    Notes: Extensive examination of large numbers of spatially orientated thin sections of orientated samples from orogens of all ages around the world has demonstrated that porphyroblasts do not rotate relative to geographical coordinates during highly non-coaxial ductile deformation of the matrix subsequent to their growth. This has been demonstrated for all tectonic environments so far investigated. The work also has provided new insights and data on metamorphic, structural and tectonic processes including: (1) the intimate control of deformation partitioning on metamorphic reactions; (2) solutions to the lack of correlation between lineations that indicate the direction of movement within thrusts and shear zones, and relative plate motion; and (3) a possible technique for determining the direction of relative plate motion that caused orogenesis in ancient orogens.

    Notes: The aim of the study was to determine whether C5 deficiency in the mouse is associated with resistance to the development of secondary amyloidosis. Chronic inflammation was induced in the F2 progeny, derived from matings between amyloid-susceptible and amyloid-resistance mice, by daily injections of azocasein for thirty days. Using a restriction fragment length polymorphism generated by digestion of genomic DNA with the restriction enzyme HindIII, C5 sufficient and deficient DNA can be clearly differentiated. Eight mice were found to be C5 sufficient, 32 were heterozygotes and 14 were found to be C5 deficient. Grading of the splenic amyloid load from negative to 4+ was performed after staining tissue squashes with Congo red and viewing them under a polarizing microscope. Seventeen mice were noted to have negative to trace, 18 had moderate (1+-2+) and 19 had heavy (3 H+-4+) amyloid deposition. There was no correlation between splenic amyloid load and C5 deficiency. Based on these results it is clear that C5 deficiency and resistance to secondary amyloidosis are not associated.

    Notes: In a first course in mathematical statistics or categorical data analysis, the maximum likelihood estimators of the parameters of the multinomial distribution are often “derived” using calculus. However, the important step of verifying that the resulting estimators indeed maximize the likelihood is omitted or done incorrectly. In this paper, two simple methods of obtaining the maximum likelihood estimates are presented.

    Notes: The unit-copy P1 plasmid depends for stability on a plasmld-encoded partition region called par, consisting of the parA and parB genes and the parS site. ParA is absolutely required for partition, but its partition-critical role is not known. Purified ParA protein is shown to possess an ATPase activity in vitro which is specifically stimulated by purified ParB protein and by DNA. ParA is responsible for regulation of expression of parA and parB, and purified ParA has an ATP-dependent, site-specific DNA binding activity which recognizes a sequence that overlaps the parA promoter. The rote of the ATP-dependence of the binding activity, as well as other possible functions of the ATPase activity in partition, is discussed.

    Notes: The amdR (intA) regulatory gene of Aspergillus nidulans encodes a 765-amino-acid polypeptide which determines the omega-amino acid induction of at least five structural genes. The AmdR polypeptide contains a potential Zn(ll)2Cys6 DNA-binding motif which has been shown to be present in the N-terminal region of a large number of fungal activator proteins. In vitro mutagenesis of the fourth cysteine of this motif abolishes AmdR function as shown by loss of complementation of an amdR− mutation and by the AmdR− phenotype of a mutant gene replacement strain. Studies using constructs in which the proposed AmdR DNA-binding motif is replaced with that from another activator, FacB, shows that induction is independent of DNA-binding specificity and that sequences in the C-terminal region of AmdR are activation domains. Sequencing of several amdR mutant alleles which affect activation and/or induction, together with studies of deletion constructs indicate that changes in the conformation of the protein determines its activity and that this is modulated by inducers.

    Notes: The broad-host-range plasmid RK2 has been shown to encode several proteins important for its maintenance within bacterial populations of a number of Gram-negative bacteria. Their genes are organized into two operons: parCBA and parD. These operons have been proposed to be transcribed from two divergent promoters, p-parCBA and p-parD, located within a sequence of approximately 150 bases. In this report we identify and characterize the sequences required for regulated transcription from these promoters in Escherichia coli, Agrobacterium tumefaciens and Pseudomonas aeruginosa. Both of these promoters are repressed by their own gene products in the same manner in all three bacteria tested, with ParA functioning as the primary repressor of p-parCBA and ParD functioning as the repressor of p-parD. The binding regions of these proteins were determined through deletion analyses, DNA mobility shift assays, and an examination of the effect of mutations in this region. Based on these observations, the ParA protein appears to bind to either two inverted repeat or two direct repeat sequences, one downstream from the transcriptional initiation site and the other upstream of the p-parCBA -35 box. The ParD protein appears to bind to one inverted repeat sequence, located between the -35 and -10 boxes of p-parD.

