    Notizen: A variety of diploid human fibroblast lines have been successfully grown to high densities (〉106 cell/ml) on recently developed microcarriers. Interferon induction using poly I·poly C and a superinduction procedure resulted in yields greater than 10,000 units/ml with one cell line. A direct comparison of microcarrier cultures to roller bottle cultures showed equivalent interferon yields on a per cell basis and some apparent differences relating to optimum inducer concentrations and kinetics of interferon accumulation.

    Notizen: An enzyme which catalyzes the synthesis of cephalexin fromD-α phenylglycinemethylester (PGM) and 7-amino-3-desacetoxy-cephalosporanic acid (7-ADCA) was prepared from Xanthomonas citri (IFO 3835) and partially purified 30-fold by ammonium sulfate fractionation, DEAE-cellulose, and Sepharose-4B column chromatography. The Km values for 7-ADCA, PGM, and cephalexin were determined as 11.1, 2.1, and 1.61 mM, respectively. The enzymatic cephalexin synthesis follows the reversible bi-uni reaction kinetics. The equilibrium constant is influenced by the initial mole ratios of 7-ADCA and PGM. The cephalexin hydrolysis is catalyzed by the same cephalexin synthesizing enzyme, but methanol does not participate in the hydrolytic reaction. The amount of enzyme in the reaction mixture affects the initial rate but does not influence the equilibrium product concentration. This cephalexin-synthesizing enzyme was immobilized onto several adsorbents. Among these, Kaolin and bentonite showed a higher retention of enzyme activity and stability for reuse. The immobilized-enzyme reaction kinetics were investigated and compared with those of the soluble enzyme. A rate expression for the enzymatic synthesis of cephalexin was derived. The results of computer simulation showed good agreement with the experimental results.

    Notizen: The influence of bulk-water bacterial cell concentration and specific growth rate history on bacterial adsorption rates to surfaces was investigated using response surface analysis. A pure culture of Pseudomonas sp. 224S was grown in a chemostat and pumped into a continuous flow reactor where the bacteria were exposed to clean, glass surfaces under turbulent flow conditions for a period of six hours. Adsorption rate decreased approximately linearly with increasing specific growth rate history. Glass surfaces became saturated with 224S at ca. 0.1% coverage and the resulting spatial pattern of the adsorbed cells deviated from random in the direction of uniformity.

    Notizen: Aerobic glucose metabolism by Pseudomonas aeruginosa biofilms at various calcium loading rates was investigated. The influence of calcium on specific growth rate, extracellular polymeric substance (EPS) formation rate, biofilm detachment rate, and biofilm calcium concentrations was determined. Calcium accumulated in the biofilm in proportion to the liquid phase concentration. Increasing calcium concentration increased the cohesiveness of the biofilm as indicated by a lower relative detachment rate. Specific activity in the biofilm was the same as that measured in a chemostat and was not influenced by changing calcium concentration. EPS formation rate in the biofilm was unaffected by calcium concentration but was higher than that observed in a chemostat.

    Notizen: One of the least understood processes affecting biofilm accumulation is detachment. Detachment is the removal of cells and cell products from an established biofilm and subsequent entrainment in the bulk liquid. The goal of this research was to determine the effects of shear stress and substrate loading rate on the rate of biofilm detachment.Monopopulation Pseudomonas aeruginosa and undefined mixed population biofilms were grown on glucose in a RotoTorque biofilm reactor. Three levels of shear stress and substrate loading rate were used to determine their effects on the rate of detachment. Suspended cell concentrations were monitored to determine detachment rates, while other variables were measured to determine their influence on the detachment rate. Results indicate that detachment rate is directly related to biofilm growth rate and that factors which limit growth rate will also limit detachment rate. No significant influence of shear on detachment rate was observed.A new kinetic expression that incorporates substrate utilization rate, yield, and biofilm thickness was compared to published detachment expressions and gives a better correlation of data obtained both in this research and from previous research projects, for both mono- and mixed-population biofilms. © John Wiley & Sons, Inc.

    Notizen: The cultural conditions for the production of enhanced formation of L-serine (up to 7 g/L) are described with the methylotrophic bacterium Pseudomonas 3 ab (DSM 672). The batch process is divided into three parts: (1) the biomass production phase, (2) substrate limitation period, and (3) L-serine accumulation phase. The initial specific production rate of qp = 0.1 g L-serine/g dry wt/h is based on the inhibition of the L-serine pathway. This is accomplished by high precursor concentrations (glycine) and a pH shift to pH 8.5. The enzymatic background is discussed. Furthermore, a coupling of the L-serine process with a L-tryptophan-producing process is demonstrated.

Variables monitored were substrate (glucose), biofilm cellular density, biofilm extracellular polymeric substance (EPS) density, and suspended cellular and EPS concentrations. A mathematical model developed to describe the system was compared to experimental data. Intrinsic yield and rate coefficients included in the model were obtained from suspended continuous culture studies of glucose metabolism by P. aeruginosa. Experimental data compared well with the mathematical model, suggesting that P. aeruginosa does not behave differently in steady-state biofilm cultures, where diffusional resistance is negligible, than in suspended cultures. This implies that kinetic and stoichiometric coefficients for P. aeruginosa derived in suspended continuous culture can be used to describe steady-state biofilm processes.

    Notizen: The ability of a number of Ustilago species, especially Ustilago maydis (DC.) Cda., to produce lysine and threonine was investigated. The organisms were grown in shake flasks or in 10-l. fermentors. Lysine and threonine were found to be excreted into the medium both in the free and bound form. The bound amino acids could be released by acid hydrolysis or by enzymes from autolyzed cells. The optimal conditions for the release by autolysis were, in the case of Ustilago maydis (DC.) Cda., pH 4.3 and 45°C. An enzyme that could liberate lysine from the bound form(s) occurring in the broth was extracted from cells of Ustilago maydis (DC.) Cda. It exhibited an apparent pH optimum near 4.0. The effect of pH and temperature during the growth phase on the yield of lysine and threonine was studied in 10-l. fermentations.

    Notizen: The ability of Ustilago maydis (DC.) Cda. to grow and to produce lysine and threonine was investigated in shake flask cultures. Growth and production of lysine and threonine increased markedly when aeration was increased. The optimal ratio of glucose to diammonium phosphate in the medium seemed to be approximately 10:1. Ammonium phosphate, ammonium sulfate, ammonium chloride, ammonium nitrate, sodium nitrate, urea, aspartic acid, glutamic acid, and glycine were readily used as nitrogen sources. Growth and amino acid production was poor on ammonium carbonate, ammonium acetate, ammonium citrate, trimethylamine, and betaine.U. maydis (DC.) Cda. was found to grow on a number of different carbohydrates. Besides D-glucose the organism could utilize D-mannose, D-galactose, D-fructose, L-arabinose, D-xylose, D-ribose, sucrose, maltose, and the polyalcoholes D-sorbitol, D-mannitol, and i-inositol. Pectin, dextrin, and corn starch treated with α-amylase could also be used but not untreated corn starch or lactose, cellobiose, D-sedoheptulose, glycerol, or D-glucosamine. The formation of lysine and threonine was better with disaccharides and hexoses than with pentoses.No specific effects on the formation of lysine and threonine could be observed from changes in the concentration of calcium, sodium, potassium, magnesium, zinc, iron, copper, manganese, cobalt, or molybdenum. The requirement of metal ions for growth is discussed.
