    Notizen: Several axenic strains of pathogenic and nonpathogenic Entamoeba histolytica were tested for their capacity to digest native radioactive type I collagen gels and to produce liver abscesses when injected into the liver of newborn hamsters. The results demonstrate that the pathogenic strains of amebas (HM1:IMSS, HM3:IMSS, HM38:IMSS, and HK9) have a collagenolytic activity that closely correlates with their in vivo capacity to produce liver lesions. The nonpathogenic isolate (Laredo) did not show collagenolytic activity and failed to produce lesions in the liver of newborn hamsters. The results also demonstrate that type I collagen obtained from rodents and cats is degraded less by amebic collagenase than is bovine collagen, which is similar to human collagen. These findings suggest that species susceptibility to invasive infection may depend, among other factors, on the characteristics of the extracellular components of host tissues.

    Notizen: A numerical model was developed to describe the bioremediation of hydrocarbons in ground water aquifers considering aerobic degradation. The model solves the independent transport of three solutes (oxygen, hydrocarbons, and microorganisms) in ground water flow using the method of characteristics. Interactions between the three solutes, in which oxygen and hydrocarbons are consumed by microorganisms, are represented by Monod kinetics, solved using a Runge-Kutta method. Model simulations showed good correlation as compared with results of soil column experiments. The model was used to estimate the time needed to remediate the columns, which varied from one to two years.

    Notizen: The present study was undertaken to identify and characterize clones whose expression increase during Cicer arietinum epicotyl growth. Two cDNAs encoding two different plant metallothionein (MT)-like proteins have been isolated from a cDNA library from epicotyls of Cicer arietinum L. cv. Castellana. The CanMT-1 deduced protein appears to have the typical structure of type 1 MT where all Cys residues are in Cys-X-Cys motifs, while the CanMT-2 has the typical structure of type 2 MT having Cys-Cys and Cys-X-X-Cys motifs within the N-terminal domain. Both chickpea CanMTs are up-regulated during epicotyl growth, showing increased expression in mature tissues, mostly CanMT-1, which is undetectable in young epicotyls. Accordingly, stem of chickpea plants displayed the highest level of CanMT-1 expression in the basal internode, with reduced growth, decreasing towards the apex. Osmotic stress by PEG, which inhibited growth, and ABA treatment induced the expression of MT-like genes, which points to a relationship between chickpea MTs and ABA-mediated stress response. Unlike CanMT-2, CanMT-1 is induced in chickpea epicotyls by cadmium indicating a different function for both clones. We conclude that these MT-like proteins, in particular CanMT-1, are regulated by the developmental stage and may participate in cell maturation process.

    Notizen: Dengue viruses are arthropod-borne, single-stranded RNA viruses. Aëdes aegypti and Aëdes albopictus are the principal vectors. In order to understand the molecular basis of dengue virus infections we explored the biochemical identity of dengue-2 (DEN-2) virus receptors in the Aëdes albopictus-derived cell line C6/36. We show here that DEN-2 interacts with two major polypeptides of 80 and 67 kDa. Polyclonal anti-C6/36 membrane antibodies block DEN-2 binding to intact C6/36 monolayers as well as to membrane extracts. Our results strongly suggest that the identified polypeptides are part of the DEN-2 receptors.

    Notizen: The interaction of Entamoeba histolytica trophozoites with collagen induces the synthesis and release of electron-dense granules containing a collagenase activity that is an important factor in the pathogenicity of the parasite. The binding is thought to be mediated by an ‘integrin-like’ collagen receptor. In the signal transduction mechanisms activated by collagen, pp125FAK and p42MAPK are involved. Using immunoprecipitation assays coupled to Western blot analysis, we demonstrate here the collagen-dependent association of paxillin and Src with pp125FAK. Furthermore, collagen induces a time-dependent increase in the DNA binding activity of the activator protein 1, which is well correlated with an increase in Fos expression. Our results suggest that a stimulus-transcription coupling triggered by collagen in E. histolytica trophozoites might activate or repress genes involved in tissue invasiveness.

    Notizen: The proline-rich proteins (PRPs) form a class of plant cell wall structural proteins. The expression of PRP genes is organ- and tissue-specific and it is induced in response to wounding, infection and other stress conditions, but the role of these proteins in the cell wall during development and stress has not yet been established. Our objective was the characterization of a cDNA clone, termed CanPRP, isolated from a cDNA library of growing epicotyls of Cicer arietinum, and the study of the relationship between its expression and cell wall modifications during growth. CanPRP differential expression was analysed by northern blot through epicotyl growth, under different growth conditions and in different organs of chickpea. CanPRP encodes a putative PRP that contains 15 alternating pentapeptide repeats: Pro-Pro-Val-Tyr-Lys and Pro-Pro-Val-Glu-Lys, typical features of cell wall PRPs. The levels of PRP transcripts are almost undetectable in young epicotyls and increase during epicotyl growth, with the highest levels of expression found once growth begins to cease and the cell wall starts to strengthen. Accordingly, the stem of chickpea displays the highest level of expression in the basal internode, with reduced growth, decreasing towards the apex. Expression of the CanPRP clone was induced by osmotic and saline stress which inhibited growth. We conclude that the protein deduced from the CanPRP clone participates in the cell wall-hardening process in either the stress-mediated inhibition of growth or when cell growth ceases as development takes place.

    Notizen: Abstract Five different interferon preparations were compared for their antiviral activity against Herpes simplex virus type 1 (HSV-1) and several RNA viruses. The interferons used were: interferon α from human buffy coats, interferon β from human fibroblasts, interferon γ from human lymphocytes after stimulation with phytohemagglutinin (PHA), lymphoblastoid interferon from Namalva cells IFN-α (Ly) and cloned α2 interferon produced by Escherichia coli containing the human gene for interferon α2. All preparations were able to protect monolayers of HeLa cells against HSV-1 infection when low multiplicities were used. The five IFN preparations were also tested against encephalomyocarditis (EMC) virus, poliovirus and vesicular stomatitis virus (VSV).

    Notizen: Abstract. Capsular polysaccharides are important virulence factors both in Gram-positive and Gram-negative bacteria. A similar cluster organization of the genes involved in the synthesis of bacterial exopolysaccharides has been postulated in both cases, suggesting that these clusters evolved by module assembly. Horizontal gene transfer has been postulated to explain the polymorphism found in these cellular polymers. The cap1K and cap3A genes coding for the pneumococcal type 1 and type 3 UDP-glucose dehydrogenases, respectively, have been compared with other UDP-sugar dehydrogenases. We have observed that the evolutionary distance between Cap1K and Cap3A is approximately equal to that found between Cap1K (or Cap3A) and other UDP-GlcDH of families evolutionarily distant like KfiD, the dehydrogenase from Escherichia coli K5. On the basis of comparisons of G + C content, patterns of synonymous and nonsynonymous substitutions, dinucleotide frequencies, and codon usage bias, we conclude that the kfiD gene has been introduced into E. coli from an exogenous source, probably from a streptococcal species.