    Notes: Reproductive performance of albino and pigmented channel catfish (Ictalurus punctarus) females was compared when spawning was induced with HCG (human chorionic gonadotropin) or Ovaprim/〉 (domperidone and [d-Ala6, Pro9 NEt]-luteinizing hormone-releasing hormone). In 1985, only HCG was used to induce spawning. Albino females required more injections, had a longer latency period from time of first injection to spawning, and had lower spawning success (30%) than pigmented females (77%). No differences were observed between albino and pigmented females for time-of-day of spawning, number of eggs obtained by manual stripping, and number of eggs/kg body weight. In 1988, HCG, Ovaprim, and saline (control) were used to induce spawning. HCG induced spawning in 80% of albino and 71% of pigmented females. Ovaprim induced 67% of pigmented females to spawn, but only 2 of 5 albino females spawned. One of four pigmented females injected with saline spawned. Albino females yielded 40–50% fewer eggs/kg body weight than pigmented females. Other performance characteristics for albino and pigmented females were similar whether injected with HCG or Ovaprim. High temperatures encountered during the 1985 spawning season my have had a differential effect on albino and pigmented fish and may account for some differences in spawning success.

    Notes: Two experiments were conducted to evaluate the dietary iron requirement of juvenile Penaeus vunnamei. Prior to initiation of 28 day (Experiment I) and 35 day (Experiment II) feeding trials, 17–20day-old postlarvae were fed the basal diet lacking iron supplementation but containing 12 mg Fe/kg for seven days. After conditioning, juvenile shrimp (mean weight: 0.038 g Experiment I, 0.047 g Experiment II) were fed one of four diets supplemented with 0, 20, 40 or 80 mg Fe/kg from ferrous sulfate heptahydrate. Final weight gain of shrimp ranged from 1.03 to 1.28 g in Experiment I and 1.62 to 1.94 g in Experiment II. Although percent weight gain was in excess of 2700% in Experiment I and 3400% in Experiment II, there were no significant differences in growth or survival of shrimp fed the different levels of iron. Iron levels of the hepatopancreas and carapace increased linearly with dietary iron; however, there were no significant differences in the iron content of muscle tissue. Based on these results, the iron content of the basal diet appeared to satisfy physiological needs of the shrimp. Therefore, practical diets containing protein sources of animal origin which are rich in iron should not require iron supplementation.

    Notes: The killifish Rivulus marmoratus is the only known selfing hermaphroditic vertebrate, and males of the species are usually exceedingly rare or non-existent in nature. Collections on several Belize cays in 1988 and 1989 yielded 13.5–24% males. Factors responsible for this unusually high proportion of males are currently not understood. Likewise, the biological significance of males in populations which otherwise consist of selfing hermaphrodites (with internal fertilization) is problematic and awaits further study.

    Notes: Diets containing a complete mineral premix or mineral premixes from which Ca, P, Na, CI, K, Mg, Mn, Fe, Zn, Cu, I, Se, or Cr were individually deleted were fed to juvenile Penaeus vunnamei (mean initial weight 79.5 mg) over a 4-week experimental period. Final mean percent weight gain ranged from 1,472 (Cu-free mineral premix) to 1,991% (Na-free mineral premix) with an average of 1,812% for all treatments. Survival during the first two week period and the second two week period was greater than 92% and 88%, respectively. There were no significant differences observed among treatments for growth and survival data.Upon termination of the growth trial, the hepatopancreas and carapace were dissected from the shrimp and evaluated for ash, Ca, P, Na, K, Mg, Mn, Fe, Zn and Cu content. Of the minerals analyzed, the individual deletion of Mg, Mn, Fe, Zn and Cu from the mineral supplement resulted in reduced tissue mineralization in P. vannamei whereas, the deletion of Ca and P from the mineral mix did not produce significant responses. Deletion of Se from the mineral premix resulted in a significant increase in the ash content of the carapace. Based on the information provided by this and other studies, further investigation of the dietary requirements of P. vannamei for Ca, P, Fe, Mg, Mn, Zn, Cu and Se is recommended.
