    Notes: The mechanisms of water loss and their relation to muscle shortening were investigated by dry cooking in a controlled environment oven. Air was pumped vertically upward in the oven at a rate of 13.7 m3/hr. Cylindrical muscle samples were cooked at 121°C, 149°C and 177°C. The axes of these samples were in vertical, 45° and horizontal directions with respect to the oven. The muscle fibers were parallel to the axes of these samples. It is observed that total weight loss is almost independent of sample orientation, implying that the draining of fluid by gravity (a mechanism which is believed to be closely related to sample orientation) is not an important mechanism of mass transfer. The main mechanism is possibly fiber shrinkage which squeezes fluid out during cooking. Linear relationships between remaining water content and sample length were observed both before and after protein denaturation. The slopes of these two linear relations are different, suggesting that the way fluid is squeezed out is different before and after protein denaturation. Also, these slopes are almost independent of oven temperature in the 121-177°C range, suggesting that the remaining moisture content depends primarily on the shortening during cooking of a sample and only weakly on the oven temperature and initial condition (i.e., frozen or frozen-thawed) of the sample.

    Notes: : Beef/turkey blended patties, containing 107 to 108 cfu/g of Salmonella Senftenberg or Listeria innocua, were battered and breaded. The effect of frying (177 °C) and air convection cooking (288 °C) on thermal inactivation of S. Senftenberg and L. innocua was evaluated. A model was obtained to correlate product internal temperature with frying and oven cooking time. Cooking method significantly affected thermal history and subsequently the thermal inactivation of S. Senftenberg and L. innocua. The effect of frying time interacted with oven cooking time. Increasing frying time reduced the oven cooking time. Mathematical models were developed to correlate the survival rate of S. Senftenberg or L. innocua with frying and oven cooking time.

Herzberg are commonly credited with the first scientific description of fresh ground water floating on deeper saline water along coastal areas of the world. As C. W. Carlston has pointed out, J. DuCommun should receive the credit for the first quantitative statement of the “Ghyben-Herzberg principle,” DuCommun's work having antedated publications by both Badon Ghyben and Herzberg by more than 60 years. Moreover, two well-known scholars, Pliny the Elder and Charles Darwin also wrote on the topic long before Badon Ghyben and Herzberg.

    Notes: Abstract.— A time-resolved fluoroimmunoassay (TR-FIA) marketed for measuring cortisol in human sera was evaluated and validated for use in the quantification of plasma cortisol concentrations of channel catfish Ictalurus punctatus. Time-resolved fluoroim-munoassays provide non-isotopic alternatives to the use of radioimmunoassays (RIA). The evaluated TR-FIA satisfied strict criteria of precision (intra-assay coefficients of variation (CV) 〈 7%) and reproducibility (inter-assay CV ≤ 9%). Accuracy of the TR-FIA, calculated as the percent of exogenous cortisol recovered from spiked catfish plasma, averaged 99.5%. Assay sensitivity (minimum detection limit) in catfish plasma was 1.2 ng/mL, and the displacement curve for serially diluted channel catfish plasma paralleled the cortisol standard curve. Plasma cortisol concentrations of channel catfish in the presence and absence of a confinement stressor were used to characterize the im-munoreactive cortisol measurable by TR-FIA, and compared favorably to RIA values for the same samples (r2= 0.95, P 〈 0.001).

    Notes: Juvenile channel catfish Ictulurus punctatus were exposed to 1× (0.44 mg/L), 3× (1.32 mg/L), or 5× (2.19 mg/L) the recommended therapeutic concentrations of waterborne potassium permanganate (KMnO4) for 36 h to determine the toxicity of the chemical. The fish were observed for 14 d after exposure. Gill, liver, and blood samples were collected before exposure, at 12, 24, and 36 h of exposure, and at 48-h intervals for 14 d thereafter. Analysis of homogenized gill tissue showed a transient increase in manganese content that quickly disappeared once exposure was discontinued. Fish exposed to the 3× and 5× concentrations of KMnO4 experienced 9 and 50.6% mortality, respectively. Plasma cortisol was elevated more than ten-fold at the 5× concentration. Both plasma chloride and osmolality were significantly reduced at the 3× and 5× concentrations but were unchanged at the 1×. Packed cell volumes (PCV) of whole blood rose significantly in response to 3× and 5× concentrations of KMnO4 Mortality may have been the result of blood electrolyte depletion as indicated by increased PCVs, loss of chloride, and reduced osmolality. All stress indicators measured, except PCV at the 5× concentration, were indistinguishable from unexposed controls within 48 h after exposure was discontinued. At the l× concentration (the concentration most like that employed in a disease treatment) no changes were observed in any stress indicators measured suggesting that KMnO4 may be safely used as a disease therapeutant for channel catfish.

    Notes: Eighty subprimals were fabricated from 20 pairs of beef strip loins from lean (fat thickness, 0.2-0.4 cm) US Standard and US Good carcasses. Three vacuum packaged (VP) and one dry aged treatments were randomly assigned to short loin sections of each carcass. Steaks from VP intact and VP not-reassembled subprimals were rated highest in overall appearance and eating quality. Dry aging reduced retail caselife, produced greater shrinkage and trim losses and lower overall satisfaction ratings by panelists. Precutting subprimals into steaks followed by VP aging tends to decrease retail caselife (especially if steaks are reassembled) and decrease juiciness and tenderness.

    Notes: A gas chromatographic technique was used to determine sulfur dioxide and carbon dioxide in the headspace of packaged apples during storage. The headspace sulfur dioxide so determined decreased more rapidly and correlated better with nonenzymatic browning of the apples than free or total sulfur dioxide. The decrease in headspace sulfur dioxide, which was not accompanied by a comparable decrease in free sulfur dioxide, was attributed to the binding of sulfite to organic components of the apple. Free sulfur dioxide was found to measure not just the inorganic sulfite, which is commonly assumed, but also bound sulfite which was rapidly released under the acid conditions. Neither free nor total sulfur dioxide in a product could be quantitatively determined from headspace gas chromatographic techniques.

    Notes: Trypsin inhibitors were isolated from seeds of Amaranthus hypochondriacus by extraction at pH 7.5, heat treatment at 70°C for 15 min and affinity chromatography with trypsin-Sepharose 4B. Inhibitors of trypsin and chymotrypsin were identified in polyacrylamide gels following electrophoresis using a stain procedure that employed the chromogenic substrate acetyl-D, L-phenylalanine-2-napthyl ester. Three of the thirteen trypsin inhibitors identified in the electrophoresis gels, also had antichymotryptic activity. The inhibitor preparation was very thermostable retaining 20% of its original activity after 7 hr at 100°C.
