    Notes: Biochemical evidence is presented for the autophagic destruction of liver mitochondria in the influenza B virus model of Reye's syndrome in mice. Separation of lysosomes and autophagic vacuoles from mitochondria was accomplished by prior treatment of the mice with Triton WR-1339, resulting in uptake of detergent by these organelles (tritosomes), reducing their densities. The organelles were banded in a discontinuous sucrose gradient. Total protein in the heavy tritosomal fraction increased from 1-2% in controls to 7-8% in virus-treated animals. Ornithine carbamoyl transferase (OCTase), a mitochondrial marker, increased from 2-3% (controls) to 11-15% (virus-treated), and glucose-6-phosphatase, a marker for endoplasmic reticulum, increased from 1-2% (controls) to 8-10% (virus-treated). β-Galactosidase, a soluble enzyme in the lysosome, and OCTase also increase in the cell extract fraction following virus treatment, indicating that there was turnover of heavy lysosomal contents.

    Notes: Macromolecules are sulfated during the vegetative growth of Dictyostelium discoideum. A characterisation of the structures of sulfated oligosaccharides associated with these macromolecules indicates that the oligosaccharides are heterogeneous. Endoglycosidase and pronase digestion were used with gel-filtration chromatography to obtain two different Oligosaccharide fractions and a glycopeptide fraction; these were further characterised by ion-exchange and lectin-affinity chromatography and by acid hydrolysis. The data indicate that up to 43% of the sulfate is associated with typical N-linked oligosaccharides, that up to 5% is associated with N-linked oligosaccharides that are either very large or extremely highly charged, and that the remaining sulfate is associated with oligosaccharides non-N-linked to protein. Each fraction was also shown to be heterogeneous at most other structural levels. Electrophoretic analyses following the endoglycosidasc and pronase treatments indicated that all of the macromolecules are glycoproteins and suggested further that at least two of the Oligosaccharide fractions are located on different groups of glycoproteins.

    Notes: Western blotting and immunoprecipitation data indicated that lysosomal enzymes represent a subset of the sulfated macromolecules present in vegetative Dictyostelium discoideum amoebae and account for less than 2.5% of the total sulfate incorporated during vegetative growth. These data suggest that the majority of the highly sulfated macromolecules of vegetative D. discoideum amoebae are not related to the lysosomal enzymes.

    Notes: The amdS gene of A. nidulans has proved extremely favourable for the isolation of mutations affecting gene regulation. Trans-acting regulatory genes involved in amdS induction by small molecular weight effectors have been identified - amdR (ω-amino acids) facB (acetate) and amdA (acetate). Another gene, the areA gene, has properties expected of a major activator gene involved in nitrogen metabolite repression of amdS. All of these regulatory genes are also involved in the control of various other functions encoded by structural genes unlinked to amdS. Mutations in the 5′-region adjacent to amdS have been isolated and allow the identification of independent cis-acting sequences which are the target sites for the regulatory genes. The involvement of these sequences in regulatory product binding has been deduced from titration studies using transformants containing multiple copies of the 5′ sequences. A combination of genetics and molecular analysis is allowing a detailed characterization of this system.

    Notes: Hepatic sinusoidal Ito cells have the capacity to produce interstitial collagen types I and Ill as well as other matrix proteins and may be involved in hepatic fibrogenesis. Transforming growth factor β (TGF β) responsiveness was evaluated during in vitro cell culture, since increasing evidence suggests that this ubiquitous polypeptide can stimulate the production of collagenous proteins in a variety of cell types. TGF β induced marked inhibition of Ito cell proliferation for cells grown on either a type I or a type IV collagen matrix. In marked contrast, the collagen synthetic response was considerably different for cells grown on a type I versus a type IV collagen matrix. When cells were grown on a type I collagen matrix, TGF β caused a significant increase in the accumulation of collagen type I and III. When Ito cells were grown on a type IV collagen matrix, there was no stimulation of collagen production. TGF β respsonsiveness was also evaluated in the setting of altered vitamin A concentrations. Freshly isolated Ito cells are engorged with vitamin A, the usual physiologic storage site for hepatic vitamin A. During in vitro culture and during in vivo fibrogenesis, Ito cells lose their vitamin A stores coincident with a transformation to a collagen-producing myofibroblast-like cell. When cultured Ito cells were grown on a type I collagen matrix and re-exposed to an increased concentration of vitamin A, the production of interstitial collagen was reduced. However, when the vitamin A-enriched Ito cells were exposed to TGF β, the production of interstitial collagen was increased, similar to cells that had not received vitamin A.

