    Description: Heterotopic ossification (HO) is the de novo formation of bone that occurs in soft tissue, through recruitment, expansion, and differentiation of multiple cells types including transient brown adipocytes, osteoblasts, chondrocytes, mast cells, and platelets to name a few. Much evidence is accumulating that suggests changes in metabolism may be required to accomplish this bone formation. Recent work using a mouse model of heterotopic bone formation reliant on delivery of adenovirus-transduced cells expressing low levels of BMP2 showed the immediate expansion of a unique brown adipocyte-like cell. These cells are undergoing robust uncoupled oxidative phosphorylation to a level such that oxygen in the microenvironment is dramatically lowered creating areas of hypoxia. It is unclear how these oxygen changes ultimately affect metabolism and bone formation. To identify the processes and changes occurring over the course of bone formation, HO was established in the mice, and tissues isolated at early and late times were subjected to a global metabolomic screen. Results show that there are significant changes in both glucose levels, as well as TCA cycle intermediates. Additionally, metabolites necessary for oxidation of stored lipids were also found to be significantly elevated. The complete results of this screen are presented here, and provide a unique picture of the metabolic changes occurring during heterotopic bone formation. This article is protected by copyright.     Description: Cell and gene therapy approaches are safer when they possess a system that enables the therapy to be rapidly halted. Human mesenchymal stem cells were transduced with an adenoviral vector containing the cDNA for bone morphogenetic protein 2 (AdBMP2) to induce bone formation. To make this method safer, a system to quickly kill these virally transduced cells was designed and evaluated. Cells were encapsulated inside poly(ethylene glycol) diacrylate (PEG-Da) hydrogels that are able to shield the cells from immunological destruction. The system involves an inducible caspase-9 (iCasp9) activated using a specific chemical inducer of dimerization (CID). Delivering AdBMP2-transduced human mesenchymal stem cells encapsulated in PEG-Da hydrogel promoted ectopic ossification in vivo , and the iCasp9 system allowed direct control of the timing of apoptosis of the injected cells. The iCasp9-CID system enhances the safety of delivering AdBMP2-transduced cells, making it a more compelling therapeutic for bone repair and spine fusion. This article is protected by copyright.     Description: We adapted Newton's Law of Cooling to model downstream water temperature change in response to stream-adjacent forest harvest on small and medium streams (average 327 ha in size) throughout the Oregon Coast Range, USA. The model requires measured stream gradient, width, depth and upstream control reach temperatures as inputs and contains two free parameters which were determined by fitting the model to measured stream temperature data. This model reproduces the measured downstream temperature responses to within 0.4 C ° for 15 of the 16 streams studied and provides insight into the physical sources of site-to-site variation among those responses. We also use the model to examine how the pre-to-post harvest change in daily maximum stream temperature depends on distance from the harvest reach. The model suggests that the pre-to-post harvest temperature change approximately 300  m downstream of the harvest will range from roughly 82% to less than 1% of that temperature change which occurred within the harvest reach, depending primarily on the downstream width, depth, and gradient. Using study-averaged values for these channel characteristics the model suggests that for a stream representative of those in the study, the temperature change approximately 300  m downstream of the harvest will be 56% of the temperature change which occurred within the harvest reach. This adapted Newton's Law of Cooling procedure represents a highly practical means for predicting stream temperature behavior downstream of timber harvests relative to conventional heat budget approaches, and is informative of the dominant processes affecting stream temperature.     Description: Till from moraines at the heads of six major outlet glaciers in the Transantarctic Mountains (TAM) and from till beneath three West Antarctic ice streams have a ubiquitous zircon U-Pb age population spanning the time of the Ross/Pan-African Orogenies (610-475 Ma). Geo- and thermochronology of detrital minerals in these Antarctic glacial tills reveal two different thermal histories for the central and southern TAM. Double-dating of the zircons reveals a geographically widespread (U-Th)/He (ZHe) population of 180-130 Ma in most of the till samples. Sandstone outcrops at Shackleton Glacier, and three Beacon Supergroup sandstone clasts from three moraines, have ZHe ages that fall entirely within this range. The similar population and proximity of many of the till samples to Beacon outcrops lead us to suggest that this extensive ZHe population in the tills is derived from Beacon Supergroup rocks and reflects the thermal response of the Beacon Basin to the breakup of Gondwana. A second population of older (〉200 Ma) ZHe ages in tills at the head of Byrd, Nimrod and Reedy Glaciers. For the tills at the head of the Nimrod and Byrd Glacier, integrating the double-dated zircon results with 40 Ar/ 39 Ar of hornblende, muscovite and biotite, and U-Pb and (U-Th-Sm)/He double-dates on apatite yields a typical pattern of early rapid orogenic cooling (∼4-10˚C/my) 590-475 Ma after the emplacement of the Granite Harbour Intrusives. Low temperature thermochronometers at these sites yield variable but quite old ages (ZHe 480-70 Ma and AHe 200-70 Ma) that require a long history at low temperature.     