    Notes: Summary The disposition of diazepam has been studied in pregnant women at parturition. The plasma concentration of diazepam was monitored for at least 3 days in 18 women who received a single intravenous injection of 10 mg during the 10 h-period prior to delivery. Fourteen mothers had uneventful puerperia (Group I) and in 13 of these cases there was a pronounced postnatal increase in the plasma concentration of diazepam. The terminal phase half-life (t1/2) was significantly greater for Group I (mean = 65 h; range = 24–114 h) than for age-matched non-pregnant patients (mean = 29 h; range = 18–44 h from literature). The prolonged t1/2 appeared to be related to changes in the distribution of diazepam and not to a reduction in hepatic elimination since the total plasma clearance (Cltp) in these 14 pregnant patients (mean = 28 ml/min; range = 18–43 ml/min) was not reduced compared to that reported for non-pregnant controls (mean = 30 ml/min; range = 22–45 ml/min). Four mothers underwent postnatal surgery for tubal ligation (Group II) and the plasma concentration-time profiles for this group did not show the same postnatal phenomenon as did the profiles obtained for Group I. The t1/2 for Group II was shorter (mean = 31 h; range = 24–37 h) than for Group I and similar to that for the non-pregnant controls. The Cltp for Group II was greater (mean = 56 ml/min; range = 48–63 ml/min) than for both Group I and non-pregnant controls. These results suggest that delivery alters the disposition of diazepam and is generally associated with a postnatal re-distribution of diazepam into the systemic circulation. The blood/plasma concentration ratio was determined in 9 patients (mean = 0.62; range = 0.54–0.77). There was no difference in the total blood clearance between the pregnant patients of Group I and the non-pregnant controls. In most cases the umbilical venous plasma concentration (Cpuv) of diazepam was greater than the peripheral maternal venous plasma concentration (Cpmv) at delivery. The foetus appears to constitute a slowly equilibrating tissue-group in which diazepam does not reach equilibrium with the maternal systemic circulation for at least 5–10 h at which time the diazepam concentration in maternal and foetal plasma is similar.

    Notes: Summary Somatostatinergic nerves in the spinal cord of the monkey were investigated utilizing immunohistochemistry with various antibodies against synthetic somatostatin. In contrast to earlier investigations, it is shown that somatostatinergic nerve endings occur in most of the areas of the grey matter of the spinal cord. The somatostatinergic axons are, however, characteristically distributed in three main regions: (1) Densely-packed endings are seen in lamina II of the substantia gelatinosa, forming a crescent-shaped pattern in the columna dorsalis. Somatostatin immunoreactivity is also seen in lamina I and in the Lissauer tract. (2) A fine network of fibers is observed around the central canal; the endings are concentrated on special cell bodies. Some single perikarya are also stained in this region. (3) A loose network of single fibers is found ending on perikarya of the columna lateralis or ventralis. The perikarya of the nerve axons, with the exception of those terminating in the columna dorsalis, have as yet not been identified. In order to better understand the somatostatinergic system of the spinal cord, these newly-detected somatostatinergic nerves must be studied and their exact pathways analyzed.

    Notes: Summary Equine pancreas was investigated with immunohistochemical methods to study the distribution of endocrine cells immunoreactive to antiinsulin, anti-glucagon, and anti-somatostatin. A-cells demonstrable by antiglucagon are located in the center of Langerhans islets and frequently in the duct epithelium. Few A-cells are seen associated to acini. Anti-insulin reactive B-cells form a large zone around the center of the Langerhans islets in which some B-cells lie between exocrine cells and others, although few, are located in the duct epithelium. D-cells stained with anti-somatostatin serum form a discontinuous outermost zone around the Langerhans islets. In some islets the D-cells are also observed among the B-cells or between the border of A- and B-cells. Single D-cells are seen in the duct epithelium or between acinar cells. In younger horses, endocrine cells are more frequently associated in bulges of the duct system. The histotopographic relation between these endocrine cell types is discussed with respect to its functional significance.

    Notes: Summary The Merkel cells from sinus hair follicles of rats were investigated by immunohistochemistry using different antisera against neuropeptides and gastroenteropancreatic (GEP)-hormones. For the first time it has been demonstrated that Merkel cells exhibit an immunoreactivity towards metenkephalin (methionine-enkephalin). The met-enkephalin immunoreactivity was restricted to Merkel cells and was not found in associated nerve axons or terminals. Denervation of Merkel cells did not affect the met-enkephalin immunoreactivity. Antisera against leu-enkephalin (leucine-enkephalin) and other polypeptides did not produce an immunoreaction. The demonstration of met-enkephalin-like immunoreactivity supports the concept that the Merkel cell is a member of the paraneuronal system and a potential neuroreceptor cell.

