    Notes: The morphological and ultrastructural changes that occur during preparation of porcine, bovine, and murine spermatozoa for flow cytometric quantification of the relative DNA content of the X- and Y-chromosome-bearing sperm populations were examined. Ejaculated spermatozoa from the boar and bull were washed using a series of dimethyl sulfoxide (DMSO) solutions prior to fixation, whereas the epididymal mouse spermatozoa were washed only in phosphate-buffered saline (PBS). Spermatozoa from all three species were then fixed in ethanol and processed for fluorochrome staining by a treatment regimen consisting of sulfhydryl reduction and proteolysis. The processed sperm nuclei were stained for DNA with the fluorochrome, 4′-6-diamidino-2-phenylindole (DAPI) before quantification by flow cytometry. Scanning and transmission electron micrographs of sperm heads taken at various steps of the preparation and staining procedures show 1) that the rigorous washing procedure disrupted the plasma and outer acrosomal membranes, 2) that ethanol fixation resulted in removal of the outer membranes and disintegration of the nuclear envelope, and 3) that thiol and proteolysis treatment removed the remaining cellular organelles including the tail and rapidly induced partial decondensation of the tightly packed chromatin. Sequential micrographs showed that the nuclear matrix of all three species increased in thickness about twofold during the preparation and staining. Consequently, the harsh procedures currently used for quantitative staining of DNA for high-resolution flow cytometric analyses destroy most cellular organelles and thereby prevent simultaneous characterization of DNA content and other sperm cell constituents.

    Description: A liquid-helium-cooled grating spectrometer (CGS) is being developed as a facility instrument for the Kuiper Airborne Observatory (KAO), primarily to study for infrared lines originating in the interstellar medium. A maximum resolving power of approximately 6000 is achieved by means of a 45 cm long Echelle grating and is optically capable of operating in the spectral range from 25 to 300 microns. An array of detectors is used to simultaneously measure a line and the adjacent continuum from astronomical sources. Currently six detectors allow measurements in the 30 to 120 micron spectral band. The instrument, its operation, and its performance are described.

    Description: VLBI and CCD astrometry is presented which establishes the coincidence of the X-ray and radio object Cir X-1 with an object lying about one arcsec south of the object 'L' of Whelan et al. (1977), a conclusion reached previously by Argue and Sullivan (1982). The question whether the object L has any physical connection with Cir X-1 cannot yet be definitely answered.

    Description: The catalog of X-ray sources detected during the NRL Large Area Sky Survey (LASS) with the HEAO 1 satellite is presented. The catalog is derived from the first six months of data from HEAO 1 and includes sources detected during one full scan. Positions and intensities for a total of 842 different sources are included, with a limiting flux of 250 nJy at 5 keV. The catalog is more than 90 percent complete at a flux level equivalent to 1.5 microjoules at 5 keV for a Crab-like spectrum. Cross-references with published literature are provided and coincidental identifications are proposed for some of the sources which have been never studied before. A cross-sectional line drawing of the sensor module of HEAO I is also provided.

    Description: Multitemporal Thematic Mapper, Thematic Mapper Simulator, and detailed ground truth data were collected for a 9- by 11-km sample segment in Webster County, IA, in the summer of 1982. Three dates were acquired each with Thematic Mapper Simulator (June 7, June 23, and July 31) and Thematic Mapper (August 2, September 3, and October 21). The Thematic Mapper Simulator data were converted to equivalent TM count values using TM and TMS calibration data and model based estimates of atmospheric effects. The July 31, TMS image was compared to the August 2, TM image to verify the conversion process. A quantitative measure of proportion estimation variance (Fisher information) was used to evaluate the corn/soybeans separability for each TM band as a function of time during the growing season. The additional bands in the middle infrared allowed corn and soybeans to be separated much earlier than was possible with the visible and near-infrared bands alone. Using the TM and TMS data, temporal profiles of the TM principal components were developed. The greenness and brightness exhibited behavior similar to MSS greenness and brightness for corn and soybeans.

    Description: The HCN emission of the pure rotation and rotation/vibration lines in comet IRAS-Araki-Alcock 1983d is examined. The prevailing excitation mechanism for the emissions was the excitation of the nu-2, 2 nu-2, and nu-3 vibrational bands by the solar infrared field. For the description of inner coma, a dynamical excitation model is presented which includes collisions. It is predicted on the basis of the model that HCN molecules in rotation and rotation vibration lines of IRAS-Araki-Alcock 1983d would be detectable with a large-millimeter wave telescope, and that the strongest infrared lines would be observable from space observatories. Subsequent searches for the J = 1-0 HCN radio lines in comet 1983d with the Five College Radio Astronomy Observatory (FCRAO) proved unsuccessful. An extremely low upper limit was found for HCN production which suggests that HCN might not be the only parent of planetary cometary CN.

    Description: Optical filter photometry, and optical and ultraviolet spectrophotometry data collected between November 1980 and June 1982 of Comet Bowell are presented. It was determined that Comet Bowell began producing significant amounts of OH, though not other species, before perihelion at a heliocentric distance near 4.6 AU. As the comet approached perihelion at 3.4 AU, OH production decreased and CN and C2 species were detected at normal concentrations. An outburst in April 1982 was dominated by OH. The grains in 1982 near perihelion showed increases in albedo between 3150 and 4500 A and 1.2 and 1.6 microns. The presence of two grains populations was suggested, together with the conclusion that Comet Bowell was physically a typical new comet entering the inner solar system from the Oort cloud for the first time.

    Description: The use of an image registration program in the studies of urban growth is described. This program permits a quick identification of growing areas with the overlap of the same scene in different periods, and with the use of adequate filters. The city of Brasilia, Brazil, is selected for the test area. The dynamics of Brasilia urban growth are analyzed with the overlap of scenes dated June 1973, 1978 and 1983. The results showed the utilization of the image registration technique for the monitoring of dynamic urban growth.

    Description: The urban growth of Brasilia within the last ten years is analyzed with special emphasis on the utilization of remote sensing orbital data and automatic image processing. The urban spatial structure and the monitoring of its temporal changes were examined in a whole and dynamic way by the utilization of MSS-LANDSAT images for June (1973, 1978 and 1983). In order to aid data interpretation, a registration algorithm implemented in the Interactive Multispectral Image Analysis System (IMAGE-100) was utilized aiming at the overlap of multitemporal images. The utilization of suitable digital filters, combined with the images overlap, allowed a rapid identification of areas of possible urban growth and oriented the field work. The results obtained in this work permitted an evaluation of the urban growth of Brasilia, taking as reference the proposal stated for the construction of the city in the Pilot Plan elaborated by Lucio Costa.
