    Beschreibung: Yeast artificial chromosomes (YACs) were obtained from a 550-kilobase region that contains three probes previously mapped as very close to the locus of the fragile X syndrome. These YACs spanned the fragile site in Xq27.3 as shown by fluorescent in situ hybridization. An internal 200-kilobase segment contained four chromosomal breakpoints generated by induction of fragile X expression. A single CpG island was identified in the cloned region between markers DXS463 and DXS465 that appears methylated in mentally retarded fragile X males, but not in nonexpressing male carriers of the mutation nor in normal males.     Notizen: Nascent high-molecular-weight (UHMW) polyethylene (PE) samples of different origins show a rather high crystallinity of about 70-75% and contain both a major portion of orthorhombic extended chain crystallites and a minor portion of triclinic crystallites. The triclinic content is greater the higher the molecular weight of the sample and the higher the activity of the used catalyser. A melting / recooling treatment results in a reduction of crystallinity by about 15-25% and disappearance of the triclinic phase. Further, an irreversible conversion of nascent orthorhombic extended chain crystallites to orthorhombic folded chain crystallites of increased lateral dimensions and crystalline perfections takes place during the melting / recooling treatment. The results are compared to those obtained for lower-molecular-weight PE samples and for high-strength / modulus PE fibers of different origins.

    Notizen: Bilirubin, a bile pigment, was studied for its extent of adsorption on substituted aminechloromethyl polystyrene by UV spectrophotometry. By performing simple displacement reactions on chloromethyl polystyrene with secondary and tertiary amines, the amount of charge density and steric factor on substituted nitrogen atom have been varied. Adsorption isotherms of bilirubin at 0°C by different amine-chloromethyl polystyrene homologs suggest the existence of electrostatic interaction of polymeric resin with bilirubin moiety. Results of adsorption of bilirubin to polymeric resin have shown that the extent of adsorption of bilirubin depends on the unit charge, and on the structure of the substituted amine-chloromethyl polystyrene. The effects of porosity of resin on bilirubin adsorption have also been discussed.

    Beschreibung: The human immunodeficiency virus-1 (HIV-1) trans-activator Tat is an attractive target for the development of antiviral drugs because inhibition of Tat would arrest the virus at an early stage. The drug Ro 5-3335 [7-chloro-5-(2-pyrryl)-3H-1,4-benzodiazepine-2(H)-one], inhibited gene expression by HIV-1 at the level of transcriptional trans-activation by Tat. The compound did not inhibit the basal activity of the promoter. Both Tat and its target sequence TAR were required for the observed inhibitory activity. Ro 5-3335 reduced the amount of cell-associated viral RNA and antigen in acutely, as well as in chronically infected cells in vitro (median inhibition concentration 0.1 to 1 micromolar). Effective inhibition of viral replication was also observed 24 hours after cells were transfected with infectious recombinant HIV-1 DNA.     Beschreibung: Severe combined immunodeficient (SCID) mice reconstituted with human peripheral blood leukocytes (hu-PBL-SCID mice) have inducible human immune function and may be useful as a small animal model for acquired immunodeficiency syndrome (AIDS) research. Hu-PBL-SCID mice infected with human immunodeficiency virus-1 (HIV-1) contained virus that was recoverable by culture from the peritoneal cavity, spleen, peripheral blood, and lymph nodes for up to 16 weeks after infection; viral sequences were also detected by in situ hybridization and by amplification with the polymerase chain reaction (PCR). Mice could be infected with multiple strains of HIV-1, including LAV-1/Bru, IIIB, MN, SF2, and SF13. HIV-1 infection affected the concentration of human immunoglobulin and the number of CD4+ T cells in the mice.     Beschreibung: Three spatially distant surface loops were found to mediate the interaction of the coagulation protein factor X with the leukocyte integrin Mac-1. This interacting region, which by computational modeling defines a three-dimensional macromotif in the catalytic domain, was also recognized by glycoprotein C (gC), a factor X receptor expressed on herpes simplex virus (HSV)-infected endothelial cells. Peptidyl mimicry of each loop inhibited factor X binding to Mac-1 and gC, blocked monocyte generation of thrombin, and prevented monocyte adhesion to HSV-infected endothelium.     Notizen: The Wurtz-type coupling of a number of representative classes of substituted dichlorosilane monomers was investigated in considerable detail. Low temperature polymerization techniques, including ultrasonic activation, were also studied. Similarly, the effect of dipolar additives on the polymerization process was examined. The conclusions drawn from these studies provide some insight into the selection of useful polymerization conditions for the conversion of various monomer types into linear high polymers.

    Notizen: High molecular weight alternating block copolymers of polyethesulphone (PES) and polydimethylsiloxane (PDMS) were prepared by the condensation of dimethylamino-terminated PDMS oligomers and hydroxy-terminated PES oligomers in 1,2-dichlorobenzene. Microphase separation of the block copolymers at exceptionally short block lengths was observed by differential scanning calorimetry (DSC) and transmission electron microscopy (TEM). The Si—O—C intersegment linkage in these materials appeared to display poor hydrolytic stability which is contrary to results obtained for other block copolymers.
