    Notes: The ratio of the total characteristic to white radiation from a chromium target spectrographic X-ray tube has been determined by a new method at operating voltages ranging from 10 to 50 kV. The ratio values K are referred to the portion of the continuous spectrum extended from the Duane and Hunt limit λo to 2.5 Å A conversion coefficient has been tabulated to consider the continuous spectrum extended from λ to any arbitrary wavelength up to 6 Å. The method can be conveniently applied to targets other than chromium. It is only required to perform a simple fluorescence intensity measurement with a standard spectrometer on a conveniently prepared sample.

    Notes: A method is derived, using information theory and the maximum entropy principle, for treating deconvolution problems. The approach is particularly suitable for the determination of instrumental effects in spectral lines. The technique is illustrated by recourse to numerical examples. A comparison is made with alternative methods. Although realistic situations are restricted to the correction of x-ray diffraction profiles (instrumental width), the formalism applies for any spectral technique.

    Notes: A system based on the EDAX 9100 analyser was developed to obtain concentration profiles. Even though the system can be used on any type of flat specimens, in this study it was used for measuring the tungsten (W) concentration on rough spherical specimens of the W-γ-Al2O3 supported catalyst. The concentration gradient of the active component from the periphery to the centre of the particle is often an important factor in determining catalyst performance. The purpose was to determine the concentration profile of W along the radius of a sphere with a large enough number of experimental points to draw accurate information in a short time. In this way, a considerable number of impregnated spheres can be analysed, and average results can be calculated and taken as representative values of the population under study. The method developed provides highly satisfactory results, making it very useful for the systematic study of this type of specimen. Obviously, the use of this system can be extended to metallurgical and mineralogical specimens.

    Notes: Yeast exo-1,3-β-glucanases are secretable proteins whose function is basically trophic and may also be involved in cell wall glucan hydrolytic processes. Since fluorescein di(β-D-glucopyranoside) is a fluorogenic substrate detectable and quantifiable by flow cytometry, it was used for testing the ability of the EXG1 gene product of Saccharomyces cerevisiae and its homologous gene in Candida albicans to function as reporter genes. These open reading frames were coupled to different promoters in multicopy plasmids, and exoglucanase activity quantified at flow cytometry. Exoglucanases were found to be useful tools for the study of promoter regions in S. cerevisiae. This technique has the advantage over other reporter gene systems - such as β-galactosidase fusions - that it does not require permeabilization of yeast cells and therefore it allows the recovery of viable cells - by sorting - after flow cytometry analysis.

    Notes: Simple methods, based on the technique of flow cytometry, have been developed for the phenotypic characterization of yeast autolytic mutants and for the analysis of the formation and regeneration of the yeast protoplasts. The expression of lytic mutations determined uptake of the fluorescent dye propidium iodide, which could be carefully monitored by flow cytometry. Mixed populations of lysed and viable cells were precisely quantified and sorted, and the technique was also applied to demonstrate protection from lysis of mutant cells with cell wall defects, in the presence of osmotic stabilizers. Protoplast formation and regeneration was monitored by analysing relative cell size; this was facilitated by the preparation of homogeneous protoplast preparations. The technique of flow cytometry proved superior to other conventional methods for these types of study.