    Notes: The electronic properties of a variety of methylated isoalloxazine derivatives have been investigated with absorption and fluorescece techniques (Table 1). The spectral effects of methyl substitution at different positions in the isoalloxazine ring are interpreted in terms of different bond orders between the C-atoms of the methyl groups and of the aromatic ring. Methyl groups at C(6) or C(9) induce quenching of the fluorescence and shortening of the lifetime whereas a methyl group at C(7) has an opposite effect. The quenching effect might be due to strain in the molecular framework caused by the methyl groups.The lowest energy levels of the isoalloxazine cations are shifted to higher energy as compared to the neutral isoalloxazines. Apart from these shifts the trends in spectral behaviour upon methylation are similar to those observed for the neutral isoalloxazine (cf. Table 2). The fluorescence of the isoalloxazine cations is strongly quenched in auqeous solution and their lifetime much shortened. These observations indicated that there is substantial interaction between the charged fluorophore and the water dipoles and that this interaction produces efficient radiationless deactivation of the first excited singlet state.

    Notes: Unbalanced growth has been studied in HeLa cell cultures maintained in perpetual division synchrony by periodic inhibition of DNA synthesis. Observation of eight independent cellular parameters indicates that once synchrony has been established the degree of unbalanced growth in later cycles is not as great as is observed with the conventional “double-block” technique. This significant diminution of unbalanced growth appears to be related to the significantly shorter periods of inhibition of DNA synthesis used in the resynchronization process.

    Notes: A variety of diploid human fibroblast lines have been successfully grown to high densities (〉106 cell/ml) on recently developed microcarriers. Interferon induction using poly I·poly C and a superinduction procedure resulted in yields greater than 10,000 units/ml with one cell line. A direct comparison of microcarrier cultures to roller bottle cultures showed equivalent interferon yields on a per cell basis and some apparent differences relating to optimum inducer concentrations and kinetics of interferon accumulation.

    Notes: Six enzymes associated with the activities of the nucleus (thymidine kinase), mitochondria (succinate dehydrogenase), lysosomes (acid phosphatase), peroxisomes (catalase), (cytosol lactate dehydrogenase), and the intermembranal mitochondrial space (alkaline DNase) were assayed at 2 hr intervals over the division cycle of repetitively resynchronized HeLa cells. The results indicated a high degree of reproductibility for cells synchronized by the method of perpetual resynchronization and may be of direct use to those interested in subcellular organellogenesis.

    Description / Table of Contents: The Use of Titanium Castings to Produce a Complex Shaped Intermediate Casing of RB 199.This paper describes the present position on a highly stressed casing and its functions in the RB 199-34 R engine. When the parts of the casing are made from solid forged titanium billets, up to 93% of the material used is machined away. For this reason, the most complicated parts of the casing were procured as castings and welded together by TIG welding. The paper describes the present quality standard of the titanium castings. The tests on the materials and their mechanical properties show that the castings are inferior to the forgings only with respect to their HCF behaviour and their elongation at rupture. Macroscopic flaws are not entirely inevitable but can be detected by X-ray tests. The future prospects for titanium castings are considered favourable.
    Notes: Der Bericht beschreibt die Lage eines hochbeanspruchten Gehäuses und dessen Funktionen im Triebwerk RB 199-34 R. Bei der Herstellung der Einzelteile des Gehäuses aus massiv geschmiedeten Titanblöcken wird bis zu 93% des eingesetzten Materials zerspant. Die kompliziertesten Partien des Gehäuses wurden deshalb als Gußteil bezogen und mittels WIG-Schweißung verbunden. Der Bericht beschreibt den heutigen Qualitätsstand der Titangußteile. Die Ergebnisse der Werkstoff- und Festigkeitsuntersuchungen weisen nur im HCF-Verhalten und der Bruchdehnung Nachteile des Gusses gegenüber dem geschmiedeten Material aus. Makroskopische Fehler sind nicht vollständing vermeidbar, aber durch Röntgenprüfung nachzuweisen. Die künftigen Aussichten des Titangusses werden positiv beurteilt.