    Notes: The yeast Brettanomyces anomalus showed the Custers effect in that under strictly anaerobic conditions, in the presence of glucose, CO2 production was negligble. CO2 production was stimulated by mixing anaerobic cell suspensions with an aerated glucose solution in astopped-flow cell. Glycolytic CO2 production continued even after oxygen exhaustion. Studies using an open reaction vessel showed that the rate of glycolytic CO2 production could be increased to a maximum level by exposing the anaerobic cell suspension to brief pulses of O2. A cell suspension CO2 at a maximal rate demonstrated the Pasteur effect on switching the mobile gas to a mixture conatining oxygen (5.05 KPa). In contrast to glycolytic CO2 production in vivo nicotinamide pool responded rapidly to changes in oxygen concentration. The addition of acetaldehyde, acetone, or 3-hydroxy-butan-2-one led to a temprorary production of CO2 at an initial rate depending on the concentration of substance added according to the Michaelis-Menten equation. The maximal rates were equal with all three substances, whereas tha apparent Km values were different. The total amount of CO2 produced was 22-fold greater than the amount of acetaldehyde added. Added organic hydrogen acceptors modulated the intracellular reedox balance of B. anomalus under conditions. These results are discussed in relation to the current hypothesis of the Custers effect.

    Notes: A computer-aided graphics approach to correlating transmission electron microscope images of freeze-fractured and thin-sectioned samples is outlined. Any three-dimensional model of the imaged structure can be mathematically sectioned to provide a two-dimensional representation of the model in the “fracture” plane. The method is used to demonstrate that the structure of lamellar liquid crystalline liposomes is based on a family of Dupin cyclides; closed, parallel surfaces with a conjugate ellipse and hyperbola as curvature defects.

    Notes: The controlled environment vitrification system (CEVS) permits cryofixation of hydrated biological and colloidal dispersions and aggregates from a temperature- and saturation-controlled environment. Otherwise, specimens prepared in an uncontrolled laboratory atmosphere are subject to evaporation and heat transfer, which may introduce artifacts caused by concentration, pH, ionic strength, and temperature changes. Moreover, it is difficult to fix and examine the microstructure of systems at temperatures other than ambient (e.g., biological systems at in vivo conditions and colloidal systems above room temperature). A system has been developed that ensures that a liquid or partially liquid specimen is maintained in its original state while it is being prepared before vitrification and, once prepared, is vitrified with little alteration of its microstructure. A controlled environment is provided within a chamber where temperature and chemical activity of volatile components can be controlled while the specimen is being prepared. The specimen grid is mounted on a plunger, and a synchronous shutter is opened almost simultaneously with the release of the plunger, so that the specimen is propelled abruptly through the shutter opening into a cryogenic bath. We describe the system and its use and illustrate the value of the technique with TEM micrographs of surfactant microstructures in which specimen preparation artifacts were avoided. We also discuss applications to other instruments like SEM, to other techniques like freeze-fracture, and to novel “on the grid” experiments that make it possible to freeze successive instants of dynamic processes such as membrane fusion, chemical reactions, and phase transitions.

    Description: Space motion sickness clinical characteristics, time course, prediction of susceptibility, and effectiveness of countermeasures were evaluated. Although there is wide individual variability, there appear to be typical patterns of symptom development. The duration of symptoms ranges from several hours to four days with the majority of individuals being symptom free by the end of third day. The etiology of this malady remains uncertain but evidence points to reinterpretation of otolith inputs as being a key factor in the response of the neurovestibular system. Prediction of susceptibility and severity remains unsatisfactory. Countermeasures tried include medications, preflight adaptation, and autogenic feedback training. No countermeasure is entirely successful in eliminating or alleviating symptoms.
