    Notes: Fraction I protein was extracted from fresh alfalfa (Medicago sativa) leaves and purified by ammonium sulfate fractionation, DEAE-cellulose and Sephadex G-200 chromatography. The isolate possessed a specific activity for ribulosebisphosphate carboxylase EC 4.1.1.39) of 1.01 units/mg protein. An approximate molecular weight of 573,000 was determined from values of So20,w (18.7) and Do20,w (2.97). The protein contained 26 disulfide and 37 sulfhydryl groups—only one of which was titratable in the undenatured protein complex. It migrated as a single boundry by Tiselius electrophoresis and as a dense zone in polyacrylamide gels (disc). In the presence of SDS, two principal subunits of ∼52,000 and ∼12,500 daltons were revealed. Based on the FAO/WHO (1973) provisional pattern for essential amino acids, the protein was slightly deficient in sulfur-containing residues.

    Notes: Abstract American flagfish (Jordanella floridae) were exposed to intermittent cyanide concentrations ranging from 0.065 to 0.15 mg/L hydrogen cyanide to determine the effects upon the embryonic, juvenile, and adult stages of development along with hatching success and fry survival of the subsequent F1 generation. The most significant effect was observed following embryonic exposure to levels as low as 0.075 mg/L hydrogen cyanide from fertilization to hatching. Hatching success was reduced to 37% when compared with controls while egg production among those embryos which successfully completed hatching and reached sexual maturity was subsequently reduced by 30%. A 5-day exposure to levels as low as 0.065 mg/L during the embryonic and juvenile stage produced a 40% reduction in egg production at sexual maturity and a further 33% reduction in the hatching success of the F1 generation. Exposure during the embryonic and adult stage produced a 27% reduction in fecundity. The results also suggest that exposure during embryonic development is responsible for the identical but shortened estrous cycle and delayed sexual maturity observed following embryonic (Group A), embryonic and juvenile (Group B) and embryonic and adult (Group C) exposure to hydrogen cyanide.

    Notes: Summary Duck muscle spindles show a large outer capsule enclosing a number of intrafusal muscle fibres which are individually encircled by an inner capsule. The outer capsule consists of a multilayer of 4–6 flattened cells with elongated nuclei, and are usually lined by a basement membrane. They resemble and are linked with perineural cells of the nerve bundle that innervates the spindle. There is overlapping and interdigitation between branching processes of these cells. Some apposing surfaces develop junctional complexes in the form of hemi-desmosomes and desmosomes. The cytoplasmic content shows numerous pinocytotic vesicles, some polyribosomes, lamellated cisternae of rough endoplasmic reticulum and microfilaments. The inner capsule consists of cells showing, at the mid-equatorial region, a large ovoid nucleus and extensive branching of thin and long processes that overlap, forming numerous layers around each intrafusal muscle fibre. Bundles of collagen fibrils in a crescentshaped configuration occur circumferentially between the multilayer of processes and the muscle fibre. Some linkages between the inner and the outer capsule cell processes provide a network that subdivides the large periaxial space into compartments. There is no basement material lining the inner capsule cells and the processes. Some cytoplasmic area occurs around the ovoid nucleus and the cytoplasm varies in density, showing some dilated short profiles of rough endoplasmic reticulum, a few pinocytotic vesicles and microfilaments.

    Notes: A new computational scheme, named the ‘Integrated Load Increment’ method (ILI), has been developed to handle elasto-plastic analysis of solids by the finite element variational technique. This scheme allows considerably larger loading steps to be applied to the structure without deviating from its nonlinear, load-deformation relationship. It has been shown that these relationships can be evaluated by integrating the nonlinear constitutive equations within each load increment under certain conditions. A numerical example on a pressurized thick-walled cylinder indicates that a 240 per cent improvement in computational efficiency and substantial reduction of loading steps over the conventional load incremental method can be achieved by the ILI technique.
