Tumorous tissue formation on inoculated stem internodes of sterile-grown plantlets, as well as expression of a β-glucuronidase marker gene interrupted by an intron in cells of inoculated stem nodes, indicate that the monocotyledon Lilium is a host for Agrobacterium.

    Notes: Summary Gene fusions between the β-glucuronidase (GUS) reporter gene and the promoters of the cauliflower mosaic virus 35S RNA transcript (CaMV 35S) and the mannopine synthase (mas) genes were introduced into rapeseed varieties via Agrobacterium-mediated transformation. Fluorometric assay of β-glucuronidase activity indicated different expression patterns for the two promoters. In seedlings, the CaMV 35S promoter had maximum activity in the primary roots, while the mas promoter was most active in the cotyledons. Etiolated seedlings cultured in the dark showed reduced activity of the mas promoter. Before vernalization at the rosette stage, both promoters were more active in older plant parts than in younger ones. At this stage the highest activity was recorded in cotyledons. After the plants had bolted reduced promoter function was detected in the upper parts of the transformed plants. Both promoters were found to be functional in the majority of the studied organs of transgenic rapeseed plants, but the promoter activity varied considerably between the organs at different developmental stages. The ability of pollen to transfer the introduced genes to other varieties and related species (e.g. Brassica napus and Diplotaxus muralis) by cross-pollination was studied in greenhouse experiments, and field trials were carried out to estimate the distance for biologically-relevant gene dispersal. In artificial crossing, the introduced marker gene was transferable into other varieties of Brassica napus. In field trials, at a distance of 1 metre from the source of transgenic plants, the frequency of an outcrossing event was relatively high (10-3). Resistant individuals were found at 16 and 32 metres from the transgenic pollen donors, but the frequency of an outcrossing event dropped to 10-5.

    Notes: Summary Leaf explants of apple cvs Gala and Golden Delicious were infected with the Agrobacterium tumefaciens strain AGL0(pMOG410). The effects of a 2 d preculture of the explants before infection and the addition of 5-azacytidine to the selection medium were studied. The percentages of GUS-positive explants after 5 w did not significantly alter due to these treatments. One of the ‘Gala’ shoots, which was removed from a leaf explant cultured for 8 w on selection medium, proved to be GUS-positive and will be analyzed further. In general, however, it should be concluded that regeneration of transgenic shoots directly from leaf tissue was not very effective.

    Notes: Die Zusammendrückbarkeit von Eis zwischen 100 und 500 Megabar ergab sich zu 0.0000120 bei -7.03°; sie beträgt also nur ¼ der Zusammendrückbarkeit des Wassers bei benachbarten Temperaturen. Es wurde gefunden, daß sie mit zunehmendem Druck nicht merklich abnimmt. Dies ist wahrscheinlich die erste direkte Bestimmung der Zusammendrückbarkeit des gewöhnlichen Eises, die jemals gemacht worden ist. Der Unterschied zwischen unserer niedrigen Zahl und dem von BRIDGMAS durch indirekte Berechnung gefundenen höheren Wert ist wahrscheinlich wenigstens zum Teil auf einen großen Temperaturkoeffizienten zurückzuführen.

    Notes: Experimental information concerning the molecular transport of the lighter hydrocarbons in the gas phase is limited. For this reason a study was made of the Maxwell diffusion coefficients of n-hexane in the gas phase of the methane-n-hexane, and propane-n-hexane systems.Maxwell diffusion coefficients were determined at steady state for pressures up to 70 lb./sq. in. at temperatures between 70° and 220° F. The effects of interfacial resistance were considered and uncertainties as to the behavior at the end of the transport path were eliminated. Coefficients were reported with partial pressure and with fugacity as the potential. Fick diffusion coefficients were calculated for each component on the assumption that the gas phase was an ideal solution.These data indicated that the Maxwell hypothesis with fugacity as the potential in an ideal solution is a fair description of the transport characteristics of the lighter hydrocarbons in the gas phase at relatively low pressures. A regular decrease in the Maxwell diffusion coefficient with an increase in the molecular weight of the stagnant component was observed.

    Notes: A heated jet of air from an 0.898-in. standard A.S.M.E. nozzle was discharged into a 4-in. steel duct, well insulated over its entire 10-ft. length. Air from the region surrounding the nozzle was entrained into the duct. At a number of points along the duct, radial profiles of air velocity and temperature were obtained by means of a probe which combined an impact tube and a thermocouple. The temperature at each of several points along the duct wall was indicated by thermocouples imbedded in the wall.In the experiments reported here the velocity at the jet was 585 ft./sec.; the temperature of the jet was about 220°F. and that of the entrained air was about 88°F. The total air flow rate through the duct was 0.67 lb./sec., and the heat flux was 4.9 B.t.u./sec., with the temperature of the entrained air taken as the datum.The radial and axial profiles of velocity and temperature are compared and discussed; the temperatures of the stream near the duct wall and of the duct wall itself are given. Conservation of mass and heat was checked by graphical integration of the radial profiles.
