-Styryl-methyl-SulfonesThe 1H and 13C n.m.r. spectra have been measured for a series of donator-acceptor-disubstituted trans-stilbenes 1 and monosubstituted β-styryl-methyl-sulfones 2. The 13C n.m.r.-signals have been identified by comparison with empirical calculated chemical shifts and quantum chemical determined HMO-π-electrons densities. The influence of substitutents to the experimental chemical shifts are discussed in detail. For 1 and 2 exist linear correlations between chemical shifts and HAMMETT values, and π-electron densities, respectively.

    Notes: Mammary-derived growth inhibitor (MDGI), a polypeptide growth inhibitor isolated from lactating bovine mammary tissue, previously shown to have extensive sequence homology with fatty acid-binding proteins, was demonstrated to meet the criteria of a fatty acid-binding protein. The protein was found to bind [3H]palmitic acid in a saturable manner and to be complexed with endogeneous free fatty acids. [3H]palmitic acid, when bound to the protein, was more rapidly taken up by the target cells (human mammary carcinoma cells [MaTu]) than was free [3H]palmitic acid, suggesting a lipid carrier function for the inhibitor. It is suggested that the fatty acid-binding properties of MDGI may relate to its ability to inhibit cell growth in vitro and to regulate other cellular functions.

    Notes: We have found that polyacrylamide gel electrophoresis in 3% gels in the presence of sodium dodecyl sulfate is suitable for the separation of cellular glycoproteins having molecular weights ranging from 200,000 to 1,000,000. The gels secured on a rigid support (Gelbond) allow blotting techniques with lectins and antibodies for the detection of glycoproteins. Using these methods we have separated lysates of HT-29 human colon carcinoma cells and detected at Jeast four distinct high molecular weight Sialoglycoproteins having molecular weights of 900,000, 740.000, 560,000, and 450,000. The expression of the 9000,000 component, as revealed by wheat germ agglutinin binding, was much higher in a subline of HT-29 cells established from liver metastases in a nude mouse than it was in the parental cells. The relative intensity of wheat germ agglutinin binding to these four sialoglycoprotein components differs depending upon their growth phase in vitro. These glycoproteins were also detectable by the binding of peanut agglutinin, provided the glycoproteins were previously treated in the gels with mild acid to remove the sialic acid from their carbohydrate chains, suggesting that mucin-type carbohydrate chains are present on these glycoproteins. The same set of glycoproteins can be detected by metabolic labeling of the cells with [3H] glucosamine in tissue culture. Very similar glycoprotein profiles are revealed by metabolic labeling of fresh colon carcinoma tissues with [3H] glucosamine in vitro.

    Notes: Early passage mouse embryo fibroblasts, mouse 3T3 cell lines, and early passage diploid human fibroblasts grew to higher cell densities in tissue culture medium supplemented with serum than in medium supplemented with defibrinogenated platelet-poor plasma (PPP). Unlike the mouse cells, the human fibroblasts displayed this differential growth response only in the presence of hypophysiologic concentrations of calcium. The addition of heat-treated extracts of human platelets to PPP-supplemented medium stimulated the replication of both the normal mouse cells and early passage human embryo fibroblasts.Human or mouse fibroblasts transformed by either retroviruses or by SV40, including SV40 infected “serum revertants” and “flat transformants,” grew to equal cell densities in medium supplemented with either serum or PPP. Infection of Balb/c-3T3 cells with SV40 rapidly induced them to grow in PPP-supplemented medium demonstrating that the ability of SV40-transformed cell lines to proliferate in PPP-supplemented medium does not arise from the cell culture selection procedures usually employed to obtain stable virus-transformed cell lines. 3T3 cells infected but not transformed by retroviruses do not replicate in PPP-supplemented medium demonstrating that reduction of the growth requirement for the platelet growth factor(s) by retroviruses is a transformation-specific response. Cell cultures that did not proliferate well in PPP-supplemented medium did not form tumors when inoculated into athymic nude mice. Many, although not all, of the lines which grew well in PPP medium were tumorigenic in nude mice. Together, these findings indicate that: (1) normal fibroblast-like cells display a growth requirement for factor(s) present in serum but not found in PPP; (2) this serum specific growth factor is derived from platelets; (3) a primary response to viral transforming genes is a reduction in the growth requirement for these platelet-derived factors; and (4) cells that have a reduced requirement for the platelet-derived growth factor are often tumorigenic.

    Notes: Bovine adrenal and brain cortex and corpus luteum-derived capillary endothelial cells have been established in culture, taking advantage of their ability to proliferate at clonal density when maintained on extracellular matrix (ECM) coated dishes in the presence of serum supplemented medium. All three cell types formed at confluency a monolayer of small, tightly packed, contact inhibited cells that express factor VIII related antigen. Their proliferative response to basic and acidic FGF when cells were maintained on plastic and exposed to serum supplemented medium was similar to that previously reported for endothelial cells derived from large vessels, with acidic FGF being 30-fold less potent than basic FGF. Their requirement for high density lipoproteins and transferrin in order to proliferate actively when maintained on ECM-coated dishes and exposed to serum-free conditions was also similar to that previously reported for endothelial cells derived from large vessels. Heparin strongly reduced the proliferative response of capillary endothelial cells to either basic or acidic FGF, as well as their response to serum alone, regardless of whether cells were maintained on plastic or on ECM-coated dishes. The present data indicate that bovine endothelial cells derived from large or small vessels are indistinguishable in so far as their response to growth factors, plasma factors, and substrata are concerned.

    Notes: Studies were conducted to explore the effects of mevinolin, a competitive inhibitor of HMG CoA reductase, on the growth and morphology of normal and transformed murine fibroblasts. Mevinolin is known to block DNA synthesis and cell growth in a number of kinds of non-transformed cells. Eight cell lines were studied, including two normal fibroblast cell lines (C3H 10T 1/2 and NIH 3T3) and derivatives of these cell lines transformed by chemical carcinogens, X-irradiation or the H-ras oncogene. All of the eight cell lines displayed appreciable growth inhibition by 5 μM mevinolin and marked inhibition by 30 μM mevinolin. Mevinolin also induced a marked rounding in the morphology of all of the cell lines. These effects of mevinolin on cell growth and morphology were blocked or reversed by the addition of mevalonic acid. Thus, both normal and transformed cells require mevalonate, or an as yet unidentified metabolite of mevalonate for their growth, even though some transformed cells have become relatively autonomous of other growth factors. Whereas mevinolin acted primarily as a cytostatic agent for most of the cell lines studied, with the transformed cell line MCA/10T 1/2, which ordinarily grows to a very high cell density, prolonged exposure to mevinolin caused marked cytotoxicity. Thus mevinolin might be useful as an anti-tumor agent for specific tumors.
