    Notes: Abstract The reannealing kinetics of denatured DNA fragments from 23 species of higher plants have been studied, using hydroxylapatite chromatography to distinguish reannealed from single-stranded DNA. The 2C nuclear DNA contents of the species varied between 1.7 and 98 pg. The proportions of DNA in species with a nuclear DNA mass above 5 pg that reannealed with the kinetics of sequences present in more than 100 copies were high (69–92% with a mean of 80±2.0%). For species with less than 4 pg of DNA, the mean proportion of repeated-sequence DNA was 62±2.9%. It is concluded that most of the variation in nuclear DNA mass in higher plant chromosomes can be accounted for by variation in repeated-sequence DNA. The consequences of altering the adapted DNA content of a species by the addition of families of repeated sequences are discussed in relation to the proportion of repeated-sequence DNA.

    Notes: Abstract Growth and nitrogenase activity were studied in cultures ofAzotobacter vinelandii growing with dinitrogen, ammonium sulfate, aspartic acid or yeast extract. Nitrogenase activity was measured by means of the C2H2 reduction test. In the presence of ammonium sulfate nitrogenase is completely repressed. After exhaustion of ammonia its activity is restored following a diauxic lag period of 30 min. With aspartic acid nitrogenase activity is partially repressed, and growth yield is higher than in the culture growing with N2 only. This is due to simultaneous use of dinitrogen and aspartate. Fluctuations of nitrogenase activity occurring during exponential growth and the mechanism of their regulation are discussed.

    Notes: Abstract A method is described which allows the quantitative determination of small ammonia concentrations in the culture of nitrogen-fixing microorganisms. With this method the ammonia concentration range was estimated in which repression of nitrogenase synthesis in Azotobacter vinelandii occurs. Both in batch and continuous cultures there was no repression below 10 μM, whereas nitrogenase synthesis stopped completely if the ammonia concentration in the medium exceeded 25 μM.

    Notes: Abstract The kinetics of renaturation of DNA from the rat thymus and ascites hepatoma was studied. Three zones were distinguished on the curve, corresponding to fast, intermediate, and slow rates of DNA reassociation. The course of the reassociation curves of DNA from tumor and normal tissues did not diverge in any of these zones. Consequently, no difference could be detected between the DNA's of normal and neoplastic cells on the basis of their reassociation kinetics.
