    Notes: Summary When the active sodium-potassium pump (Na−K-ATPase) of shark rectal glands is blocked by ouabain, the concentration of intracellular ions changes in the direction of equilibrium with extracellular fluid. These changes were examined when isolated perfused glands were in the basal state and also when they were stimulated to secrete with cAMP and theophylline, to see whether stimulation affected the passive movement of sodium, potassium and chloride across cell membranes. In basal glands 10−4M ouabain induced an increase of 30 meq/l in intracellular [Na+] and a decrease in intracellular [K+] of about 50 meq/l after 30 min, while intracellular [Cl−] was unchanged. In stimulated glands, these movements were exaggerated. The increase in intracellular [Na+] averaged 112 meq/l, and the decrease in intracellular [K+], 96 meq/l (P〈0.01), while mean intracellular [Cl−] rose by 80 meq/l. Furosemide, 10−4M, partially reversed the accelerated changes in intracellular electrolytes seen after ouabain was added to stimulated glands. These results are consistent with an action of cAMP upon a ouabaininsensitive cotransport of sodium, potassium and chloride in the rectal gland, analogous to that described in avian erythrocytes.

    Notes: Summary Available evidence indicates that hypercalcemia in pulmonary tuberculosis results from increases in circulating 1α,25-dihydroxyvitamin D [1α,25(OH)2D]. To further characterize vitamin D metabolism in this disorder, the effects of vitamin D, 100,000 units a day for 4 days, were compared in 25 normal subjects and 11 patients with active pulmonary tuberculosis who were normocalcemic and had not had hypercalcemia. Serum calcium, phosphorus, 25-hydroxyvitamin D (25-OHD) and 1α,25(OH)2D were measured. Whereas vitamin D increased mean serum 25-OHD from 20±2 (±SE) to 40±5 ng/ml (P〈0.001) and did not change mean serum 1α,25(OH)2D in the normals (33±2 vs. 31±2 pg/ml), it increased mean serum 25-OHD from 21±4 to 55±13 ng/ml (P〈0.05) and mean serum 1α,25(OH)2D from 28±2 to 35±3 pg/ml (P〈0.05) in the patients. Serum calcium was normal and remained within the normal range in all subjects and patients. The findings indicate that there is a modest but significant abnormality in the regulation of circulating 1α,25(OH)2D in normocalcemic patients with pulmonary tuberculosis. The results are similar to those previously reported by us in normocalcemic patients with sarcoidosis.

    Notes: Abstract Mutants of the EB virus-transformed cell line T5-1 (HLA-Al, 2; B8, 27), bearing well-characterized alterations in HLA-A2 antigen expression and unable to bind the HLA-A2-specific monoclonal antibody 13137.2, have been tested for their susceptibility to EB virus-specific cytolysis using effector T-cell preparations functionally restricted through relevant HLA antigens. Initial experiments first confirmed that the parent line T5-1 was susceptible to cytolysis by both “common” A2-restricted and 1327-restricted effector cells. While those T5-1 mutants with little or no surface A2 expression were not lysed by A2-restricted effectors, those targets with quantitatively normal expression of mutant A2 molecules were as susceptible to A2-restricted lysis as the parent line itself. In contrast, all the T5-1 mutant lines were susceptible to B27-restricted cytolysis. The results demonstrate that experimentally induced mutations of HLA-A2 antigen structure, affecting a serologically defined site on the molecule, can occur without altering that same molecule's expression of the T cell-restricting determinant(s). Such experimentally induced mutations are quite different from the naturally occurring “variant” A2 antigens which are present within the serologically defined A2 antigen group and which show changes at the T cell-restricting site.

    Notes: Abstract We consider one parameter analytic hamiltonian perturbations of the geodesic flows on surfaces of constant negative curvature. We find two different necessary and sufficient conditions for the canonical equivalence of the perturbed flows and the non-perturbed ones. One condition says that the “Hamilton-Jacobi equation” (introduced in this work) for the conjugation problem should admit a solution as a formal power series (not necessarily convergent) in the perturbation parameter. The alternative condition is based on the identification of a complete set of invariants for the canonical conjugation problem. The relation with the similar problems arising in the KAM theory of the perturbations of quasi periodic hamiltonian motions is briefly discussed. As a byproduct of our analysis we obtain some results on the Livscic, Guillemin, Kazhdan equation and on the Fourier series for the SL(2, ℝ) group. We also prove that the analytic functions on the phase space for the geodesic flow of unit speed have a mixing property (with respect to the geodesic flow and to the invariant volume measure) which is exponential with a universal exponent, independent on the particular function, equal to the curvature of the surface divided by 2. This result is contrasted with the slow mixing rates that the same functions show under the horocyclic flow: in this case we find that the decay rate is the inverse of the time (“up to logarithms”).