Description: Soil moisture is a critical variable in the water and energy cycles. The prediction of soil moisture patterns, especially at high spatial resolution, is challenging. This study tests the ability of a land surface hydrologic model (Flux-PIHM) to simulate high-resolution soil moisture patterns in the Shale Hills watershed (0.08 km 2 ) in central Pennsylvania. Locally measured variables including a soil map, soil parameters, a tree map, and lidar topographic data, all have been synthesized into Flux-PIHM to provide model inputs. The predicted 10-cm soil moisture patterns for fifteen individual days encompassing seven months in 2009 are compared with the observations from 61 soil moisture monitoring sites. Calibrated using only watershed-scale and a few point-based measurements, and driven by spatially uniform meteorological forcing, Flux-PIHM is able to simulate the observed macro spatial pattern of soil moisture at ~10 m resolution (spatial correlation coefficient ~ 0.6) and the day-to-day variation of this soil moisture pattern, although it underestimates the amplitude of the spatial variability and the mean soil moisture. Results show that the spatial distribution of soil hydraulic parameters has the dominant effect on the soil moisture spatial pattern. The surface topography and depth to bedrock also affect the soil moisture patterns in this watershed. Using the National Land Cover Database (NLCD) in place of a local tree survey map makes a negligible difference. Field measured soil type maps and soil type-specific hydraulic parameters significantly improve the predicted soil moisture pattern as compared to the most detailed national soils database (Soil Survey Geographic Database, or SSURGO, 30-m resolution).     Description: Flow paths and residence times in the hyporheic zone are known to influence biogeochemical processes such as nitrification and denitrification. The exchange across the sediment-water interface may involve mixing of surface water and groundwater through complex hyporheic flow paths that contribute to highly variable biogeochemically active zones. Despite the recognition of these patterns in the literature, conceptualization and analysis of flow paths and nitrogen transformations beneath riffle-pool sequences often neglect to consider bed form driven exchange along the entire reach. In this study, the spatial and temporal distribution of dissolved oxygen (DO), nitrate (NO 3 - ) and ammonium (NH 4 + ) were monitored in the hyporheic zone beneath a riffle-pool sequence on a losing section of the Truckee River, NV. Spatially-varying hyporheic exchange and the occurrence of multi-scale hyporheic mixing cells are shown to influence concentrations of DO and NO 3 - and the mean residence time (MRT) of riffle and pool areas. Distinct patterns observed in piezometers are shown to be influenced by the first large flow event following a steady 8 month period of low flow conditions. Increases in surface water discharge resulted in reversed hydraulic gradients and production of nitrate through nitrification at small vertical spatial scales (0.10 to 0.25 m) beneath the sediment-water interface. In areas with high downward flow rates and low MRT, denitrification may be limited. The use of a longitudinal two-dimensional flow model helped identify important mechanisms such as multi-scale hyporheic mixing cells and spatially varying MRT, an important driver for nitrogen transformation in the riverbed. Our observations of DO and NO 3 - concentrations and model simulations highlight the role of multi-scale hyporheic mixing cells on MRT and nitrogen transformations in the hyporheic zone of riffle-pool sequences.     Description: Abstract Hazard analysis at caldera volcanoes is challenging due to the wide range of eruptive and environmental conditions that can plausibly occur during renewed activity. Taupo volcano, New Zealand, is a frequently active and productive rhyolitic caldera volcano that has hosted the world's youngest known supereruption and numerous smaller explosive events. To assess ashfall hazard from future eruptions, we have simulated atmospheric ash dispersal using the Ash3d model. We consider five eruption scenarios spanning magma volumes of 0.1–500 km3 and investigate the main factors governing ash dispersal in modern atmospheric conditions. Our results are examined in the context of regional synoptic weather patterns (Kidson types) that provide a framework for assessing the variability of ashfall distribution in different wind fields. For the smallest eruptions (~0.1 km3 magma), ashfall thicknesses 〉1 cm are largely confined to the central North Island, with dispersal controlled by day‐to‐day weather and the dominance of westerly winds. With increasing eruptive volume (1–5 km3 magma), ashfall thicknesses 〉1 cm would likely reach major population centers throughout the North Island. Dispersal is less dependent on weather patterns as the formation of a radially‐expanding umbrella cloud forces ash upwind or crosswind, although strong stratospheric winds significantly restrict umbrella spreading. For large eruptions (50–500 km3 magma), powerful expansion of the umbrella cloud results in widespread ashfall at damaging thicknesses (〉10 cm) across most of the North Island and top of the South Island. Synoptic climatology may prove a useful additional technique for long‐term hazard planning at caldera volcanoes.
