    Notizen: The three nodD genes of a strain of Rhizobium leguminosarum biovar phaseoli were cloned to study their effects on transcription of themselves and of the nodC genes of biovars phaseoli and viciae. Efficient transcription of nodD1 required nodD1 and was enhanced by exposure of the cells to bean exudate consistent with the presence of a nod-box preceding the nolE-nodD1 operon. Transcription of nodD2 and nodDZ was constitutive. nodC of R. leguminosarum biovar phaseoli was activated by each of the nodD genes of that biovar in the absence of inducers but expression was enhanced in cells grown with bean exudate or the flavonoids genistein or naringenin. A mutant of nodD2, tacking 60bp at its 3’end, activated nodC in the presence of inducer, but was defective in regulating certain of the nodD genes. The nodC gene of R. leguminosarum biovar viciae responded differently to the various nodD genes of R. leguminosarum biovar phaseoli than did the nodC of the latter biovar.

    Notizen: Streptomyces coelicolor produces spores whose development of a grey colour requires the activity of the whiE locus. The cloned whiE locus was identified after mobilization into a whiE mutant of a library of S. coelicolor DNA inserted into a transmissible plasmid vector. The whiE region of the cloned DNA was localized both by subcloning and by mutagenesis of the cloned DNA with the Streptomyces transposon Tn4560. Nucleotide sequencing of this region revealed seven open reading frames, of which six show homo-logy at the level of deduced gene products with genes involved in the synthesis of polyketide antibiotics. A previously described S. coelicolor DNA segment encoding biosynthesis of a brown pigment (Horinouchi and Beppu, 1985) corresponds to the cloned whiE DNA. It is proposed that whiE is normally expressed only in the aerial hyphae, where the biosynthetic product is responsible for spore colour.

    Notizen: In a strain of Rhizobium leguminosarum biovar phaseoli, three copies of the regulatory nodulation gene nodD were identified on the Sym plasmid and sequenced. Two were closely linked to each other and the third was near, but not adjacent, to the nodABC genes. Each of these nodD genes could correct the Nod defect of a nodD mutant strain of R. leguminosarum biovar viciae on peas. A truncated form of nodD2 could also correct this mutant, indicating that the C-terminus of NodD2 is not needed for inducing activity. Upstream of nodD1 and in the same operon is a newly described gene, nolE, whose product appears to be exported into the periplasm. Close to nodD2 is another gene, nolP, with no known counterpart in other rhizobia. Both nolP and nolE-nodD1 are preceded by ‘nod-box’ sequences and, in the former case, there appear to be two tandemly repeated nod-box sequences. Mutations in each of the nodD genes and in the nolE and nolP genes did not abolish nodulation or nitrogen fixation on beans.

    Notizen: The nucleotide sequence of the known portions of the mannitol operon in Escherichia coli (mtlOPAD) has been determined. Both the operator-promoter region and the intercistronic region between the mtlA and mtID genes (encoding the mannitol-specific Enzyme II of the phosphotransferase system and mannitol–1- phosphate dehydrogenase, respectively) show parallels with corresponding regions of the glucitol (gut) operon, but neither the mtlA nor the mtlD gene products show obvious homology with the corresponding gene products of the glucitol operon. Five potential cyclic AMP receptor protein binding sites were identified in the mtlOP region, all showing near identity with the consensus sequence. Four regions of dyad symmetry (four to seven bases in length), serving as potential repressor binding sites, overlap with the potential cyclic AMP receptor protein binding sites. Repetitive extragenic pallndromic (REP) sequences, forming stem-loop structures in the intercistronic region between mtlA and mtlD and following the mtlD gene were identified. Probable terminator sequences were not found in any of these three regulatory regions. Mannitol–1-phosphate dehydrogenase exhibits two overlapping, potential NAD+ binding sites near the W-terminus of the protein. Computer techniques were used to analyse the mtlD gene and its product.

    Notizen: Quadruplicate groups of juvenile red drum (initial mean weight 2.7 g; 20 fish per replicate) were fed experimental diets containing 35% crude protein and graded levels of lysine for eight weeks. Lysine concentration in the basal diet was 1.2% and was supplied by a combination of peanut meal and shrimp-head meal. The basal diet was supplemented with lysine-HCl to provide 1.6, 2.0, and 2.4% lysine. Each of these diets contained an essential amino acid (EAA) premix. Two additional diets were formulated to contain 1.2 and 2.4% lysine without the EAA premix. Weight gain and feed efficiency (FE) data indicated between 1.2 and 1.6% dietary lysine was adequate. However, serum lysine concentrations indicated 1.6-2.0% lysine was required. Fish fed 1.2 or 2.4% lysine, without the EAA premix, exhibited reduced weight gain and feed efficiency. Results indicated that red drum were able to utilize crystalline amino acids when incorporated into diets containing intact protein and, when lysine was adequate, the proteins were deficient in at least one other essential amino acid. It is recommended that a dietary lysine level of 5.7% of the dietary protein be used in formulating red drum diets.

    Notizen: Samples were extracted in Erlenmeyer flasks with hexane-ethanol-acetone (50:25:25). After extraction, 15 mL of water were added to separate the solution into polar and nonpolar layers. Lycopene was concentrated in the upper nonpolar phase while other components and polar pigments were contained in the lower polar phase. The lycopene phase was analyzed using isocratic HPLC with a C-18 column and methanol-THF-water (67:27:6) mobile phase. Lycopene and (β-caro-tene eluted as chromatographically pure peaks.

    Notizen: The effects of various chloride salts (NaCl, MgCl2, KCl or 65% NaCl + 35% KCl) in combination with mechanically separated beef (MSB at 0, 10 or 20%) on the quality of restructured steaks formulated from bullock chucks were determined. As level of MSB increased, objectionable connective tissue, flavor desirability and overall palatability decreased and off-flavors increased. The development of oxidative rancidity during frozen storage was not affected by MSB level and MSB had little effect on textural traits. NaCl and NaCl + KCl treated steaks received superior flavor and overall palatability ratings over steaks containing MgCl2 or KCl. Oxidative rancidity developed faster in steaks containing NaCl than the other salt types. Partial replacement (35%) of NaCl with KCl resulted in restructured steaks with sensory properties equal to and storage life superior to steaks with NaCl alone.

    Notizen: Soybeans were grown hydroponically and labeled with 75Se-selenite or 75Se-selenate. The seeds were then processed into a full fat soy flour, a defatted soy flour, and an acid-precipitated high-protein concentrate. Analysis for 75Se and total selenium showed that selenium concentration was dependent on the protein levels of the processed fractions indicating that the major portion of 75Se in soybeans is incorporated in proteins.

    Notizen: Vine-ripened and ethylene-treated tomatoes (cv. ‘Sunny’) were ripened at 15, 18, 21, and 25°C and evaluated for color using a colorimeter. Computer models were developed from the objective data to predict changes in color components (value, hue angle, chroma). Computer-predicted results were then converted to sensory equivalent scores to provide a basis for quality management of tomatoes within a handling and distribution system. The models obtained reasonably estimated changes in vine-ripened tomatoes (cv.‘Flora-Dade’) ripened at 15, 21, and 25°C, but were less successful at predicting changes in ethylene-treated fruit.