    Notes: Abstract Cowey and Rolls have shown that the magnification factor with which the retina is imaged onto the striate cortex is proportional to visual acuity. Schwartz has used this to derive how visual peripheral acuity in the human varies with distance from the fovea, in good agreement with experiment. The same reasoning applied to the map of the lower half of the field of view as imaged onto the striate cortex of the cat indicates that the cat fixating a point up to about 100 cm in front of him sees the foreground portion of the horizontal surface on which he is standing approximately uniformly blurred.

    Notes: Abstract Evidence is presented here for a close linkage between Akv-2, an ecotropic provirus found uniquely on chromosome 16 of AKR/N mice, and the immunoglobulin λ 1 light chain locus, Igl-1. No recombinants between the Igl-1 locus and Akv-2 were found by Southern blot analysis of DNA obtained from progeny of the backcross of (AKR/N × SJL/J)F1 to SJL/J, indicating that these genes map within 5.9 cM of each other. A probe specific for the flanking sequence of Akv-2 was used to detect the provirus, while one specific for the IgI-1 constant region was used to determine which allele of the structural gene was expressed in the backcross mice. The constant region of Igl-1 differs between AKR/N and SJL/J with respect to a site for the restriction endonuclease KpnI. This backcross was also used to seek recombinants between the regulatory, Igl-1r, and structural, Igl-1, loci of the immunoglobulin light chain locus, since the existence of such recombinants would prove that these loci are distinct. Since only parental types were recovered in the offspring, the structural and regulatory loci are no more than 2.3 cM apart, and the implications of this finding are discussed.

    Notes: Summary Enzymatic activity was investigated in metal-binding proteins from rat epidermal cells. Tris-HCl buffer soluble and KSCN solubilized proteins were extracted stepwise from granular and cornified cells of 2-day old rat epidermis. Each extract was separately applied to a Cu2+ or Zn2− chelate Sepharose 6B column and the proteins were eluted with buffers of different pHs and finally with EDTA solution. Metal chelate-binding proteins were found in both soluble and solubilized proteins but there was a larger amount in the latter. Affinity of the proteins to bind with Cu2+ chelate was greater than that with Zn2+ chelate. In Tris-HCl buffer extract, histidase activity was detected in Cu2+ chelate-binding proteins, but not in Zn2+ chelate-binding proteins. Acid phosphatase, cysteine proteinase, dipeptidase, cathepsin D, β-galactosidase, gelatin hydrolase, and superoxide dismutase did not bind to metal chelates although these enzymes, except acid phosphatase, were inhibited by Cu2+, but not by Zn2+. In contrast, KSCN solubilized metal chelate-binding proteins showed plasminogen activator, acid phosphatase, and gelatin and casein hydrolases while histone hydrolase did not bind to either chelate column. Since metal-binding proteins in rat epidermal cells have been shown previously to be histidine- and cysteine-rich proteins concentrated in keratohyalin granules, interaction of metals and the structural proteins with certain enzymes may be involved in the regulation of epidermal cell functions.

    Notes: Abstract In recent times major changes have occurred in extensive areas of Lapland inhabited by the reindeer-herding Lapps of northern Fennoscandia (Norway, Sweden, Finland, and the Kola Peninsula in the Soviet Union). Their customary way of life, delicately attuned to the ecological conditions of the arctic and subarctic areas, is buffetet between the environment-adapted life of the timeless tundra, forests, and swapms and the modern industrialized economy of northern Europe. The Lapps in the northern part of the reindeer-herding region have maintained some of the old nomadic ways and are closely tied to the habits of the reindeer, providing a simple but excellent example of the way that man can use nature without destroying it. The continued practice of reindeer ecology may serve as the catalyst that will keep the occupation of reindeer husbandry alive and make possible an orderly fusion with modern trends.
